        Dual aptamer-functionalized silica nanoparticles for the highly sensitive detection of breast cancer

        In this study, we synthesized dual aptamer-modified silica nanoparticles that simultaneously target two types of breast cancer cells: the mucin 1 (MUC1)(+) and human epidermal growth factor receptor 2 (HER2)(+) cell lines. Dual aptamer system enables a broad diagnosis for breast cancer in comparison with the single aptamer system. The dye-doped silica nanoparticles offer great stability with respect to photobleaching and enable the accurate quantification of breast cancer cells. The morphological and spectroscopic characteristics of the designed Dual-SiNPs were demonstrated via diverse methods such as DLS, zeta potential measurements, UV-vis spectroscopy, and fluorescence spectroscopy. Negatively charged Dual-SiNPs with a homogeneous size distribution showed robust and strong fluorescence. In addition, Dual-SiNPs did not affect cell viability, implying that this probe might be readily available for use in an in vivo system. Through ratio optimization of the MUC1 and HER2 aptamers, the binding capacities of the Dual-SiNPs to both cell lines were maximized. Based on Dual-SiNPs, a highly sensitive quantification of breast cancer cells was performed, resulting in a detection limit of 1cell/100μL, which is significantly lower compared with those reported in other studies. Moreover, the developed detection platform displayed high selectivity for only the MUC1(+) and HER2(+) cell lines. It is expected that this valuable diagnostic probe will be a noteworthy platform for the diagnosis and prognosis of breast cancer.

      • 초등학교 1학년 통합단원 "함께하는 한가위" 재구성 실행연구

        본 연구는 초등학교 1학년 2학기 ``바른 생활``, ``슬기로운 생활``, ``즐거운 생활``과의 통합 단원인 ``함께하는 한가위``를 통합적 관점에서 재구성하여 실행한 연구이다. 34명의 초등학교 1학년 학생을 대상으로 재구성한 통합단원을 교사연구자가 실행하는 과정에서 일어나는 교사의 교육과정 재구성에 대한 인식과 학생들의 수업태도 및 참여의 변화를 이해하고 묘사하는데 초점을 두었다. 통합단원의 재구성은 한 차시의 내용에 ``바른 생활``, ``슬기로운 생활``, ``즐거운 생활``과의 내용을 학생들의 생활과 밀접한 통합 활동으로 재구성하여 통합교과가 한 단원 당 2주간 배정된 시량인 총 22차시로 재구성하여 적용하였다. 이를 위해 교수·학습과정안, 학생용 워크북, 교사용 수업자료 등을 개발하였다. 자료 수집은 연구일지, 참여관찰과 비구조화된 면담, 실행과정에서의 실행일지, 학생들이 작성한 학습일지, 동영상 및 사진촬영 등을 통해 이루어졌다. 연구를 통해 학생들은 학습에 있어 함께하는 방법을 알고, 학교는 즐거운 배움터라는 인식을 하였으며, 교사의 변화로는 학습자 중심 교육에 대한 인식변화, 학생을 바라보는 관점의 변화, 교육과정 재구성 시도에 대한 두려움의 극복 등이 있었다. This is an action research on restructuring the integrated unit, ``Sharing together Hangawee`` that consisted the same theme across three integrated subjects. The purpose of this study was to describe and understand the changes of the action researcher`s awareness of curriculum restructuring and of the student participants` attitudes toward learning and classroom participation. Restructuring the integrated unit was mainly done by the way of designing activities closely related with students` real life and three integrated subjects of the first grade. The unit consisted of twenty-two class periods which was the standardized class period for one unit. For the implementation, teaching materials such as lesson plans, student workbook, powerpoints, hand-outs were developed and applied. Based on the research, the changes of the student were ``knowing how to cooperate with peers`` and ``recognizing school as a place of enjoyable learning``. The action researcher changes her paradigm on student-centered instruction and on the viewpoints of the student as a learner. In addition, her ability of curriculum restructuring was improved.

