Secretion of Bovine β-Casein by Saccharomyces cerevisiae

Yeast expression plasmids containing an appropriate leader sequence and bovine β-casein cDNA were constructed to produce β-casein for the study of its functional characteristics. Two kinds of expression systems for β-casein were constructed using pCGY1444 as a precursor plasmid. This plasmid is a yeast-E. coli shuttle vector which contains the chelatin promoter. The plasmid pISB202 contains the invertase leader sequence and β-casein gene. The plasmid pDEB303 contains the original bovine β-casein leader sequence gene. These two plasmids were introduced into S. cerevisiae AB116 which is a strain deficient in the major yeast proteinases. Each clone was grown in minimal media for 24h before induction by CuSO_4. The cells were thus grown under expression conditions. Both strains harbouring pISB202 and pDEB303 expressed bovine β-casein. The β-casein was detected using immunochemical staining after western blot. Secretion of β-casein was detected in the culture broth. The estimated amount of secreted β-casein was approximately 50㎍/ℓ.
Spatial Distributions of 3H and 14C in the Shielding Concrete of KRR-2

연구로 2호기 해체과정에서 발생한 방사화된 수조 콘크리트 내의 깊이에 따른 및 의 방사능 분포를 고온연소로와 액체섬광계수기를 이용하여 분석하였다. 또한 향후 연구로 2호기 해체과정에서 발생된 콘크리트폐기물의 핵종재고량 평가에 활용하기 위하여 및 측정결과와 감마방출핵종과의 상관관계를 도출하였다. 및 의 검출하한값은 0.048 및 0.028 Bq/g이다. 연구로 2호기 수조 콘크리트 내의 및 의 깊이별 방사능 분포는 콘크리트 표면으로부터 멀어질수록 지수적으로 감소하는 경향을 보여주었다. 또한, 및 의 비방사능은 콘크리트에 존재하는 의 비방사능과 좋은 상관관계를 나타내었다. The depth distributions of total and activities were characterized for the activated shielding concrete from a decommissioning of KRR-2 using the commercially available tube furnace and a liquid scintillation counter. The correlation of measurement results between and gammer emitter was evaluated to apply for estimating radionuclide inventory of the concrete waste generated from decommissioning KRR-2. The detection limits for and are 0.048 and 0.028 Bq/g respectively. The specific activities of the and tend to decrease exponentially as the depth of the concrete becomes deeper from the surface. In addition, the and activities were in good correlation with the activities analysed for the shielding concrete of KRR-2.
Removal of Dissolved Oxygen from the Make-up Water of NPP Using Membrane-based Oxygen Removal System

Corrosion control, in the end-shield cooling system of Wolsung Nuclear Power Plant, is directly related to the control of dissolved oxygen (DO). The current method, being used to deoxygenate the end-shield cooling water, is a chemical treatment by addition of reducing agent, hydrazine, to react with DO. This method has several limitations including high reaction temperature of hydrazine , unwanted explosive hydrogen gas production, and its intrinsic harmful property. A new approach to remove DO using a membrane-based oxygen removal system (MORS) was tried to overcome limitations of the hydrazine treatment. The DO removal efficiency of the MORS was found to be in the range 87% to 98%: The higher vacuum, the lower water flow rate and the higher water temperature tend to increase the DO removal efficiency.

고장수목 정점사상에 대한 이용불능도의 불화실성을 분석하기 위한 방법 및 전산코드를 개발하였으며 그 유용성을 검증하였다. 이 방법은 몬테카를로 방법과 모멘트 방법을 고장수목 축소 기법과 함께 조합하여 개발하였고 WASH-1400에 있는 고장수목과 신뢰도 자료를 이용하여 본 연구에서 개발된 코드의 효율성을 검증하였다. 몬테카를로 방법과의 비교결과 이 방법을 이응하면 계산시간을 상당히 줄일 수 있으며 충분히 정확한 결과를 얻을 수 있음을 입증하였다. A method and computer code for the uncertainty analysis in the top event unavailability are developed and tested by combining Monte Carlo Method and Moments method with fault tree reduction technique. Using system fault trees and unavailability data selected in WASH-1400, the efficiency of the proposed method is tested and these results are compared with those obtained by Monte Carlo method. It is shown that the results are sufficiently good in accuracy and computation time is considerably reduced compared with those by Monte Carlo method.
Evaluating the Factors That Influence E-commerce Performance on SMEs in China

