        Metabolomic analysis of biochemical changes in the tissue and urine of proteoglycan-induced spondylitis in mice after treatment with moxibustion

        Background: Moxibustion is widely used in East Asian countries to manage the symptom of rheumatic diseases. The aim of this study was to identify potential metabolic profiles of moxibustion on relieving ankylosing spondylitis (AS) mice through UHPLC-Q-TOF/MS metabolomic study. Methods: Thirty-two female Balb/c mice were randomized into healthy control (HC), AS model, moxibustion at acupuncture points (MA) in AS model, and moxibustion at non-acupuncture points (MNA) AS model groups. Moxibustion was administered daily at GV4, bilateral BL23 and bilateral ST36 acupuncture points for four weeks in the MA group. The overall health status, the thickness of hind paws and the tissue concentrations of IL-1β, PGE2, IL-6 and TNF-α were assessed. The UHPLC-Q-TOF/MS was used to explore the perturbations of endogenous metabolites in tissue and urine of AS model mice intervened by moxibustion. Results: Compared with the AS group, the overall health status was significantly improved after 4-week moxibustion intervention (p < 0.05). The results also showed that MA significantly reduced the levels of paw thickness and decreased the levels of four cytokines in the tissue (p < 0.01). Thirty-seven endogenous metabolites identified by the OPLS-DA were considered to be contributing to therapeutic effects of moxibustion. Moreover, metabolic pathway analysis further revealed that the identified metabolites were mainly involved in TCA cycle, Lipid metabolism, Amino Acid metabolism, Intestinal flora metabolism and Purine metabolism. Conclusions: UHPLC-Q-TOF/MS based metabolomics approach, as a novel and powerful tool, can help us to gain the insights into potential mechanisms of action of moxibustion for AS.

      • miR-10b Promotes Migration and Invasion in Nasopharyngeal Carcinoma Cells

        MicroRNA-10b (miR-10b) has been reported to play an important role in some types of cancer, but the effects and possible mechanisms of action of miR-10b in the metastasis of nasopharyngeal carcinoma cells (NPC) have not been explored. The aim of the present study was to investigate the function of miR-10b in nasopharyngeal carcinoma and to determine the molecular mechanisms underlying its action. The MTT assay was used to assess proliferation of CNE-2Z cells. Wound healing and transwell migration assays were applied to assess cell migration and invasion, while and expression of E-cadherin and MMP-9 were detected using Western blot analysis. Real-time PCR was employed to detect the expression of genes related to migration and invasion and the $2^{-{\Delta}{\Delta}Ct}$ method was used to calculate the degree of expression. MTT assay showed the expression of miR-10b to have no effect on the proliferation of NPC cell lines. The wound healing assay showed that miR-10b mimics promoted the mobility and invasion of NPC cell lines. Inhibitors of miR-10b reduced the ability of NPC cell lines to migrate and invade. In addition, the expression of genes related to migration and invasion, such as E-cadherin, vimentin, and MMP-9, were confirmed to be different in the CNE-2Z NPC cell line transfected with miR-10b mimics and with miR-10b inhibitors. In the present study, miR-10b was found to upregulate the expression of MMP-9 and knockdown of miR-10b was found to significantly downregulate the expression of E-cadherin. On the whole, these results showed that miR-10b plays an important role in the invasion and metastasis of NPC cells.

        Cross-sectional geometry dependence of spontaneous phase transformation of copper nanowires

        Molecular dynamics simulations have been performed to investigate the spontaneous phase transformation of copper nanowires. It is found that the spontaneous phase transformation exhibits distinct dependence on the cross-sectional geometry of the nanowires and can lead to the reconstruction of atoms into different atomistic configurations, e.g., pure hexagonal-close-packed crystals, fivefold deformation twins, and core/shell structures. For single-crystal copper nanowires, the critical crosssectional size, above which no spontaneous phase transformation can occur, is determined. The physical mechanisms underlying the complicated transformation behavior are analyzed from the viewpoints of energy and stresses.

        Power series solution of circular membrane under uniformly distributed loads: investigation into Hencky transformation

        In this paper, the problem of axisymmetric deformation of the circular membrane fixed at its perimeter under the action of uniformly-distributed loads was resolved by exactly using power series method, and the solution of the problem was presented. An investigation into the so-called Hencky transformation was carried out, based on the solution presented here. The results obtained here indicate that the well-known Hencky solution is, without doubt, correct, but in the published papers the statement about its derivation is incorrect, and the so-called Hencky transformation is invalid and hence may not be extended to use as a general mathematical method.

        Microscopic Mechanisms and Acoustic Emission Characteristics of Sandy Mudstone under Different Water Saturations

        In order to study the damage mechanism of surrounding rocks under water-rock coupling, an experimental study on the microscopic law of water absorption and mechanics of sandy mudstone was carried out in the sandy mudstone stratum of Wanfu coal mine in Shandong, China. The results indicate that the strength of sandy mudstone exhibits three stages of relative softening, rapid softening, and decelerated softening. The speed of water molecules entering the microscopic pore structure was analyzed based on the percentage of water molecules in the pore structure. The acoustic emission energy release process is divided into energy accumulation stage, stable release stage and intense release stage. With the increase of water content saturation,  (Spectral width of the multifractal spectrum) tends to decrease and the distribution of events is gradually uniform. The action of water gradually expands and penetrates the cracks and pores inside the sandy mudstone, which makes f (Frequency of occurrence of the signal) change from negative to positive values.

