Sulfonated pomegranate (Punica granatum) peel biochar (SPPBC) was developed via thermal activation with sulfuric acid (H2SO4) to act as a promising biochar material for the adsorptive removal of toxic cationic dye namely methylene blue (MB) dye from contaminated water. A Box-Behnken design (BBD) and numerical desirability function were adopted to optimize the input adsorption variables (SPPBC dosage, temperature, pH, and contact time). The maximum removal of the MB dye can be accomplished by simultaneous significant interaction between SPPBC dosage with solution pH, SPPBC dosage with time, SPPBC dosage with temperature, solution pH with time, and time with temperature. The numerical desirability function identified the highest MB dye removal (93.9%) can be achieved at the following optimum numerical adsorption conditions: SPPBC dosage 0.18 g, temperature 49 oC, pH 9.7, and time 4.3 h. Equilibrium data were well fitted to the Temkin and Langmuir isotherm models. The maximum recorded adsorption capacity of SPPBC for MB dye adsorption by using Langmuir isotherm model was 161.9mg/g. This research work reveals the possibility of converting lignocellulose pomegranate peel into a renewable and environmentfriendly biochar via a relatively fast acid-activation process with the great potential to be promising adsorbent for removal of MB dye.

Octopamine (OA) and tyramine (TA) are biogenic amines that act as neurotransmitters, neurohormones, andneuromodulators in the invertebrate nervous system. Tyramine-β-hydroxylase (TβH) catalyzes the biosynthesisof OA from TA. In this study, cDNA encoding Bombyx mori TβH (BmTβH) was cloned from the brain of the silkwormB. mori. The BmTβH mRNA comprised 2204 nucleotide residues and contained an open reading frameencoding 592 amino acids. The deduced amino acid sequence shared homology to several proteins belongingto the insect TβH family. Functional expression of the cloned cDNA was obtained using a B. mori baculovirus expressionvector system. Western blot analysis revealed an immunoreactive band with a molecular mass of~67.4 kDa. Reverse-phase high-performance liquid chromatography (HPLC) was used to identify the productsformed during incubation of the enzyme reaction mixture. The optimumpH and temperature for the conversionof TA to OA were 7.5 and 25 °C, respectively. During incubation, the reaction was linear for the first 30 min at25 °C and pH 7.5. Inhibitory experiments carried out with various concentrations of an inhibitor showed thatthis method can be used for screening of BmTβH inhibitors.

The insect nervous systemcontains biogenic amines such octopamine (OA),which is synthesized fromtyramine (TA) by catalysis of tyramine-β-hydroxylase (TβH). In this study, the Drosophila 70 kDa tyramine-β-hydroxylase (DmTβH) protein was purified after the recombinant nucleopolyhedrovirus isolated from Bombyx mori (BmNPV) containing the TβH gene was injected into the hemocoel of the fifth instar larvae from the d17B. mori strain. Western blot analysis revealed an immunoreactive band with a molecular mass of 70 kDa. The products formed by incubating the enzyme reaction mixture were separated and detected by reverse phase high-performance liquid chromatography. The optimum pH, temperature, and incubation time for the conversion of TA to OAwere 7.6, 25 °C, and 30 min, respectively. The inhibitory experiments using various concentrations of 1-(2-methoxy-5-methylphenyl) imidazole-2(3H)-thione (MMIT) showed that MMIT inhibited DmTβH dose-dependently and that this method can be applied for screening DmTβH inhibitors.

Flexural performance of cold-formed square CFST beams strengthened with internal stiffeners

