        자폐적 행동 양상을 보이는 아동과 어머니간의 애착 발달에 관한 분석적 연구

        This study was conducted to examine the feature of attachment and the main factors of attachment development between autistic child and his or her mother. Data was collected at Day Care Center for Children with Developmental Disorder in S National University Hospital through nurses's in S National University Hospital through nurses's participating observation, interview with the mothers, and review of nursing records, from April 2 to June 30, 1994. The subjects were all 6 children, one was a girl, the others were boys, and their mothers who participate at the sensory-integrative play for attachment promotion served at this center. Content analysis was done for data analysis. First, all data were categorized by main concepts, reciprocity and affect. Then these wee analyzd and discussed on the aspects of specificity, proximity, synchrony, joint attention, strong affect, emotional availability which were chosen as the components of attachment through literature review. The results of this study were summarized as follows : all subjects were underdeveloped, unstable and showed different level of mother-child attachment. But during the program their attachment behaviors improved little by little. The behaviors which represented specificity, proximity, and strong affect were increased considerably, on the other hand the behaviors which represented joint attention and emotional availability were scarce. The more mother and child had contact and interacted merrily, the more their attachment behaviors developed. Another important factors of attachment development in these cases were mother and child's each characteristics. Especially the child's autistic peculiarity, such as indifference, severe resistance, distracterbility, stereotype tendency, physical condition were major barriers of maintatining the mother-child interaction. Also mother's attitude to her child, raring skill and physical condition were influencing factors. A few mothers who were anxious and stubborn had much more difficulty to adjust to their children. The mother who had experienced happiness with her child, especially if the mother experienced more fun at the play by herself became more positive, active and flexible, and that mother-child interaction maintained much longer and improved. It was more critical to help them enjoy their interaction. Thus nurses had to plan and intervene more joyful and various play, and consider mother's need in addition to child's need. And The program should be strengthened to support the mother emotionally to maximize the therapeutic effect.

        G+C 함량이 높은 Primer를 사용하는 중합효소 연쇄반응에서 변성제가 미치는 영향

        G+C 함량이 높은 primer를 이용한 중합효소 연쇄반응을 실시하는 경우 높은 annealing 온도로 인하여 특이 염기서열의 합성정도가 매우 미약하게 나타나는 경우가 많이 있다. 이와 같은 문제점을 보완하기 위하여 glycerol, formamide 및 dimethyl sulfoxide (DMSO) 등의 변성제를 반응용 완충액에 첨가하고 중합효소 연쇄반응을 실시하여 그 결과를 비교 검토하였다. G+C 함량이 낮은 Borrelia burgdorferi의 Ly1 유전자의 primer set인 Bb 679와 Bb 680를 이용한 중합효소 연쇄반응에서는 변성제 첨가에 따른 합성 DNA의 양의 변화가 뚜렷하지 않았다. 그러나 G+C 함량이 높은 primer set인 Mycobacterium paratuberculosis의 IS900 유전자의 IS900/150C와 IS900/921를 이용한 중합효소 연쇄반응을 유도한 경우에는 변성제를 첨가함에 따라 합성된 DNA의 양의 증가가 뚜렷하였으며, 2.5% glycerol과 1.25% formamide를 혼합 첨가한 경우와 또는 2.5% DMSO를 반응용 완충액에 첨가하였을 때 비특이적인 증폭비율이 낮아 특이 염기서열의 합성 결과가 양호한 것으로 나타났다. Poor yields of amplified DNAs could be resulted in polymerase chain reaction(PCR) processes with G+C-rich DNA primers because of their high Tm values. To maximize the yields of amplification in PCR processes with G+C-rich primers, we compared the yields of amplified DNA fragments according to the concentrations of specific denaturants added to the reaction mixture of PCR system. With addition of the mixture of 2.5% glycerol and 1.25% formamide, or 2.5% dimethyl sulfoxide to the reaction cocktail, respectively, remarkable increases in the yields of amplified DNA fragments were not observed in the PCR systems with G+C-low primers of Lyl chromosomal gene from Borrelia burgdorferi but observed in the PCR system with G+C­rich primers of IS900 gene from Mycobacterium paratuberculosis. Although we were not practically able to discriminate the yields of PCR DNAs according to the concentrations used in this study, addition of the mixture of 5% glycerol and 2.5% formamide, or 5% DMSO tended to increase the production of extra bands.

