        Evaluation of mango saponin in broilers: effects on growth performance, carcass characteristics, meat quality and plasma biochemical indices

        Objective: The objective of the present study was to determine whether mango saponin (MS) could be used as a feed additive in broiler chicks by evaluating growth performance, carcass characteristics, meat quality, and plasma biochemical indices. Methods: A total of 216 1-d-old Arbor Acres male broiler chicks were randomly assigned into three dietary treatments supplemented with 0 (control), 0.14% (MS 0.14%), or 0.28% (MS 0.28%) MS. Each treatment had six replicates (cages) with 12 chicks each. The feeding trial lasted for six weeks. Results: Compared with the control, dietary supplemented with 0.14% or 0.28% MS increased average daily weight gain of chicks in the grower (22 to 42 d) and the whole (1 to 42 d) phases, and the final body weight of chicks on d 42 was higher in MS supplemented groups (p<0.05). Lower L45 min* (lightness) and L24 h* values, lower b24 h* (yellowness) value, and higher a45 min* (redness) and a24 h* values of the breast muscle were observed in chicks fed with 0.28% MS on d 42 (p<0.05). The total antioxidant capacity in plasma increased in MS 0.14% group on d 21 (p<0.001). Lower contents of plasma total cholesterol and triglyceride were observed in chicks fed with 0.28% MS on d 21 and d 42, whereas the group supplemented with 0.14% MS only decreased plasma triglyceride content on d 21 (p<0.05). The glucose content in plasma decreased in MS 0.28% group on d 42 (p<0.001). Conclusion: Overall, MS could be used as a feed additive in broiler chicks, and the supplemental level of 0.28% MS in diet could improve growth performance, meat quality, and plasma lipid metabolism in broiler chicks.

        <P>The various generations of Silicon Vertex Detectors (SVX, SVX', SVXII) for Collider Detector at Fermilab (CDF) at the Fermilab Tevatron have been fundamental tools for heavy-flavor tagging via secondary vertex detection. The CDF Run IIb Silicon Vertex Detector (SVXIIb) has been designed to be a radiation-tolerant replacement for the currently installed SVXII because SVXII was not expected to survive the Tevatron luminosity anticipated for Run IIb. One major change in the new design is the use of a single mechanical and electrical element throughout the array. This element, called a stave, carries six single-sided silicon sensors on each side and is built using carbon fiber skins with a high thermal conductivity on a foam core with a built-in cooling channel. A Kapton bus cable carries power, data and control signals underneath the silicon sensors on each side of the stave. Sensors are read out in pairs via a ceramic hybrid glued on one of the sensors and equipped with four SVX4 readout chips. This new design concept leads to a very compact mechanical and electrical unit, allowing streamlined production and ease of testing and installation. A description of the design and mechanical performance of the stave is given. Results on the electrical performance obtained using prototype staves are also presented.</P>

        Effects of Keratinase on Performance, Nutrient Utilization, Intestinal Morphology, Intestinal Ecology and Inflammatory Response of Weaned Piglets Fed Diets with Different Levels of Crude Protein

        Two experiments were conducted to investigate the in vitro ability of keratinase to hydrolyze soybean glycinin and ${\beta}$-conglycinin and to evaluate the in vivo effects of keratinase when included in corn-soybean diets with different levels of crude protein and fed to nursery pigs. In experiment 1, a saturated keratinase solution (1 ml) was added to two blank controls of either glycinin or ${\beta}$-conglycinin resulting in the hydrolysis of 94.74% glycinin and 88.89% ${\beta}$-conglycinin. In experiment 2, 190 pigs (8.3${\pm}$0.63 kg BW) were allotted to one of four treatments in a 2${\times}$2 factorial arrangement on the basis of body weight, and sex was balanced among the pens. The effects of crude protein (19 vs. 22%) and keratinase (0 vs. 0.05%) were studied. Each treatment was applied to six pens with seven (two pens) or eight pigs per pen. Pigs were fed the experimental diets for 21 d. Weight gain and feed conversion ratio were improved (p<0.05) with keratinase supplementation while feed intake was reduced (p<0.05). Keratinase supplementation increased (p<0.05) the apparent total tract digestibility of dry matter, energy, crude protein and phosphorus. Keratinase supplementation also increased n-butyric acid in the cecum and colon, lactobacilli and total anaerobe counts in the colon as well as the ratio of villus height to crypt depth in the ileum. Additionally, fecal score, ammonia nitrogen and branch chain volatile fatty acids in the colon, E. coli and total aerobe counts in the colon, crypt depth in the jejunum and ileum as well as serum interleukin-1 and interleukin-6 concentrations were also decreased (p<0.05) by keratinase supplementation. A reduction in dietary crude protein decreased (p<0.05) colon ammonia nitrogen concentration and cecal propionic acid and branch chain volatile fatty acid concentrations. In addition, cecal E. coli counts, colon total anaerobe counts, ileal crypt depth, and serum interleukin-1 and interleukin-6 concentrations were also decreased (p<0.05) with the reduction of dietary crude protein. With the exception of fecal scores, there were no significant interactions between crude protein and keratinase. This study provides evidence that dietary keratinase supplementation improved nursery pig performance by improving intestinal morphology and ecology, thus improving nutrient digestibility and alleviating the inflammatory response.

