분무 건조 방식으로 제조한 에프로살탄 고체분산체 제조 및 특성 분석

본 연구에서는 분무건조방법을 이용하여 고혈압 치료제로 사용되고 있는 난용성 약물인 에프로살탄의 고체분산체 제형을 다양한 조성 하에서 제조 및 평가하고, 효과적인 약물의 용해도 향상을 위한 제조 조건을 최적화하였다. 친수성 고분자 기제로 hydroxylpropylcellulose(HPC)와 poly(vinyl pyrrolidone)(PVP)를 사용하였고, 고분자 계면 활성제로 poloxamer 407을 첨가하여 약물과 고분자의 다양한 조성비를 갖는 고체분산체를 재조한 후, 물리화학적 특성을 비교 평가하였다. 친수성 고분자의 조성비 증가와 함께 약물 결정성이 감소되었으며, HPC보다는 PVP가 약물과의 우수한 상용성을 바탕으로 결정화도 감소와 용출거동 개선 효과가 상대적으로 우수하였다. 친수성 고분자로 PVP를 사용한 경우 약물의 결정성이 대부분 사라져 고체분산체 내 대부분의 약물 분자들이 무정형으로 분산되어 있으며 약물의 용출률이 에프로살탄 대비 3~7배 이상 향상되었다. In this study, we developed and optimized hydrophilic polymer-based solid dispersion formulations (SDs) using a spray drying method for improving the aqueous solubility of eprosartan, one of poorly soluble drugs that has been broadly used for the treatment of high blood pressure. Hydroxylpropylcellulose (HPC) and poly(vinyl pyrrolidone) (PVP) were used as hydrophilic polymer matrices and poloxamer 407 (P407) added as a polymeric surfactant. Various kinds of solid dispersions with different drug/polymer compositions were prepared and their physico-chemical properties were compared. As the polymer content increased, the drug crystallinity in the SDs significantly decreased and the dissolution properties were enhanced. The PVP based SDs were observed to have relatively reduced crystallinity and an enhanced dissolution rate than HPC-based SDs, due to better miscibility between drug and polymer matrix. For PVP based SDs, the drug crystallinity almost disappeared and the dissolution properties significantly increased by more than 3~7 times.
인터넷 VOD 이용의도에 영향을 미치는 요인에 관한 연구

인터넷과 정보통신 기술이 발전함에 따라 영화 유통의 창구도 다양화 되고 있으며, 인터넷 VOD 극장은 저렴한 가격과 인터넷이라는 시간과 공간의 제약성이 없는 특징을 통해 새로운 영화 창구로서 자리 매김 하고 있다. 본 연구에서는 인터넷 VOD 극장을 이용하는 데 있어서 기존 연구에서 나타난 기술수용이론의 지각된 유용성과 몰입이론의 변수 그리고 인터넷 VOD 극장에서 제공하는 영화에 대해 소비자가 느끼는 흘드백 기간에 대한 민감도와 인터넷 VOD 극장에서 제공되는 영화 장르 선택의 다양성이 소비자의 이용 의도에 미치는 영향에 대해 살펴보았다. 본 연구의 의의는 기존 오프라인 영화관 및 다른 영화매체와의 경쟁 속에서 새로운 유통 창구로 자리매김하고 있는 인터넷 VOD 극장의 매체적인 특성을 찾고, 인터넷 VOD 극장 이용자의 이용 의도에 영향을 미치는 요인을 분석함으로써 향후 인터넷 VOD 극장 수익 발전 방향을 모색했다는 점에서 실용적인 의미가 있을 것이다. The development of the Internet and telecommunication technology has lead to the diversification of the distribution channel of movies. Internet users can easily watch movies through the Internet VOD(Video On Demand) theaters without a restriction of time and space. In this study, we try to understand the intention to use of internet VOD movies using the concepts of Technology Acceptance Model and Flow Model. We also consider the concepts of sensitivity of holdback period and availability of various choice in movie genre, along with demographic factors such as age and gender. Through our study we enhance our understanding on how and when users use the Internet VOD for their movie watching.
친수성 고분자를 이용한 에프로살탄 고체분산체의 제조 및 특성 분석

본 연구에서는 고혈압 치료제로 사용되고 있는 난용성 약물인 에프로살탄의 용해도 개선을 위한 고체분산체 제제를 개발하기 위한 제조방법 및 조성비를 최적화하였다. 친수성 고분자 기제로 poly(ethylene glycol)(PEG)와 poly(vinyl pyrrolidone)(PVP)를 이용하여, 약물과 고분자의 조성비가 1:1에서 1:5 사이의 고체분산체를 용매증발법과 열용융법에 의해 제조한 후 비교 평가하였다. 용매증발법이 균일한 고체분산체 제조에 효과적이었고, 고분자의 조성 비 증가와 함께 약물 결정성이 감소되었다. PEG 보다는 PVP가 약물과의 우수한 상용성을 바탕으로 결정화도 감소 와 용해도 개선 효과가 우수하였다. 또한 고체분산체 제조 시 고분자 계면활성제인 poloxamer 407를 첨가한 경우, 약물의 결정성이 대부분 사라져 고체분산체 내 대부분의 약물 분자들이 무정형으로 분산되어 있음을 알 수 있었고, 약물의 용해도 또한 3~4배 이상 향상되었다. Poly (ethylene glycol) (PEG) and poly (vinyl pyrrolidone) (PVP) based SDs were prepared by hot melting and solvent evaporation methods and the drug/polymer composition varied in the range of 1:1~1:5 with or without poloxamer 407 (P407) as a polymeric surfactant. The SDs prepared by solvent evaporation showed more reduced crystallinity than ones by hot melting, and PVP based SDs showed more enhanced solubility and lower crystallinity than PEG based SDs. Furthermore, it was observed from DSC and PXRD analysis that the SDs with P407 (drug:polymer: P407=1:5:1) demonstrated no crystallinity and the most enhanced solubility (more than 3~4 times).
