        Impact of NR1I2, adenosine triphosphate-binding cassette transporters genetic polymorphisms on the pharmacokinetics of ginsenoside compound K in healthy Chinese volunteers

        Background: Ginsenoside compound K (CK) is a promising drug candidate for rheumatoid arthritis. This study examined the impact of polymorphisms in NR1I2, adenosine triphosphate-binding cassette (ABC) transporter genes on the pharmacokinetics of CK in healthy Chinese individuals. Methods: Forty-two targeted variants in seven genes were genotyped in 54 participants using Sequenom MassARRAY system to investigate their association with major pharmacokinetic parameters of CK and its metabolite 20(S)-protopanaxadiol (PPD). Subsequently, molecular docking was simulated using the AutoDock Vina program. Results: ABCC4 rs1751034 TT and rs1189437 TT were associated with increased exposure of CK and decreased exposure of 20(S)-PPD, whereas CFTR rs4148688 heterozygous carriers had the lowest maximum concentration ($C_{max}$) of CK. The area under the curve from zero to the time of the last quantifiable concentration ($AUC_{last}$) of CK was decreased in NR1I2 rs1464602 and rs2472682 homozygous carriers, while $C_{max}$ was significantly reduced only in rs2472682. ABCC4 rs1151471 and CFTR rs2283054 influenced the pharmacokinetics of 20(S)-PPD. In addition, several variations in ABCC2, ABCC4, CFTR, and NR1I2 had minor effects on the pharmacokinetics of CK. Quality of the best homology model of multidrug resistance protein 4 (MRP4) was assessed, and the ligand interaction plot showed the mode of interaction of CK with different MRP4 residues. Conlusion: ABCC4 rs1751034 and rs1189437 affected the pharmacokinetics of both CK and 20(S)-PPD. NR1I2 rs1464602 and rs2472682 were only associated with the pharmacokinetics of CK. Thus, these hereditary variances could partly explain the interindividual differences in the pharmacokinetics of CK.

        Risk factors associated with impaired ovarian reserve in young women of reproductive age with Crohn’s disease

        Background/Aims: Crohn’s disease (CD) primarily affects young female adults of reproductive age. Few studies have been conducted on this population’s ovarian reserve status. The aim of study was to investigate potential risk factors associated with low ovarian reserve, as reflected by serum anti-Müllerian hormone (AMH) in women of reproductive age with CD. Methods: This was a case-control study. Cases included 87 patients with established CD, and healthy controls were matched by age, height and weight in a 1:1 ratio. Serum AMH levels were measured by enzyme-linked immunosorbent assay. Results: The average serum AMH level was significantly lower in CD patients than in control group (2.47±2.08 ng/mL vs. 3.87±1.96 ng/mL, respectively, P<0.001). Serum AMH levels were comparable between CD patients and control group under 25 years of age (4.41±1.52 ng/mL vs. 3.49±2.10 ng/mL, P=0.06), however, serum AMH levels were significantly lower in CD patients over 25 years of age compared to control group (P<0.05). Multivariable analysis showed that an age greater than 25 (odds ratio [OR], 10.03; 95% confidence interval [CI], 1.90-52.93, P=0.007), active disease state (OR, 27.99; 95% CI, 6.13-127.95, P<0.001) and thalidomide use (OR, 15.66; 95% CI, 2.22-110.65, P=0.006) were independent risk factors associated with low ovarian reserve (serum AMH levels <2 ng/mL) in CD patients. Conclusions: Ovarian reserve is impaired in young women of reproductive age with CD. Age over 25 and an active disease state were both independently associated with low ovarian reserve. Thalidomide use could result in impaired ovarian reserve. (Intest Res 2020; 18:200-209)

        A Data-driven Approach to Estimate the Probability of Pedestrian Flow Congestion at Transportation Bottlenecks

        In public gathering places, pedestrian flow congestion may happen in transportation bottlenecks. Limitations exist in the conventional judgment of congestion by considering the crowd density or the walking speed merely. In this paper, a data-driven mathematical approach based on Kernel Density Estimation (KDE) to analyze the probability of pedestrian flow congestion is proposed, which comprehensively considers the walking speed, the crowd density and the flow rate when pedestrians walk towards possible bottlenecks. With the case study of Nanjing metro station during rush hours, the advantages of non-parametric KDE compared with traditional parametric normal distribution estimation are analyzed and the optimal bandwidth for KDE is also discussed. The case study shows that the proposed method can obtain a more reliable quantitative assessment of congestion risk, as it overcomes the limitation of parametric estimation that relies on experience, and also avoids biased assessment of congestion that merely concerns single parameter of pedestrian flow. Finally, an assessment framework for dynamic congestion risk at bottlenecks is suggested. With this framework, the change of congestion situations of a monitored region can be mastered through data-driven approach, and thus the transformation of normal flowing to congestion of the crowd can be quantified through probabilistic analysis.