        사업장 소음 폭로에 의한 일과성 역치 상승과 회복

        To determine the recovery time from noise-induced temporary threshold shift(TTS), a prospective field study was conducted at three worksites where workers are known to be exposed high level of noise. Subjects were selected according to answers on a questionnaire which inquired about otological history and previous noise exposure, including avocational, military and occupational exposures. After excluding employee with past otologic problems, recent exposure to high level noise, and under medications, total 92 employees participated in the study. Among 92 participants, complete consecutive audiometric examinations were carried out at 0∼2 hours, 5∼7 hours, 14∼16 hours after worktime noise exposure on 26 participants wearing hearing protectors and 22 participants wearing no protective devices. The difference between the hearing level 0∼2 hours after noise exposure and 5∼7 hours is statistically significant by paired t-test(p<0.01). The median recovery times calculated from the data of 22 participants wearing no protective hearing devices are 15.6 hours at 4000Hz, and 7.7 hours, 10.3 hours, 8.4 hours at 1000Hz, 2000Hz and 8000Hz respectively. These data suggest that when measuring the pure tone audiometry for noise exposed workers, at least 16 hours noise-free interval is required.

        국내에서 채집한 진드기에서 중합효소연쇄반응을 이용한 라임병균 및 Ehrlichiosis 원인체의 검출

        국내에서 채집한 진드기의 라임병 및 ehrlichiosis원인체 보균 상태를 조사하기 위하여 총 516마리 (Ixodes spp. 22마리, Haemaphysalis spp. 494마리)의 ixodid 진드기를 봄과 가을에 걸쳐 강원도 고산지대 일원에서 채집하였다. 수집한 진드기에서 DNA를 추출 ·정제한 후 추출한 DNA를 template로 이용하여 , Borrelia burgdorferi sensu lato 및 Ehrlichia spp.에 특이하게 반응하도록 제작한 primer를 이용한 중합효소연쇄반응 (polymerase chain reaction, PCR)을 실시하였으며, 이 결과를 oligonucleotide probe를 사용한 southern blotting을 통하여 다시 확인하였다. 총 516마리의 진드기중 B.garinii sensu lato DNA 양성 인 진드기는 68(13.2%)마리 (B. burgdorferi sensu stricto,2; B. afzelii,1; B. garinii,33; B.tanukii,8; B.turdae, 4)로 나타났으며 이 중 37 (7.2%)마리의 진드기 는 southern blot analysis에서도 양성으로 확인되었다. 또한 101 (19.2%)마리의 진드기가 Ehrlichia spp.에 대한 PCR에서 양성 이었으나, 이들 중 25 (4.8%)마리 만이 southern blot analysis에서 양성으로 확인되었다. 그러나 사람의 병인체로 추정 되는 human granulocytic ehrlichiosis (HGE) agent DNA를 보균한 진드기는 확인되지 않았다. 한편 라임병균과 ehrlichiosis원인체를 동시에 보균한 것으로 밝혀진 진드기가 3마리에서 (0.6%)확인되어,국내에서도 진드기 교상시 이들 두 가지 열성질환이 동시에 감염될 가능성이 있는 것으로 사료됨으로 진드기 매개성 열성질환에 대한 적절한 진단법 등을 보다 체계적으로 연구하여야 할 것으로 판단된다. To investigate the distribution of Borrelia Burgdorferi and human granulocytic ehrlichiosis(HGE) agent in ticks, adult ixodid ticks of Ixodes spp. and Haemaphysalis spp. were collected from the high mountain areas of Kangwon Province. Using DNAs extracted and purified in the collected ticks, polymerase chain reaction (PCR) was performed to amplify the specific nucleotide sequences of both agents. Of the 516 ticks, a total of 68 (13.2%) ticks was positive for Borrelia burgdorferi sensu lato with PCR analysis (2 for B. burgdorferi sensu stricto; 1 for B. afzelii; 33 for B. garinii; 8 for B.tanukii; 4 for B. turdae). However a little more than half of PCR-positive ticks (37/68) was found to be positive in the southern blot analysis with Bl6S oligonucleotide probe. One hundred and one (19.2%) ticks were positive for Ehrlichia spp. in PCR, and a quarter of them (25/101) was positive in southern blot with E16S oligonucleotide probe. But none of them was found to be the DNA of HGE agent. And 0.6% (3/516) ticks were positive for both of B. burgdorferi sensu lato and Ehrlichia spp. These findings might implicate the possibility of the outbreak of lyme borreliosis and ehrlichiosis in Korea, and more extensive studies may be need for the diagnosis of multiple tick-borne diseases.

