<P>Ginsenoside Rg3 (Rg3), one of the most effective ginseng saponins, has anti-inflammatory and anti-cancer effects. This study examined the effects of Rg3 on cytokine-induced expression of adhesion molecules, which is a key early event in atherogenesis. Rg3 treatment inhibited tumor necrosis factor-alpha (TNF-alpha)-induced protein and mRNA expression of two cell adhesion molecules, vascular cell adhesion molecule-1 (VCAM-1) and intercellular cell adhesion molecule-1 (ICAM-1) in ECV 304 human endothelial cells. In addition, expression of two pro-inflammatory cytokines, TNF-alpha and interleukin-1beta (IL-1beta), was suppressed by Rg3. Reporter gene analyses revealed that minimal reporter activities of nuclear factor-kappaB (NF-kappaB) and activator protein-1 (AP-1) were blocked by Rg3 in a concentration-dependent manner. Taken together, these results indicate that Rg3 may have anti-inflammatory and anti-atherosclerotic activities in the vasculature, which is mediated partly by down-regulation of the expression of cell adhesion molecules and proinflammatory cytokines in endothelial cells.</P>

<P>Multidrug resistance (MDR) is a major obstacle in cancer chemotherapy and its inhibition is an effective way to reverse cancer drug resistance. In the present study, we investigated that puerarin, a natural isoflavonoid from Pueraria lobata, down-regulated MDR1 expression in MCF-7/adriamycin (MCF-7/adr), a human breast MDR cancer cell line. Puerarin treatment significantly inhibited MDR1 expression, MDR1 mRNA and MDR1 promoter activity in MCF-7/adr cells. The suppression of MDR1 was accompanied by partial recovery of intracellular drug accumulation, leading to increased toxicity of adriamycin and fluorescence of rhodamine 123, indicating that puerarin reversed the MDR phenotype by inhibiting the drug efflux function of MDR1. Moreover, nuclear factor κ-B activity and IκB degradation were inhibited by puerarin. Puerarin stimulated AMP-activated protein kinase (AMPK), acetyl-CoA carboxylase and glycogen synthase kinase-3β phosphorylation, but puerarin decreased cAMP-responsive element-binding protein phosphorylation. The puerarin-induced suppression of MDR1 expression was reduced by AMPK inhibitor (compound C). Furthermore, both MDR1 protein expression and the transcriptional activity of cAMP-responsive element (CRE) were inhibited by puerarin and protein kinase A/CRE inhibitor (H89). Taken together, our results suggested that puerarin down-regulated MDR1 expression via nuclear factor κ-B and CRE transcriptional activity-dependent up-regulation of AMPK in MCF-7/adr cells.</P>

<P>We revealed previously that nectandrin B isolated from Myristica fragrans (nutmeg, Myristicaceae) functions as a potent AMP-activated protein kinase (AMPK) activator and showed its antiobesity effect. In this study, we investigated whether nectandrin B affects phosphorylation of endothelial nitric-oxide synthase (eNOS) in human endothelial cells. Nectandrin B increased the phosphorylation of eNOS and nitric oxide (NO) production in a concentration-dependent manner and maximal effect was found at 10 관g/ml. Nectandrin B activates AMPK, presumably via Ca(2+)/calmodulin kinase II activation and nectandrin B-stimulated eNOS phosphorylation was reversed by AMPK inhibition. Both the enzyme activity of phosphatidylinositol 3-kinase (PI3K) and the estrogen receptor (ER)-dependent reporter gene transcription were enhanced by nectandrin B. ER관 inhibition by specific antagonist or small interfering siRNA (siRNA) suppressed nectandrin B-mediated eNOS phosphorylation. Moreover, AMPK inhibition significantly reversed the activation of ER-dependent transcription and PI3K activation in response to nectandrin B. Nectandrin B evoked endothelium-dependent relaxation in rat aortic rings, and this was blocked by inhibition of AMPK, ER, or PI3K. These results suggest that potent AMPK activator nectandrin B enhances NO production via eNOS phosphorylation in endothelial cells and ER관-dependent PI3K activity is required.</P>

<P>Traditional Chinese medicines have attracted increasing interest as potential sources of novel drugs with a wide range of biological and pharmacological activities. Annona glabra Linn (Annonaceae) is used in traditional medicine as an anticancer drug. Phytochemical investigation of this plant led to the isolation of acetogenins, ent-kauranes, peptides, and alkaloids. In addition, compounds exhibited anticancer, anti-HIV-reserve, and antimalaria.</P>

The world has witnessed the outbreak of the Covid-19 epidemic. Mainstream and social media are playing an important role in Covid-19 pandemic prevention. This research explores awareness, communication channels and effectiveness of communication in the Covid-19 pandemic in rural areas of Thua Thien Hue province, Central Vietnam. Primary information was collected from 181 respondents, who are farmers, non-farmers and students. Secondary information was collected from reports and statistical data. Television, word of mouth and local loudspeakers are the main channels of mainstream media while social media mentions the role of Facebook and Zalo to transfer Covid-19 pandemic information. Mainstream media is still the main channel of farmers and old people while non-farmers and young people tend to access information through social media. Communication has significantly contributed to improving awareness and action of rural people in the Covid-19 epidemic prevention.