This paper investigated the factors that influence the e-commerce performance of Chinese small and medium enterprises (SMEs). E-commerce provides a platform the fair competition between SMEs and large enterprises. In fact, within a decade, e-commerce has brought economic benefits to many SMEs. Hence a number of SMEs rapidly adopted this platform in recent years. SMEs are facing critical barriers to e-commerce application. First, there are e-commerce implementation and adoption decision issues. Second, there is a need to consider the key success factors and its relationship with other variables. Third, these is a dilemma on how to measure the effect on the firm of implementing e-commerce and the appropriate performance evaluation procedure. In this paper, theoretical and empirical explorations and critical research were conducted on e-commerce among SMEs in China. Considering the downward trend in the performance of SMEs and the existing theoretical models, this study presented relevant hypothesis. Past studies were also reviewed and organized to support the argument. This paper analyzed the factors that influenced e-commerce performance based on a number of research papers. Using the statistical SEM verification method, this study also discovered the critical success factors in doing business through e-commerce among SMEs. Two critical factors were verified from about 200 cases which are subjected to statistical processing. It was found that internal human resources and e-commerce strategies are the statistically significant success factors for e-commerce performance among Chinese SMEs.

Peroxisome proliferator-activated receptors (PPARs), members of the nuclear hormone receptor family, are key regulators of various metabolic pathways related to lipid and glucose metabolism as well as inflammation. We examined the effect of zingerone, a major ingredient of ginger, on PPAR, hepatic nuclear factor-4 (HNF-4), and nuclear factor-${\kappa}B$ (NF-${\kappa}B$) expression in 21-month-old male Sprague-Dawley rats. Two experimental groups receiving doses of either 2 or 8 mg/kg/day zingerone for 10 days were compared with young rats (6 months old) and an age-matched control group. For molecular work, the endothelial cell line YPEN-1 was used. Both the 2 and 8 mg/kg/day dose of zingerone significantly increased DNA binding activities of PPARs (2.8-fold). Expression of HNF-4 was also increased in the group receiving the 8 mg/kg/day dose. We further showed that zingerone partially prevented the age-related decline in PPAR expression. In vitro experiments revealed zingerone ($10\;{\mu}M$) increased PPAR expression (2.5-fold) to a similar extent as the PPAR agonist fibrate ($5\;{\mu}M$) and suppressed pro-inflammatory transcription factor NF-${\kappa}B$ activity. Collectively, our findings suggest that zingerone exerts its potent anti-inflammatory action by increasing HNF-4 and PPAR activities, while suppressing NF-${\kappa}B$ activity.

Feline calicivirus (FCV) is a highly infectious pathogen in cats and widely distributed worldwide with high genetic variation. Full-length open reading frame 2 of 5 from recently isolated Korean FCV isolates were sequenced and compared with those of global isolates. The results of phylogenetic analysis supported dividing global FCV isolates into two genogroups (type I and II) and demonstrated the presence of genogroup II in Korea, indicating their geographic spread in East Asia. High sequence variations in region E of the FCV isolates emphasizes that a novel vaccine needs to be developed to induce protective immunity against various FCV strains.

Acute total obstruction of the left main coronary artery (LMCA) is a serious emergency condition requiring prompt diagnosis and treatment. Unless properly treated, it will likely progress to cardiogenic shock and a high mortality rate. We report a case of acute LMCA total obstruction presenting with atypical momentary electrocardiogram (ECG) changes including right bundle branch block with left axis deviation, and ST-segment elevation in aVR and aVL. We focus on the unusual ECG changes associated with LMCA obstruction which should be noted in order to ensure revascularization without delay, especially when this condition is accompanied by cardiogenic shock.

On-line determination of trace uranium in sea water by flow injection-inductively coupled plasma-mass spectrometry(FI-ICP-MS) has been investigated. The method of flow injection equipped wth microcolumn was used for on-line separation and enrichment of uranium prior to determination by ICP-MS because the influences of concomitant matrix in sea water are serious problem in introducing to ICP-MS. In order to take high accuracy and precision, flow injection parameters such as time response, acid concentration, sampling volume, flow rate, etc., and other instrumental conditions were optimized. A sampling volume of 3mL resulted in a preconcentration factor of about 30, and precision at the 3 ng/mL level was 3.5%(RSD). Developed method was applied to natural sea water samples off the coast of Young Kwang nuclear power plant in Korea and the results were assessed through either analysis of synthetic sea water reference sample or recovery experiment