      • 어린이 김치 표준화에 대한 연구(1) : 어린이 김치의 담금법 개발 Development of the Preparation of Kimchi for the Children

        The purpose of this study is reporting the results of first trial for the standardization of children's kimchi in terms of brining standardization, development for the recipe ad pannel test for the children's kimchi. In order to develop the standard recipe for children's kimchi, our research team have visited kimchi factories, temples and houses located in Pusan and Kyung-Sang Nam Do province. Three kinds of chinese cabbage kimchi (hamyang, haeundae, and jinrae kimchi named by region where we collected) which got the top three highest score in panel testing was selected. The original recipes of these kimchi were sightly modified to prepared the kimchi at laboratory. Among three of them, hamyang kimchi which contained dried anchovy power was selected as a model kimchi for development of the children's kimchi recipe. We find out that the optimal salt concentration of brined cabbage and kimchi for the children's kimchi (sliced style kimchi, approximately 2.5X3cm a piece) that showed the hightest pennel score were approximately 1.5% and 2.0%, respectively. The salt concentration of brined cabbage became 1.5, 1.43, and 1.60% after 60 min at 10% brining solution, 45 min at 12% and 30 min at 15%, respectively. The developed recipe used for the children's kimchi at our team is as follows. The major ingredients used for the preparation of kimchi is brined sliced chinese cabbage (salt concentration is 1.5%) 1kg galic 11g, ginger 5.0g, red pepper powder 20.0g, fermented anchovy juice 25.0g, fermented shrimp juice 5.0g dried anchovy and sea weed extracts 30.0g, anchovy power 2.4g glutinious rice gel 40.0g, starch syrup, 10.0g, and apple juice 22.5g. The total acceptability test of developed children's kimchi (test kimchi) was carried out with 84 of 6th grade student during lunch time at elementary school in Pusan where has a school lunch program. Sixty nine percent of student answered test kimchi is very good, 25% of them said it is fair, and only 6% of students answered they didn't like test kimchil. However, for the kimchi supplied from school at lunch, 52% students answered the kimchi is good, 26% of them said it is fair, and 22% of them said they don't like school supplied kimchi. This result tells us that the test kimchi is better than school supplied kimchi and the test kimchi can be developed further as a children's kimchi.

        Sequential Induction of Ethylene , Lipoxygenase , and Ascorbate Peroxidase in Senescing Soybean Callus

        Bursts of ethylene production occurred in twice at an early exponential (EEP) and prestationary (PSP) phases, respectively, during growth of callus tissue isolated from the root of soybean seedlings. The second burst of ethylene production at PSP was smaller in magnitude than the earlier one at EEP, but was followed by increases in both guaiacol peroxidase (GuPOX) and ascorbate peroxidase (AsPOX). The increase in AsPOX activity was also preceded by an increase in lipoxygenase (LOX) activity. Treatment of the tissue with the ethylene antagonist 2,5-norbonadiene (NBD) resulted in substantial reduction in LOX and AsPOX activities during this period. GuPOX activity was reduced only slightly, if any, by NBD. Role of ethylene in the sequential induction of LOX and AsPOX in senescing callus tissue is discussed.

        Currently, information and communication technology provides, within a very short time, user friendly environment without limitation of time and geographical location to the people who try to search traffic information. Especially, rapidly developed wireless communication services such as wireless internet and Wibro, and Web-based traffic information introduced by Web 2.0 concept are considered as the most potential usage, effectiveness, and customized traffic information tool in the next generation. Based upon the background, this study tried to introduce web based traffic information reflected by recent developed techniques and concepts, and assist to establish it as one of the most effectiveness traffic information device. Firstly, this study examined and analyzed the users’ behavior and their satisfaction associated with current web-based traffic information. Secondly, this study investigated the user preference behavior implementing Stated Preference (SP) survey method as if they were provided traffic information same as Web 2.0 concept. Finally, as it was implicated, this study proposed a business strategy and a policy to substantiate the traffic information platform in Gyeonggi-Do from the analysis and results.