        Genetic polymorphism in pvmdr1 and pvcrt-o genes in relation to in vitro drug susceptibility of Plasmodium vivax isolates from malaria-endemic countries

        Treatment failure of chloroquine for Plasmodium vivax infection has increased in endemic countries. However, the molecular mechanisms for resistance and in vitro susceptibility of P. vivax to chloroquine remain elusive. We investigated the prevalence of mutations in the pvmdr1 and pvcrt-o genes, and the copy number of the pvmdr1 gene in isolates from the Republic of Korea (ROK), Thailand, the Union of Myanmar (Myanmar), and Papua New Guinea (PNG). We also measured in vitro susceptibility of Korean isolates to antimalarial drugs. The pvmdr1 analysis showed that mutations at amino acid position Y976F of pvmdr1 were found in isolates from Thailand (17.9%), Myanmar (13.3%), and PNG (100%), but none from the ROK, and mutation at position F1076L was present in isolates from the ROK (100%), Thailand (60.7%), and Myanmar (46.7%). One copy of the pvmdr1 gene was observed in most isolates and double copy numbers of the gene were observed in two Thai isolates. In the exons of the pvcrt-o gene that were sequenced, a K10 insertion was present in isolates from Thailand (56.0%) and Myanmar (46.2%), and the wild type was found in all Korean isolates. The results suggest that gene polymorphisms and copy number variation was observed in isolates of P. vivax from Southeast Asian countries. In Korean isolates polymorphism as limited to the F1076L variant, and no isolates with high level of resistance were found by in vitro susceptibility determinations. Moreover, our results provide a baseline for future prospective drug studies in malaria-endemic areas.

        Energy and Ileal Digestible Amino Acid Concentrations for Growing Pigs and Performance of Weanling Pigs Fed Fermented or Conventional Soybean Meal

        A new strategy of co-inoculating Bacillus subtilis MA139 with Streptococcus thermophilus and Saccharomyces cerevisiae was used to produce fermented soybean meal (FSBM). Three experiments were conducted to determine the concentration of digestible energy (DE) and metabolizable energy (ME) (Exp. 1), apparent ileal digestibility (AID) and standardized ileal digestibility (SID) of amino acids (AA) (Exp. 2), and feeding value (Exp. 3) of FSBM produced by this new strategy (NFSB) compared with soybean meal (SBM) and conventionally available FSBM (Suprotein). In Exp. 1, twenty-four barrows (initial body weight [BW] of $32.2{\pm}1.7kg$) were randomly allotted to 1 of 4 diets with 6 replicates per diet. A corn basal diet and 3 diets based on a mixture of corn and 1 of 3 soybean products listed above were formulated and the DE and ME contents were determined by the difference method. The results showed that there were no differences in DE and ME between SBM and either FSBM product (p>0.05). In Exp. 2, eight barrows (initial BW of $26.8{\pm}1.5kg$) were fitted with ileal T-cannulaes and used in a replicated $4{\times}4$ Latin square design. Three corn-starch-based diets were formulated using each of the 3 soybean products as the sole source of AA. A nitrogen-free diet was also formulated to measure endogenous losses of AA. The results showed that the SID of all AA except arginine and histidine was similar for NFSB and SBM (p>0.05), but Suprotein had greater (p<0.05) SID of most AA except lysine, aspartate, glycine and proline than NFSB. In Exp. 3, a total of 144 piglets (initial BW of $8.8{\pm}1.2$ kg) were blocked by weight and fed 1 of 4 diets including a control diet with 24% SBM as well as diets containing 6% and 12% NFSB or 12% Suprotein added at the expense of SBM. During d 15 to 28, replacing SBM with 6% NFSB significantly improved average daily gain (ADG) and average daily feed intake (ADFI) (p<0.05) for nursery piglets. During the overall experiment, ADG of piglets fed diets containing 6% NFSB was significantly greater (p<0.05) than that of piglets fed SBM. In conclusion, fermentation with the new strategy did not affect the energy content or the AID and the SID of AA in SBM. However, inclusion of 6% NFSB in diets fed to nursery piglets improved performance after weaning likely as a result of better nutritional status and reduced immunological challenge.