        심리기술훈련이 경쟁 상태 불안과 운동 수행에 미치는 영향

        The purpose of this study was to investigate the effects of psychological skills training on competitive state anxiety and sport performance. The participants were 76 physical education students (40 males, 36 females) ranging in age from 18 to 27 from the Dong-shin University in Naju city. In order to measure competitive state anxiety, the Competitive State Anxiety Inventory-2 (CSAI-2) was used. It was composed of 27 questions covering cognitive, physical instabilities and self-confidence respectively with 9 questions. To measure sports performance, the participants were performed golf putting. Prior to performing the golf putting, the participants completed the CSAI-2 to measure competitive state anxiety and a test of golf putting. After pre-test, the participants divided randomly four different groups to received 16 weeks psychological skills training. One of the groups was a control group and the others were three experimental groups. Three experimental groups were performed a 16 weeks psychological skills training program by using the Progressive Muscle Relaxation and the Imagery Training. The control group was not contacted in any way during the 16 weeks. After 16 weeks, the participants were re-tested using the same methods as in the pre-test to examine the effects of the psychological skills training. The results were as follows: 1) The psychological skills training programs were effective in reducing anxiety levels of the participants. Three experimental groups were meaningful for reducing his/her cognitive anxiety. Its were also effective for lessening his/her physical instability and improving self-confidence. 2) The psychological skills training programs were effective in improving the golf putting scores. The score gap between the control group and three experimental groups were particularly large. This means that the psychological skills training programs were affected to his/her competitive state anxiety, thereby contributing to improving scores in the Golf putting' performance.

        Integrated chassis control for optimized tyre force coordination to enhance the limit handling performance

        <P>This paper proposes a coordinated control algorithm of the differential braking, the front and rear traction torques and the active roll moment to enhance the limit-handling performance. The coordinated algorithm is designed to maximize the driving velocity while keeping the vehicle in a lane. First, the analysis of the cornering dynamics is described to consider the non-linear characteristics of the tyres during acceleration or deceleration. The target vehicle motions are determined on the basis of the driver's intention and the current states of the vehicle. An optimization-based control allocation strategy is utilized to distribute the actuator control inputs optimally by considering the tyre and vehicle limitations. Closed-loop simulations of a driver-vehicle-controller system were conducted to investigate the performance of the proposed control algorithm. The performance of the coordinated algorithm was compared with those of the individual coordination algorithms. The simulation results show that the proposed integrated chassis controller improves the performance in high-speed cornering with respect to the driving speed without losing stability compared with simple coordination chassis control systems.</P>

        Enzyme-linked antibody aptamer assays based colorimetric detection of soluble fraction of activated leukocyte cell adhesion molecule

        <P><B>Abstract</B></P> <P>Patients with pancreatic and colorectal cancers have elevated levels of activated leukocyte cell adhesion molecule (ALCAM) in their blood compared to healthy individuals. To this end, sensitive detection of soluble fraction of ALCAM (sALCAM), also called CD166, was performed using enzyme-linked antibody aptamer (ELAA) assay. For the assay, aptamer specific for sALCAM was used as a capturing probe and polyclonal antibody modified with an enzyme was used as a detecting probe for colorimetric visualization. The ELAA assay was able to detect sALCAM as low as 0.31ng/mL. In addition, the assay enabled the differentiation of sALCAM from other proteins in buffer as well as in human serum. The ELAA assay provides cost-effective, highly sensitive, and reproducible detection of sALCAM.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Schemes of enzyme-linked antibody aptamer assays are proposed. </LI> <LI> The enzyme-linked antibody aptamer assay was able to successfully differentiate sALCAM among other proteins. </LI> <LI> The enzyme-linked antibody aptamer assay presents an alternative way of detecting sALCAM. </LI> </UL> </P>

        An Integrated Control of Differential Braking, Front/Rear Traction, and Active Roll Moment for Limit Handling Performance