        Steam reforming of α-methylnaphthalene as a model compound of biomass tar over Ni-based catalyst for hydrogen-rich gas

        Tar is a barrier to limit the development of biomass gasification. Catalytic steam reforming experiments using α-methylnaphthalene (MNP) as a model tar compound were carried out in the two-stage reactor system (TSR). Based on response surface methodology, the effects of TSR temperatures and the molar ratio of steam to carbon (S/C) on MNP reforming performances were analyzed using the Li-modified Ni-based catalyst (NBC). The results show that the proper introduction of H2 is able to improve significantly the MNP conversion, specially at lower temperatures. Furthermore, it is more appropriate for the modified catalysts by Li and Mg to be loaded in the first reactor due to their significant promotion to hydrocracking reactions, and it is favorable to place the Ni/Al catalyst in the second reactor for H2-rich gas. Additionally, the carbon deposition resistance of the NBC modified by Li exhibits better than that of the NBC modified by Mg.

        Multiple-motion mode switching robot platform

        Developing an intelligent robot platform that has multiple motion modes and can actively adjust the motion mode according to an operation situation is necessary in resolving the inability of single-mode robots to adapt completely to complex environments. In this study, we designed a robot that has multiple motion modes and can switch freely in these existing modes. The robot has the following modes:Four-legged walking, two-legged walking, pulse scrolling, and crawler movement. In the robot’s design, these modes are not just simple mechanisms involving superposition and motion switching. These modes involve a degree of freedom in reuse, thereby reducing the number of drives and obtaining a compact structure while achieving a coherent transition of each action mode. For higher load ratios, motion continuity are increased. The robot platform can balance itself during movement by adjusting its own mechanism or providing additional force and change its shape and posture over some obstacles by adjusting its own mechanism.

        Molecular cloning and characterization of two novel DREB genes encoding dehydration-responsive element binding proteins in halophyte Suaeda salsa

        The dehydration-responsive element-binding(DREB) proteins play an important role in regulatingexpression of stress-inducible genes under abiotic stresses. In this study, two genes encoding putative DREB proteins,named SsDREBa and SsDREBb, were cloned from halophyteSuaeda salsa L. using RACE method. The deducedSsDREBa and SsDREBb proteins contain a typical AP2/ERF domain. Multiple sequence alignments and phylogeneticanalysis revealed that the two SsDREB genes of S. salsa were highly similar in AP2/ERF domains at thenucleotide and amino acid levels and belong to the A-6subgroup of the DREB transcription factor subfamily. Asubcellular localization assay showed that both SsDREBslocalized to the nucleus. Yeast one-hybrid experimentstestified that both proteins were able to specifically bind tothe DRE sequence and activate the expression of the downstreamHIS reporter gene in yeast. Quantitative real-timePCR analysis demonstrated that under normal conditions,the expression level of SsDREBa was the most high in theroots and no SsDREBa mRNAs were detected in the stems;SsDREBb expressed at relatively higher levels in the leavesthan in the roots and stems. The expression of SsDREa andSsDREBb genes in S. salsa roots and leaves was remarkablyinduced by high-salt and dehydration treatments, butnot by cold and ABA, and exhibited stronger induction inroots and leaves, respectively. These results indicate thatthe SsDREBa and SsDREBb are novel stress-responsivetranscription factors, which are involved in the drought andhigh-salt stress responses through ABA-independent pathwaysand could be used for production of stress-toleranttransgenic crops.

      • Single Nucleotide Polymorphisms in the NER Pathway and Clinical Outcome of Patients with Bone Malignant Tumor

        The effects of polymorphisms in ERCC5, ERCC6, XPC, CCNH and MMS19L on osteosarcoma response to chemotherapy and the survival of the affected patients were assessed. Genotyping of ERCC5, ERCC6, XPC, CCNH and MMS19L was performed by PCR-RFLP assay. The median PFS was 12.8 months, and the median OS was 18.6 months. Individuals carrying homozygous genotypes of ERCC5 rs17655 and ERCC5 rs1047768 were more like to have good response to treatment, while those carrying homozygous genotypes of MMS19L rs29001322 showed poor response. Osteosarcoma patients carrying TT genotype of ERCC5 rs1047768 showed a significantly longer PFS (16.8 months) and OS (21.4 months) than CC genotype, with HRs(95% CI) of 0.31 (0.10-0.93) and 0.32 (0.06-0.97), respectively. Conversely, those with the TT genotype of MMS19L rs29001322 demonstrated shorter PFS and OS, the HRs (95% CI) being 2.23 (1.08-4.15) and 4.62 (1.45-16.08), respectively. Our findings showed polymorphisms in ERCC5 rs1047768 and MMS19L rs29001322 to be associated with clinical outcome of osteosarcoma patients undergoing chemotherapy.