        A New Chemiluminescence Method for Determination of Cytosine Arabinoside in Pharmaceutical Preparations

        A novel chemiluminescence (CL) system was established for the determination of cytosine arabinoside (Ara-C) in pharmaceutical preparations. It was showed that a clear CL signal was observed when Eosin Y mixed with Fenton reagent. The CL intensity was decreased significantly when Ara-C was added to the reaction system and partially scavenged the hydroxyl radicals in the solution. The extent of decrease in the CL intensity had a good stoichiometrical relationship with the Ara-C concentration. Based on this, we developed a new method for the determination of Ara-C using a flow injection analysis (FIA) technique with CL detection. Under the optimal conditions, the linear range of Ara-C concentration was 6.0 × 10−9 ~ 1.0 × 10−7 mol/L (R = 0.9982) with a detection limit of 7.6 × 10−10 mol/L (S/N=3) , the RSD was 5.6% for 6.0 × 10−8 mol/L Ara-C (n = 11). The method was successfully applied to the determination of Ara-C in injection samples. The possible chemiluminescence reaction mechanism was discussed.

      • The Impact of Room Light and Speculae on Spectroscopic Measurements for Screening for Cervical Cancer: an Experiment Using a Simulated Gynecological Patient

        Fluorescence spectroscopy has been investigated as a means of detecting oral, lung and cervical neoplasia. However, there are many issues in developing a clinical protocol for routinely making such measurements. We conducted an experiment with a simulated gynecological patient to study the effect of room light and speculum type. The speculum is needed for the medical provider to access the cervix. We investigated whether: (1) room lights present or absent had an effect and (2) the type of speculum (no speculum, coated speculum, or metal speculum) made a difference. In recent years, simulation-based medicine has gained prominence in clinical science. A simulated gynecological model was employed as a surrogate for a human cervix, which provides a physical environment similar to the clinical setting and mitigates ethical considerations. Measurements on fluorescence standards placed inside the simulated patient were made at all combinations of lighting and speculum type. Both light and the type of speculum were significant factors, and there was a statistically significant interaction between the two factors. When the room light was absent, the measurements made with either the metal speculum and the coated speculum did not exhibit significant differences, so either type of speculum may be used when the lights are absent. When the lights are present, the coated speculum shows less room light contamination in the measurements. Because the room light can contaminate the fluorescence spectroscopic measurements, making the measurement with room lights absent is highly recommended.

        Determination of N* amplitudes from associated strangeness production in p+p collisions

        <P><B>Abstract</B></P> <P>We present the first determination of the energy-dependent amplitudes of N<SUP>⁎</SUP> resonances extracted from their decay in KΛ pairs in p+p → <SUP> pK + </SUP> Λ reactions. A combined Partial Wave Analysis of seven data samples with exclusively reconstructed p+p → <SUP> pK + </SUP> Λ events measured by the COSY-TOF, DISTO, FOPI and HADES Collaborations in fixed target experiments at kinetic energies between 2.14 to 3.5 GeV is used to determine the amplitude of the resonant and non-resonant contributions into the associated strangeness final state. The contribution of seven N<SUP>⁎</SUP> resonances with masses between 1650 MeV/c<SUP>2</SUP> and 1900 MeV/c<SUP>2</SUP> for an excess energy between 0 and 600 MeV has been considered. The Σ–p cusp and final state interactions for the p–Λ channel are also included as coherent contributions in the PWA. The N<SUP>⁎</SUP> contribution is found to be dominant with respect to the phase space emission of the pK Λ + final state at all energies demonstrating the important role played by both N<SUP>⁎</SUP> and interference effects in hadron–hadron collisions.</P>
