Effect of a polyherbal formulation on anxiety and behaviour mediated via monoamine neurotransmitters

We investigated the effect of Arogh, a polyherbal formulation (PHF) on animal models of anxiety based on exploratory behavior. The anxiolytic activity of polyherbal formulation (30, 100, 300 and 500 mg/kg) was studied using various behavioural paradigms such as elevated plus maze (EPM), light/dark apparatus (LDA), open field apparatus (OFA), hole board apparatus (HBA). Diazepam (1 mg/kg) was used as a standard anxiolytic drug. The effect of PHF (100 and 300 mg/kg) on serotonin, dopamine and noradrenaline mediated behaviour was studied by lithium induced head twitches in rats, haloperidol induced catalepsy in mice and clonidine induced hypothermia in rats respectively. In EPM, PHF (100, 300 and 500 mg/kg) significantly (P < 0.05) increased the time spent in open arms and the number of entries in open arms. In LDA, PHF (100, 300 and 500 mg/kg) significantly (P < 0.05) increased the time spent in lit zone. In OFA, PHF (100, 300 and 500 mg/kg) significantly (P < 0.05) increased the number of assisted rearing and the number of squares traversed. In HBA, PHF (100, 300 and 500 mg/kg) significantly (P < 0.05) increased the number of head poking. In lithium induced head twitches, PHF (100 and 300 mg/kg) significantly (P < 0.05) decreased the number of head twitches. In haloperidol induced catalepsy, PHF (300 mg/kg) decreased the duration of catalepsy significantly (P < 0.05) at 60 min. In clonidine-induced hypothermia, PHF (300 mg/kg) did not modify the effect. Drugs must be carefully assessed on EPM test and therefore in the present study EPM is supported by other tests. Present study indicates that Arogh, a polyherbal formulation possess anxiolytic activity. It diminished serotonergic transmission and decreased the duration of catalepsy indicating potentiation of dopaminergic transmission. Thus, Arogh a polyherbal formulation contains bioactive principles which possess anxiolytic activity and modified 5-HT and DA mediated behaviour.
Estimation of Biomass and Carbon Stocks of Trees in Javadhu Hills, Eastern Ghats, India

Tropical dry forests are one of the most threatened, widely distributed ecosystems in tropics and estimation of forest biomass is a crucial component of global carbon emission estimation. Therefore, the present study was aimed to quantify the biomass and carbon storage in trees on large scale (10, 1 ha plots) in the dry mixed evergreen forest of Javadhu forest of Eastern Ghats. Biomass of adult (≥10 cm DBH) trees was estimated by non-harvest methods. The total biomass of trees in this tropical dry mixed evergreen forest was ranged from 160.02 to 250.8 Mg/ha, with a mean of 202.04±24.64 Mg/ha. Among the 62 tree species enumerated, Memecylon umbellatum accumulated greater biomass and carbon stocks (24.29%) more than the other species in the 10 ha study plots. ANOVA revealed that there existed a significant variation in the total biomass and carbon stock among the three plant types (Evergreen, brevi-deciduous and deciduous (F (2, 17)=15.343, p<0.001). Basal area and density was significant positively correlated with aboveground biomass (R2 0.980; 0.680) while species richness exhibited negative correlation with above ground biomass (R2 0.167). Finding of present study may be interpreted as most of the trees in this forest are yet to be matured and there is a net addition to standing biomass leading to carbon storage.
Amyloglucosidase Catalyzed Syntheses of Bakuchiol Glycosides in Supercritical Carbon Dioxide

Enzymatic syntheses of water soluble Bakuchiol glycosides were carried out in di-isopropyl ether organic media using amyloglucosidase from Rhizopus mold. The reactions were carried out under conventional reflux conditions and in supercritical CO2 atmospheric conditions. Out of the eleven carbohydrate molecules employed for the reaction, D-glucose, D-ribose and D-arabinose gave glycosides in yields of 9.0% to 51.4% under conventional reflux conditions. Under supercritical CO2 atmosphere (100 bar pressure at 50 oC), bakuchiol formed glycosides with Dglucose, D-galactose, D-mannose, D-fructose, D-ribose, D-arabinose, D-sorbitol and D-mannitol in yields ranging from 9% to 46.6%. Out of the bakuchiol glycosides prepared, 6-O-(6-D-fructofruranosyl)bakuchiol showed the best antioxidant (1.4 mM) and ACE inhibitory activities (0.64 mM).

