효모에서 SHC1 유전자의 이온 농도 조절에 의한 세포내 pH 항상성 유지

출아효모는 주변 환경 pH의 커다란 변화 속에서 적응할 수 있는 효과적인 체계를 지니고 있으며 SHC1 유전자는 알칼리 pH 조건에서 세포의 성장에 필요한 유전자 중에 하나임을 확인하였다. SHC1 유전자의 세포내 pH 조절 기작을 보다 구체적으로 알아보기 위하여 이 유전자가 소실된 돌연변이주를 제조하였다. 성장률의 차이가 나타나는 원인을 세포 내부의 pH 완충능력 결여에 의한 것으로 추측하고 pH 감수성 형광물질인 C.SNARE를 사용하여 외부 pH의 변화에 따른 세포 내부의 pH를 측정하였다. 알칼리 pH 완충효과는 소실 돌연변이의 경우는 야생종 대비 70% 수준을 보였다. 또한 pH 조절에 관여하는 효모세포 내부의 $Na^{+}$과 $K^{+}$의 농도를 원자흡광계를 사용하여 조사한 바, $K^{+}$ 이온의 경우에는 돌연변이주에 비하여 야생형 세포내에 더 많이 존재하는 것으로 나타났으나 $Na^{+}$ 이온의 경우는 별다른 차이점을 보이지 않았다. 이러한 결과는 $K^{+}$ 이온의 조절이 효모에서 세포내 pH조절 기작에 중요하며 SHC1 유전자는 이 $K^{+}$ 이온의 세포내 농도 유지에 관여하고 있다는 것을 제시해 주었다. Budding yeasts maintain an effective system to regulate intracellular pH in response to environmental pH fluctuation. In a previous study we reported that SHC1 plays a role in cell growth at alkaline condition, not at acid pH. We constructed a null mutant deleted an entire open reading frame for SHC1. To test whether the retardation in cell growth was caused by the absence of intracellular pH buffering capacity, we measured intracellular pH with a pH-sensitive fluorescent dye, C.SNARE. The intracellular pH of the mutant cell was much higher than that of wild-type cells, indicating that the mutant cells lack some types of buffering capacity. We also investigated the level of $Na^+ and K^+$ content with atomic mass spectroscopy after alkali shock. Wild-type cell showed a higher level of intracellular K^+$ content, whereas there was no difference in $Na^+$ level. The result suggested that K^+$ is more important in the regulation of intracellular pH in yeasts.

이 글은 이규보의 서사시 「동명왕편」의 ‘창작동기’를 당시의 역사적 사실에서 착안하여 ‘민족’이란 시각 대신에 작가 개인에서 찾는 데서 비롯하였다. 당시는 다른 나라와의 특별한 전쟁이 없었기 때문이다. 따라서 이규보가 주로 20대 후반에서 30대 초반까지 집중적으로 쓴 求官詩와 같은 맥락에서 그 창작동기를 파악하였다. 이러한 ‘求官的 性格’은 무신집권기라는 당시의 시대적 상황과 맞물려서 작품의 성격에 일정 수준의 영향을 미친 것으로 보인다. 「동명왕편」은 그 序文에서 밝힌 인식의 전환을 통하여 고구려의 건국과정에서 벌어지는 奇異한 異蹟들을 모두 神聖한 것으로 수용하였다. 또한 「동명왕편」은 해모수, 주몽, 송양, 유리 등 무인들의 모습과 그들의 대결구도 속에서 전개되는 서사가 구체적으로 형상화되었다. 이러한 서사의 특징을 ‘武人形象’이라고 하였고, 이것의 문학적 형상화는 ‘神異’한 성격을 갖는다고 할 수 있다. 동명왕과 관련된 고려시대 다른 자료들인 『삼국사기』, 『삼국유사』, 『제왕운기』 등에 비하여 이규보의 「동명왕편」은 ‘무인형상의 神異性’이 작가의 윤색에 의하여 더욱 선명하게 드러나 있다. 따라서 「동명왕편」은 고려중기 서사에서 ‘문학적 형상화’란 측면에서 분명한 선편으로 자리하고 있다고 볼 수 있다. 이러한 고려시대 자료들의 ‘신이성’은 조선시대와도 다른 것으로 앞으로 더욱 규명해야 할 중요한 과제로 생각된다. This study attempts to find the motive of creating the Epic of King Dong-myeong(東明王篇) from its author, Lee Kyu-Bo's personal background, beyond the standpoint of a national hero, based on the historical fact there was no war with its neighbors when this epic poetry was composed. Therefore, this article approaches the creation motive in the Epic of King Dong-myeong from the same viewpoint regarding Lee's other poems for his government post(求官詩), mainly made in his late 20s to early 30s. It seems that such character of demanding government service had an effect on the work of Lee under the military regime of Koryo dynasty. As seen in the preface, Lee Kyu-Bo adopted strange miracles in the process of establishing Koguryeo as sacred events in the lay of King Dong-myeong through the change of his recognition. In this epic, both warrior figures(e.g. Haemosu 해모수, Jumong 주몽, Songyang 송양, Yuri 유리 etc.) and their confrontation structure are configured concretely. Warrior figures' mythical and strange character in this epic poetry is more clearly appeared than in other works regarding King Dong-myeong such as Samkuksagi三國史記, Samkukyusa三國遺事 and Jaewangunki帝王韻紀, through Lee's embellishment. As a result, the Epic of King Dong-myeong could be brought to the fore among works representing warriors in the middle of the Koryo. Warrior figures' mythical and strange character in the Koryo period is distinguished from that of the Choseon, and this theme deserves to be explored continuously.
