The objective of this study is to provide the basic information for developing a high-value ginseng product using ginseng saponin and prosapogenin. In order to achieve such aim, Ginsenoside compositions of black ginseng (BG) extracts with various solvent conditions were examined by HPLC. The total saponin and the prosapogenin content of 95% ethyl alcohol extract were higher than that of the either 50% ethyl alcohol extract or distilled water extract. As a result, the order of the total saponin and the prosapogenin content was 1) 95% ethyl alcohol, 2) 50% ethyl alcohol, 3) the first and second mixture of 95% ethyl alcohol, distilled water, and 4) distilled water extract. In the case of fine black ginseng (FBG), the first and second mixture extracts of 95% ethyl alcohol and distilled water were the highest. In addition, the ratio of the protopanaxadiol group and the protopanaxatriol group (PD/PT) showed that the ratio of BG ranged from 0.304 to 0.601, while the ratio of FBG ranged from 1.166 to 1.657.

Human foamy virus (HFV) integrase mediates integration of viral c-DNA into cellular DNA. In this process, HFV integrase recognizes its own viral DNA specifically and catalyzes insertion of viral c-DNA. In order to study catalytic domains and residues, three deletion mutants and two point mutants of HFV integrase were constructed and analyzed with respect to enzymatic activities. The C-terminal deletion mutant showed decreased enzymatic activities while the N-terminal deletion mutant lost the activities completely, indicating that the N-terminal domain is more important than the C-terminal domain enzymatic reaction. The point mutants, in which an aspartic acid at the 164th position or a glutamic acid at the 200th position of The HFV integrase protein was changed to alanine, lost the enzymatic activities completely. However, they results suggest that the aspartic acid and glutamic acid at the respective 164th and 200th positions are catalytic residues for enzymatic reaction.

Arterioportal (AP) shunt is related to hepatocellular carcinoma (HCC) with variable frequency but its clinical significance is not well known. We retrospectively studied the prevalence and clinical significance including mortality of the AP shunt combined with HCC. Methods : The clinical data and radiologic features of HCC patients who were performed hepatic angiography from 1992 to 1997 at St. Mary's Hospital in Korea were reviewed. The data of HCC patients with AP shunt were compared with that of randomized samples of HCC patients without AP shunt. Results : The prevalence of AP shunt in HCC was 7.3%(45/616 HCC patients). There was no significant difference in clinical symptoms and signs such as ascites, encephalopathy, or variceal bleeding and laboratory findings between the HCC patients with AP shunt and those without. The AP shunt was more common in diffuse, poorly demarcated, large HCC. Especially, portal vein thrombosis (PVT) was one of the most common causes of AP shunt. Cumulative survival rate of the HCC patients with AP shunt was lower than that of those without. But only the size of HCC was significantly related with poor prognosis. Conclusion : AP shunt occurred in some HCC which was large in size or combined with PVT. AP shunt did not increase the severity of symptoms and signs, but decreased the survival because of its relation to tumor size.
수삼 증자 시 생성되는 유출액을 이용한 ginsenoside-Rg₃ 강화 효모 제조

홍삼의 유효성분인 ginsenoside-Rg₃가 강화된 식용 효모를 제조하기 위하여 수삼 증자 유출액을 이용하여 4종의 식용효모를 배양하였다. 효모의 생육도는 Saccharomyces cerevisiae가 가장 높았으며, YEPD 배지에 배양한 경우에 비하여 약 75%의 생육도를 나타내었다. YEPD 배지에 배양한 Saccharomyces cerevisiae에는 ginsenoside가 함유되어있지 않았으나 수삼 증자 유출액에 배양한 Saccharomyces cerevisiae에는 ginsenoside-Rg₃가 0.033 mg 함유되어 있는 것으로 나타났다. 4종의 효모 중 Saccharomyces cerevisiae가 향긋하고 구수한 향취를 나타내어 기호도가 가장 우수하였다. 이러한 결과로 보아 수삼 증자 유출액에 식용 효모를 배양하면 ginsenoside-Rg₃가 강화된 효모를 제조할 수 있으며, Saccharomyces cerevisiae가 생육율과 ginsenoside-Rg₃ 함량이 높고 향미의 기호도가 우수하여 식용 효모로 활용하기에 가장 적합하였다. To produce ginsenoside-Rg3 enriched edible yeast, ginseng steaming effluent (GSE) was used for yeast cultivation in this study. Four kinds of edible yeasts were cultured in sterilized GSE (2% w/v, pH 6.5), without any nutrient, for 48 h at 30℃, and their growth and ginsenoside compositions were determined. Among the yeasts, Saccharomyces cerevisiae showed the highest growth in the GSE medium. 267.1 ㎎ of Saccharomyces cerevisiae biomass was produced from 1 g of GSE solid and ginsenoside-Rg₃ contents was determined with 0.033 mg. Saccharomyces cerevisiae also showed the best overall acceptability, with a herbal and fermentative flavor and a slightly bitter taste. From these data, we conclude that Saccharomyces cerevisiae is the excellent strain for production of ginsenoside-Rg₃ enriched edible yeast using GSE.

Small hepatocellular carcinoma (HCC) can be treated by surgery, transarterial chemoembolization (TACE), percutaneous ethanol injection (PEI) and percutaneous microwave coagulation therapy (PMCT). However, surgical resection is not a viable option for all patients due to poor liver function. Furthermore, TACE is sometimes ineffective because of inadequate angiogenesis in small HCC. PEI does not always bring complete necrosis of the tumor because of inhomogeneous distribution within the tumor. PMCT heats tissue by molecular vibration of water in tissue, and induces thermal coagulation. Regardless of the presence of a capsule, PMCT is the most reliable method in inducing tissue coagulation. Slight pain, mild fever, transient elevation of liver enzyme, subcutaneous hematoma, ascites, pleural effusion and tumor seeding can be developed after PMCT. We report a case of cutaneobiliary fistula complicated after PMCT with a review of literatures.