Effect of ring and straight stiffeners in the buckling as well as vibration characteristics of metal-ceramic functionally graded plates with cutout subjected to various uniaxial and localized in-plane compressive edge loadings was explored in the present work. In the current work, the distinguishing characteristics of metal and ceramic are merged in a single volume, and power law was used for estimating the material composition throughout thickness. Buckling and free vibration characteristics were studied initially for unstiffened Al/SiC functionally graded plates with cutout. Subsequently, the influence of cutout ratio on buckling load as well as natural frequency for different power law indices was discussed. The functionally graded plate was stiffened by three different stiffener patterns, namely; ring stiffener, straight stiffener, as well as a combination of the ring and the straight stiffener, to enhance the buckling as well as vibration characteristics. The effect of stiffener depth ratio for different stiffener patterns was also presented for functionally graded plates having different cutout sizes under various loading conditions. Such studies on functionally graded material have potential applications in a variety of technological fields including the aerospace and defense sectors.

Protective effect of green tea extract on doxorubicin induced cardiotoxicity in rats

Doxorubicin induces oxidative stress leading to cardiotoxicity causing electrocardiogram abnormalities and increases in biomarkers associated with toxicity. Green tea extract (GTE) is reported to possess antioxidant activity mainly via its polyphenolic constituent, catechins. This study was intended to determine the effect of various doses of GTE (25, 50 and 100 mg/kg/day p.o. for 30 days) on doxorubicin-induced electrocardiographic and biochemical changes in rat heart. The latter included lactate dehydrogenase, creatine kinase, and glutamic oxaloacetate transaminase in serum and superoxide dismutase, catalase, and reduced glutathione, as well as membrane bound enzymes like $Na^+K^+ATPase,\;Ca^{2+}ATPase,\;Mg^{2+}ATPase$ and decreased lipid peroxidation in heart tissue Results demonstrated that rats which received GTE were less susceptible to such changes indicating protection afforded by GTE.

We investigated the antihypertensive effect of Pet ether extract (PE) of ginger rhizome; its toluene fraction (TF) and Korean ginseng extract (KGE) in deoxycorticosterone acetate (DOCA) - salt induced and fructose induced hypertensive rats. In DOCA model, DOCA (25 mg/kg, once a week; s.c) was administered in uninephrectomised animals for 4 w. PE (50 mg/kg/day; p.o), TF (10 mg/kg/day; p.o) and KGE (30 mg/kg/day; p.o) were evaluated for their antihypertensive effect. In the fructose model, drinking water was replaced with fructose (10%) for five weeks to induce hypertension. PE (50 mg/kg/day; p.o) and KGE (30 mg/kg/day; p.o) were assessed for its antihypertensive effect in fructose model. After completion of the treatment schedule, vascular reactivity to various agonists like 5-HT, noradrenaline, adrenaline, phenylbiguanide and acetylcholine were recorded in rats of both the models. A cumulative dose response curve (CDRC) of 5-HT was carried out in isolated rat fundus strip of the fructose induced hypertensive rats. Chronic administration of PE (50 mg/kg/day; p.o), TF (10 mg/kg/day; p.o), and KGE (30 mg/kg/day; p.o) significantly reduced the blood pressure in DOCA salt whereas PE (50 mg/kg/day; p.o) and KGE (30 mg/kg/day; p.o) reduced the blood pressure in fructose induced hypertensive rats. Treatment with PE (50 mg/kg/day; p.o) and KGE (30 mg/kg/day; p.o) in fructose model for five weeks shifted the CDRC towards the right on rat fundus. The mechanism of action may partly involve the serotonergic antagonistic property.

Anxiolytic activity of standardized extract of Korean ginseng - a study on exploratory behavior

The roots of the plant Korean ginseng have been extensively used in the traditional Chinese herbal medicine. We investigated the standardized extract of Korean ginseng on animal models of anxiety based on exploratory behavior. Korean ginseng extract (KGE) (3, 10 and 30 mg/kg) was administered intra-peritoneally. The anxiolytic activity was studied using elevated plus maze (EPM) paradigm, light/dark apparatus (LDA), open field apparatus (OFA) and the hole board apparatus (HBA). Diazepam (1mg/kg) was used as a standard anxiolytic drug. In EPM, KGE (10 mg/kg) significantly (P < 0.05) increased the time spent in open arms and the number of entries in open arms. In LDA, KGE (10 mg/kg) increased the number of transitions. In OFA, KGE (3 and 10 mg/kg) significantly increased (P < 0.05) the number of squares traversed. In HBA the number of head pokes were significantly increased with KGE (3 and 10 mg/kg). KGE at all selected doses did not affect the motor coordination. Thus, the study suggests that saponin containing standardized Korean ginseng extract possess anxiolytic activity.
