STD61종 열간금형강의 수명향상을 위하여 ion nitriding처리를 실시하였다. 이온질화전의 모재의 열처리시에는 1030˚C에서 quenching한 후 530˚C에서 2시간 tempering했을 때 Hv=600의 높은 경도값을 나타내었다. 580˚C, 600˚C에서 2시간 tempering했을 때는 각각 Hv=500, Hv=400으로 경도값이 오히려 감소했다. 이온질화 처리시에는 처리시간이 길고 N₂와 Ar첨가유량이 많을 수록 깊은 화합물층과 확산층을 얻었다. 그리고 각 처리조건에서 안정된 Hv=1150의 질화물층을 얻을 수 있다. Ion nitriding treatment has been practiced to improve the life of hot die steel of STD61. Before nitriding the parent STD61 tempered at 530˚C, 2hrs after quenching at 1030˚C, It showed the high hardness distribution of Hv=600. The hardness distribution has decreased by Hv=400, Hv=500 when tempered for 2hrs at 580˚C, 600˚C. In the ion nitriding, the more treat-time and the more N₂and Ar flux, the deeper the compounded layer and the diffused layer. The stable hardness, Hv=1150, of nitride layer was obtained at various treatment condition.

A great interest has been rising on securing safety and the maintenance of structures in the civil engineering and construction fields due to the increase in the number of large structures and social infrastructures as well as collapses of bridges and high rise buildings. MEMS (Micro-Electro-Mechanical System), one of the technologies that are rapidly applied to the evaluation of the integrity of structures, has advantages of being adequate for wireless monitoring owing to the low cost and low electricity consumption, and ease of installation. In this paper, the evaluation of the wireless MEMS system has been conducted using the shaking table and a full scale shear building. The effectiveness of wireless MEMS system has been verified by comparing the results of the evaluations of MEMS accelerometer and traditional accelerometer. To assess the applicability, the wireless MEMS monitoring system is applied to a full-scale building structure with hybrid mass damper(HMD) system. The building is a 5-story steel building and the HMD is mounted on the 4th floor to vibrate the building. The field evaluation results indicate that the wireless MEMS monitoring system is reliable and robust for the health monitoring of buildings.

Objective The objective of the present study was to explore causal pathways to understand how second traumatic experiences could affect the development of emotional exhaustion and psychiatric problems. Methods A total of 582 workers who had jobs vulnerable to secondary traumatic experiences were enrolled for this study. Emotional exhaustion, secondary trauma, resilience, perceived stress, depression, anxiety, and sleep problems were evaluated. A model with pathways from secondary traumatic experience score to depression and anxiety was proposed. The participants were divided into three groups according to the resilience: the low, middle and high resilience group. Results Resilience was a meaningful moderator between secondary traumatic experiences and psychiatric problems. In the path model, the secondary trauma and perceived stress directly and indirectly predicted perceived stress, emotional exhaustion, depression, anxiety, and sleep problems in all three groups. Direct effects of perceived stress on depression and anxiety were the largest in the low resilience group. However, direct effects of secondary trauma on perceived stress and emotional exhaustion were the largest in the high resilience group. Conclusion Understanding the needs of focusing for distinct psychological factors offers a valuable direction for the development of intervention programs to prevent emotional exhaustion among workers with secondary traumatic experiences.

<P>Interleukin-12 (IL-12) gene was shown to produce both IL-12 and p40 subunit. The excess production of the p40 subunit as a natural antagonist of IL-12 is a major obstacle of IL-12 gene-based cancer therapy. We previously reported that IL-12N220L gene, which selectively reduces the secretion of the p40 subunit, induces long-lasting stronger type 1 helper T cells (T(H)1) and cytotoxic T lymphocyte (CTL) immunity in hepatitis C virus (HCV) E2 DNA vaccination model and higher protection from challenge with tumor cells expressing E2 than IL-12 in a prophylactic setting. Here, we demonstrated that intratumoral injection of IL-12N220L-expressing adenovirus showed better tumor growth inhibition and higher survival rate than that of IL-12 or granulocyte macrophage-colony stimulating factor (GM-CSF)-expressing adenovirus in a therapeutic setting. In particular, the mice cured by IL-12N220L treatment were protected against intravenous rechallenge of the same tumor cells better than those by IL-12 treatment. In addition, the enhanced antitumor activity of IL-12N220L was confirmed in B16F10 lung metastasis model, which correlated with the frequency of tumor-specific interferon (IFN)-gamma-secreting cells. When tested in CT26/NP tumor that expresses influenza nucleoprotein (NP) as a tumor antigen, IL-12N220L induced stronger NP-specific T(H)1 and CTL responses than IL-12, particularly at a later time point, indicating the generating long-term tumor-specific memory T-cell responses. Moreover, the potent antitumor effects of IL-12N220L were further augmented by combination with chemotherapy using farnesyltransferase inhibitor (FTI), LB42908. Taken together, our results suggest that IL-12N220L is superior to IL-12 in cancer immunotherapy, which can be further enhanced by combination with chemotherapy.</P>
Cloning of the Transketolase Gene from Erythritol-Producing Yeast Candida magnoliae

The entire nucleotide sequence of the TKL1 gene encoding transketolase (TKL) in an erythritolproducing yeast of Candida magnoliae was determined by degenerate polymerase chain reaction and genome walking. Sequence analysis revealed an open reading frame of C. magnoliae TKL1 (CmTKL1) that spans 2,088 bp and encodes 696 amino acids, sharing 61.7% amino acid identity to Kluyveromyces lactis TKL. Functional analysis showed that CmTKL1 complemented a Saccharomyces cerevisiae tkl1 tkl2 double mutant for growth in the absence of aromatic amino acids and restored transketolase activity in this mutant. An enzyme activity assay and RT-PCR revealed that the expression of CmTKL1 is induced by fructose, H2O2, and KCl. The GenBank accession number for C. magnoliae TKL1 is KF751756.
Neuregulin induces CTGF expression in hypertrophic scarring fibroblasts.

<P>Hypertrophic scarring (HTS) is a common fibroproliferative disorder that typically follows thermal and other injuries involving the deep dermis. These pathogenic mechanisms are regulated by connective tissue growth factor (CTGF) and transforming growth factor-관. We found that neuregulin-1 (NRG1), as well as NRG receptors, HER-2, and HER-3 were upregulated in HTS fibroblasts (HTSF), compared with normal fibroblasts. Furthermore, NRG1 stimulation increased the expression of CTGF in HTSF. In the presence of inhibitors of PI3K, Src, Smad, or reactive oxygen species, the effect of NRG1 on CTGF expression decreased significantly. In particular, the combination of LY294002 or PP2 with SB431542 blocked NRG1-mediated CTGF expression in HTSF. Finally, we demonstrated that siRNA for CTGF, AG825, LY294002, and PP2, either alone or in co-treatment, effectively reduced extracellular matrix expression. Taken together, our results suggest that NRG1 is involved in fibrotic scar pathogenesis via PI3K- or Src-mediated CTGF expression.</P>