        <P>This paper describes an integrated chassis control (ICC) algorithm of differential braking, front/rear traction torque, and active roll moment control. The integrated control algorithm is designed to maximize driving velocity and enhance vehicle lateral stability in cornering. The target longitudinal acceleration is determined based on the driver's intention and vehicle current status to ensure vehicle lateral stability in high-speed maneuvering. An optimization-based control allocation strategy is used to distribute the actuator control inputs optimally under consideration of tire and vehicle limitations. Closed-loop simulations of a driver-vehicle-controller system were conducted to investigate the performance of the proposed control algorithm. The performance of the ICC has been compared with those of individual chassis control systems, such as electronic stability control, four-wheel drive (4WD), and active roll control system. The simulation results show that the proposed ICC algorithm improves the performance in high-speed cornering with respect to driving speed without losing stability, compared with individual chassis control systems.</P>

      • Formula for estimating the uncertainty of manufacturer's power curve in pitch-controlled wind turbines

        <P>This study proposes a new formula for estimating the uncertainty in pitch-controlled wind turbines regardless of the type of wind turbines or the availability of measurement data. The results obtained using a manufacturer's power curve (MPC), which is defined as the representative value of the power performance of wind turbines, may differ from the prediction results of power generation obtained using an actual power curve measured at a real wind farm. Therefore, the uncertainty of the MPC can considerably influence the calculation of power generation. However, the current methods used for uncertainty calculation either use a random constant chosen by the judgment of individual analyzers based on their experience or rely on a limited set of empirical formulas suggested through the analysis of data measured from specific wind turbines. To solve this problem, we suggest a new formula for uncertainty estimation that has general applicability. The suggested formula was validated by comparing its results with the data output measured from a real wind farm. In addition, a comparison of the real-time estimation results of wind power generation and measurement results demonstrates that the proposed formula produces more reliable results than the existing formula.</P>

        Estimating USLE Soil Erosion through GIS-based Decision Support System

        The objective of this study was to develop a GIS-based decision support system (GIS-USLE system) to estimate soil erosion and evaluate its effect on concentrated upland plots in Godang district, Korea. This system was developed for the ArcView environment using A VENUE script. Three modules were used in the GIS-USLE system, namely pre-processing, the USLE factors calculator module, and post-processing. This system benefits from a user friendly environment that allows users with limited computer knowledge to use it. This system was applied to 1,285 individual upland plots ranging from 0.005 to 1.347 ha in size with an average slope steepness of 14 %. The rainfall distributions were estimated using the three methods, namely Mononobe and Yen-Chow with Triangle and with Trapezoid type, and then used to calculate the rainfall erosivity factor. The soil erosion amounts from the 1,285 individual plots in the study area by 2 year return period with a 24h maximum rainfall amount of 154.6 mm were estimated at 5 tons/ha on average. Slope appeared to be the most important factor affecting soil erosion estimation, as expected. The prototype model was applied to the project area, and the results appeared to support the practical applications. By examining many fields simultaneously, this system can easily provide fast estimation of soil erosion and thus reveal the spatial pattern of erosion from fields in a region. This study will help estimate and evaluate soil erosion in concentrated upland districts and identify the best management practices.

        <P>The adrenergic and serotonergic stimulations of rat C6 glioma cells have previously been shown to induce the activation of steroid 5alpha-reductase (5alpha-R) gene expression, resulting in their differentiation through the production of neuroactive 5alpha-reduced steroid metabolites. In addition, progesterone and histone deacetylase (HDAC) inhibitors have also been reported to promote the glial cell differentiation with the enhancement of serotonin-stimulated brain-derived neurotrophic factor gene transcription through the production of 5alpha-reduced neurosteroids, thus suggesting that glial cell differentiation is probably implicated in the protection and survival of neuronal cells in the brain. Therefore, the expression of 5alpha-R gene in glial cells seems physiologically important in maintaining the neural function in the brain, but little is known about the mechanism underlying the regulation of 5alpha-R gene transcription. In the present study, the effect of a HDAC inhibitor trichostatin A (TSA) on 5alpha-R gene transcription in the glioma cells was examined, and TSA was shown to induce the elevation of 5alpha-R mRNA levels through the activation of the 5alpha-R promoter via a mechanism involving Sp1 and Sp3 transcription factors in a time- and concentration-dependent manner. Thus, both Sp1 and Sp3 are considered to play a physiological role in the regulation of 5alpha-R gene expression, and hence the production of 5alpha-reduced neurosteroids in glial cells.</P>