The tube outward local buckling of Concrete-Filled Steel Tube (CFST) beam under high compression stress is still considered a critical problem, especially for steel tubes with a slender section compared to semi-compact and compact sections. In this study, the flexural performance of stiffened slender cold-formed square tube beams filled with normal concrete was investigated. Fourteen (14) simply supported CFST specimens were tested under static bending loads, stiffened with different shapes and numbers of steel stiffeners that were provided at the inner sides of the tubes. Additional finite element (FE) CFST models were developed to further investigate the influence of using internal stiffeners with varied thickness. The results of tests and FE analyses indicated that the onset of local buckling, that occurs at the top half of the stiffened CFST beam’s cross-section at mid-span was substantially restricted to a smaller region. Generally, it was also observed that, due to increased steel area provided by the stiffeners, the bending capacity, flexural stiffness and energy absorption index of the stiffened beams were significantly improved. The average bending capacity and the initial flexural stiffness of the stiffened specimens for the various shapes, single stiffener situations have increased of about 25% and 39%, respectively. These improvements went up to 45% and 60%, for the double stiffeners situations. Moreover, the bending capacity and the flexural stiffness values obtained from the experimental tests and FE analyses validated well with the values computed from equations of the existing standards.
Micro-grid Monitoring and Supervision: Web-based SCADA Approach

Owing to the widespread use of the micro-grid concept to serve many real life applications, the main concern of this paper is to monitor, evaluate and manage the operational performance of an existent, already installed micro-grid that consists of On & Off grid PV systems in addition to the main grid supply. With the aid of customized web based SCADA system fully designed by authors, the performance evaluation of the proposed micro-grid is carried out in terms of energy consumption, power quality indices and energy cost based on Egypt’s energy current tariff . The designed system helps in both micro-grid status monitoring as well as load sharing between the main grid supply and the off grid PV energy system. The data of the on grid PV energy system are accessed and collected online directly from the inverter web server instead of downloading them offl ine from the PV inverter. In this case, data are analyzed with a reasonable refresh rate, which saves time and eff ort. Remote access platform is developed securely through a multi-level security authentication and strong data encryption using the OpenVPN protocol under the Dynamic Domain Name System service. Smart wired and wireless technology—used as integrated sensing devices to the developed SCADA system—enables easy system installation and running cost saving. Finally, to validate the collected data from the newly SCADA designed monitoring system, a performance characterization for the on grid PV system following international standards of yields calculations is carried out for 3 months in 2019.

In this paper, we prove common fixed point theorems for $\mathcal{L}$-fuzzy mappings under implicit relation in b-metric spaces. Further, results obtained for an integral type contractive condition. These theorems generalize and improve previous corresponding results.
Anti-cancer Effect of Hyoscyamus muticus Extract via Its Activation of Fas/FasL-ASK1-p38 Pathway

Hyoscyamus muticus L. is a traditional medicine used as antispasmodic and sedative. Herein, we aimed to determine the phytochemical constituents and for the first time its anti-cancer activities. The phytochemical constituents of the different extracts were evaluated by calorimetric methods. The anti-cancer activities of the extracts were tested against leukemia, breast, renal, and prostate cancers cell lines. 4, 6-Diamidino-2-phenylindole (DAPI) staining, flow cytometric analysis, knockdown of ASK1, and reactive oxygen species (ROS) production were evaluated to clarify the mechanism of action. Phytochemical screening confirmed the presence of wide range of phytoconstituents. Hyoscyamus muticus methanolic extracts (HMME) showed the highest anti-cancer activities against leukemia, breast, renal, and prostate cancers as compared to ethanol and aqueous extracts. Specifically, HMME exerted cytotoxic effect against the MDA-MB-231 and MDA-MB-468 triple-negative breast cancer (TNBC) cell lines with IC50 values of 8.75 and 7.25 μg/mL, respectively. Mechanistically, DAPI staining and flow cytometric analysis revealed that HMME induces apoptosis via the death receptor, FAS, but not the mitochondrial pathway. Moreover, ASK1 and p38 were rapidly activated in response to HMME, and knockdown of ASK1 by a small interference of RNA specific to Ask1 attenuated p38 and caspase-3 activation and suppressed apoptosis, implying that HMME-induced apoptosis relies on the ASK1-p38-caspase-3 pathway. Furthermore, we confirmed that cellular ROS generation was a critical mediator in HMME-induced apoptosis because the ROSscavenger N-acetyl cysteine significantly decreased the phosphorylation of ASK1 and HMME-induced apoptosis. Our results confirmed HMME cytotoxic effects in TNBCs via ROS-dependent activation of the Fas/FasL-ASK1-p38 axis.