        Modulation electronic structure of NiS nanoarray induced by Fe, V doping for high efficiency water and urea electrolysis

        Exploring high-efficient and stable low-cost electrocatalysts is of significant importance for boosting theefficiency of water splitting and purifying urea-enriched wastewater. Herein, bimetallic doping strategywas adopted to obtain jasminum nudiflorum-like Fe, V doped NiS arrays (Fe, V-NiS/NF) via typicalhydrothermal process and subsequent anion exchange reaction. The as-obtained Fe, V-NiS/NF array displayshigh catalytic activity and stability toward oxygen evolution reaction (OER) and urea oxidationreaction (UOR) in alkaline media, with reduced overpotentials of 273 and 214 mV to deliver the currentdensity of 50 mA cm2 for OER and UOR, respectively. More notably, when employing Fe, V-NiS/NF assymmetric electrolytic cell for urea electrolysis, a low cell voltage of 1.45 V is needed at 10 mA cm2,which is about 110 mV lower than the conventional water electrolysis. Meanwhile, the catalyst also displayssuperior stability for over 72 h. Such outstanding performance is attributed to the following points:(i) 3D porous flower-like structure facilitates the mass transfer and abundant exposure of active sites; (ii)in situ growth of catalysts on conductive substrate and the effective interface engineering of differentcomposition shorten the charge transport pathways and expedite electron transfer. Density functionaltheory calculations demonstrate that the Fe and V dopants regulate the electronic environment of Ni sitesand optimize the adsorption free energy of urea. This work provides a universal pathway to design highefficientand non-noble electrocatalysts for H2 production in an energy-saving way via urea electrolysis.

        Coexistence of Subcutaneous Panniculitis-Like T-Cell Lymphoma and Dermatomyositis in a 12-Year-Old Boy

        Subcutaneous panniculitis-like T-cell lymphoma (SPTL) is an extremely rare, indolent skin malignancy that can be difficult to distinguish from autoimmune disease-associated panniculitides. Here, we describe a 12-year-old boy who was diagnosed at age 7 years with dermatomyositis with classical manifestations, including poikiloderma, Gottron’s sign, and symmetric muscle weakness. Recently, the boy presented multiple subcutaneous nodules and fever. Histopathological examination and immunohistochemical staining revealed co- existence of SPTL. To our knowledge, this is the first case of dermatomyositis accompanied with SPTL. This case alert clinical physicians of the possibility of SPTL should be considered when a patient with dermatomyositis has new lesions presenting as nodules and unknown fever.

        Dickson Charge Pump with Gate Drive Enhancement and Area Saving

        This paper presents a novel charge pump scheme that combines the advantages of Fibonacci and Dickson charge pumps to obtain 30 V voltage for display driver integrated circuit application. This design only requires four external capacitors, which is suitable for a small-package application, such as smart card displays. High-amplitude (<6.6 V) clocks are produced to enhance the gate drive of a Dickson charge pump and improve the system's current drivability by using a voltage-doubler charge pump with a pulse skip regulator. This regulation engages many middle-voltage devices, and approximately 30% of chip size is saved. Further optimization of flying capacitors tends to decrease the total chip size by 2.1%. A precise and simple model for a one-stage Fibonacci charge pump with current load is also proposed for further efficiency optimization. In a practical design, its voltage error is within 0.12% for 1 mA of current load, and it maintains a 2.83% error even for 10 mA of current load. This charge pump is fabricated through a 0.11 μm 1.5 V/6 V/32 V process, and two regulators, namely, a pulse skip one and a linear one, are operated to maintain the output of the charge pump at 30 V. The performances of the two regulators in terms of ripple, efficiency, line regulation, and load regulation are investigated.

        Proteolysis Characteristics of Sarcoplasmic, Myofibrillar, and Stromal Proteins Separated from Grass Carp and Antioxidant Properties of Their Hydrolysates

        Proteolysis of grass carp sarcoplasmic, myofibrillar,and stromal proteins by 5 commercial proteases were studied. Sarcoplasmic and myofibrillar protein could be well hydrolyzed by Alcalase 2.4 L to reach high protein recoveries (PR) (71.86±2.46 and 80.77±3.05%, respectively),while the maximum PR for stromal protein was only 42.83±2.84%. However, stromal hydrolysates, containing mostly 6-10 kDa fraction, exhibited higher ·OH scavenging activities due to its high content of antioxidant-assisting amino acids. Alcalase 2.4 L and pancreatin 6.0, which produced hydrolysates with relative high degree of hydrolysis (DH), were used for further hydrolysis of whole grass carp protein with the assistance of response surface methodology (RSM). The results showed that serine proteases (Alcalase 2.4 L and pancreatin 6.0) could produce sarcoplasmic, myofibrillar, or stromal hydrolysates with relatively high PR, DH, and strong ·OH scavenging activity, which may be used to prepare antioxidant hydrolysates from grass carp.

        The potential neuritogenic activity of aqueous extracts from Morchella importuna in rat pheochromocytoma cells

        The aim of this study was to explore the neuritogenic effects of aqueous extracts from the fruiting bodies of Morchella importuna (MEA). 3-(4, 5-dimethythiazol-2- yl)-2, 5-diphenyltetrazolium bromide (MTT) assay was carried out to assess the cytotoxicity of MEA. Neurite outgrowth stimulation assay was used to evaluate the potentiation of neuritogenic activity induced by MEA. The specific inhibitors for TrkA, MEK/ERK and PI3K signaling pathway were served to clarify the mechanism of MEA’s neuritogenic effects. It was shown that MEA could mimic neuritogenic activity of NGF, a kind of representative neurotrophic factors with no significant cytotoxicity, and stimulate neurite outgrowth in a dose-dependent manner of PC12 cells. The neuritogenic activity induced by MEA required activity of PI3K/Akt and MEK/ERK1/2 signaling pathways, as well as parts of TrkA receptor. Accordingly, MEA could be used as a promising neuritogenic-stimulation compound for nervous diseases treatment.
