      • Nuclear Thiol Peroxidase 단백질 결정조건의 최적화

        Yeast nucleus-localized thiol peroxidase(nTPx) is a functional peroxidase with two cysteine residues. For the structural analysis of reduced conformation of this protein, two cysteine residues are mutated by serine. The nTPx-mutant protein is crystallized by sparse matrix crystal screening using hanging drop method. Initial crystal has a rare hairy looking thin needle-like morphology. A series of optimization process was undertaken by a systematic approach. Traditional homology modeling technique is used for the prediction of nTPx-mutant protein. The predicted structure showed the typical thioredoxin fold with the topology which is similar with AhpC peroxiredoxin family. Compared with predicted nTPx structure, this enzyme system should have a typical 2-Cys redox mechanism.

        두개강내 원발성 악성 흑색종 : 증례보고 Case Report

        Intracranial malignant melanomas are commonly secondarily metastatic from the other primary lesion but intracranial primary malignant melanomas are very rare in its incidence, and its clinical course and prognosis are very often different from the metastatic (secondary)lesion. The 34 years old female was admitted to the our hospital because of headache. vomiting and visual disturbance for about 1 month. Neurological examination revealed only papilledema. Brain CT demonstrated the heterogenous high density lesion with peritumoral low density in the left frontal lobe which was enhanced homogenously by contrast media. The images of their CT was mimicking the meningioma. We performed the craniotomy for total removal of tumor. The pathological findings revealed a malignant melanoma. We could not find the other melanomas in skin. eyes. GI system, lung etc and she was survived for 1 year and 9 months now. Following review of literatures we report the rare intracranial primary malignant melanoma.

      • 3세대 뉴클레오사이드 유도체인 4’-Selenonucleoside 들의 결정구조 비교를 통한 삼차원 구조적 특징 분석

        4’-selenonucleosides are third-generation of nucleosides which has been developed for anti-viral agent. So far seven 4’-selenonucleoside's structures are determined by x-ray crystallography. Seven crystal structures are compared and characterized for the detailed furanose ring structure and environment. Most 4’-seleno nucleosides shows south conformation on the furanose ring puckering because of the electron-rich selenium atom. However N7 regio-isomer of selenoadenosine which is substituted by chlorine at N6 position, has a north conformation due to the steric effect of bulky chlorine atom. Also the x-ray crystal structure of 4’-selenoadenosine revealed an unusual mixture of north and south conformations by DNA-like base paring and base stacking. It is expected that this structural investigation of 4’-selenonucleosides is useful for the design of therapeutically effective antiviral agents.

      • 여성관련 입법성과와 의회리더십 : 민법중개정법률안 결정과정을 중심으로

        The purpose of this study is to analyze the parliamentary leadership and legislative outcome related to womens' area in the 16th and 17th National Assembly in Korea. To understand the legislation process, comparative case study was employed as a tool. The cases were nine revised bills among civil law which were related to womens' areas. The results of this study were as follws: The cases of the 16th National Assembly failed to be made as laws, on the other hand, the cases of the 17th National Assembly succeeded. The most important reasons for the legislative success were caused by the strong will of MOPs and the parliamentary leadership to achieve the legislation. Above all, this study found that the parliamentary leadership, over-party solidarity, and enlarging the number of the female MOPs affected most the legislation process like bill-proposal and bill-review and legisltaive outcome. 본 연구는 여성의원들이 상대적으로 증가한 16대 국회와 17대 국회에서 민법중개정법률안 중 여성과 관련된 내용을 담고 있는 입법사례를 중심으로 분석해 봄으로써 여성관련 입법성과와 의회리더십을 살펴보았다. 분석 대상은 동성동본금혼 폐지를 다룬 3건의 법률안(의원발의안 2건, 정부제출안 1건)과 호주제 폐지를 다룬 6건의 법률안(의원발의안 3건, 정부제출안 2건, 위원회안 1건)이고, 분석 방법은 회의록 내용을 중심으로 한 사례비교분석을 시도하였다. 분석 결과, 이해관계가 분명하게 대립되는 쟁점이었던 여성관련 호주제폐지 법안이, 16대 국회와는 달리 17대 국회에서 실질적인 입법성과를 이룰 수 있었던 점은 사회적 공감대가 확산되어 시대적 대세로 형성되었다는 점과 정부에서 지지하였다는 점 등이 영향 요인으로 작용하였지만 무엇보다도 입법주체인 의원들의 입법을 이루려는 적극적 의지와 의회리더십에 바탕했다고 볼 수 있다. 특히 여성의원들의 수적 증가와 활발한 초당적 공조 활동 등, 여성의원의 의회리더십이 여성관련 법률안 발의와 입법에 큰 영향을 미쳤다는 점이 확인되었다.

      • 학습 기술 개입에 대한 개관

        본 논문에서는 학생들의 학습능력 향상을 목표로 하는 학습 기술 개입에 관한 연구들을 개관하였다 본 논문의 목적은 첫째, 학습 기술 개입의 정의, 기본원리, 모델, 종류 및 학습기술평가 도구에 관해 소개하고, 둘째, 선행 연구 결과에 기초하여 효과적인 학습기술 개업의 특성을 살펴보며, 셋째, 교육 및 임상적 활용을 위한 합의점과 향후 연구 방향에 대해 제시하고, 넷째, 실전 활용을 위한 학습 기술 개입 프로그램의 한 예를 제시해 주고자 하는 것이다 이와 같은 목적을 위해 학습 기술과 관련된 일련의 국내외 저서, 학술 논문 및 연구 결과들이 조사되었다. 학습 기술 관련 선행연구 결과에 따르변, 대략 1980년대부터 1990년대 사이에 학습 기술 개입에서 학습 기술, 동기, 자기 개념, 자기 효능감, 자기 조절, 전이 및 개업의 맥락과 같은 요인들이 연구의 쟁점이 되었다. 일련의 연구를 통해 성공적인 학습 기술개입의 특성에 대한 몇가지 동의가 이루어졌지만, 학습 기술 개업의 효과의 크기 면에서 여전히 다양한 결과를 보여 주고 있다. 즉, 개입의 종류에 따라 성과에 차이가 있고, 학습 기술 개입이 수행보다는 정서적 변인에 더 많은 영향을 미친다는 결과가 있었으며, 개입 성공의 일반화의 문제점 등이 보고되었다. 이와 관련하여 동기, 의지, 자존감과 같은 정서적 변인에 대한 강조가 이루어졌다 본 개관에서 살펴본 연구 주제, 관심 및 쟁점 등은 향후 연구 방향의 지침으로 참고될 수 있을 것이다. 국내의 경우, 사실상 학습 기술 개업과 관련된 연구가 거의 이루어지지 않았음을 감안해 볼 때, 앞으로의 연구에서는 학습 모델에서 강조되고 있는 초인지에 대한 검증으로부터 시작하여, 학습 기술 개입의 특성, 구성 요소 및 평가 등에 대한 기초적인 연구가 이루어져야 할 것이다 더 나아가 이러한 연구 결과들이 국내 교육 환경에 맞는 고유한 학습기술 개입 프로그램의 개발로 이어져야 할 것이며, 그에 따른 지속적인 타탕성 검증이 요구된다고 하겠다.

      • Homology modeling에 의한 항산화 단백질 Bacterioferritin Comigratory Protein (BCP)의 구조 분석

        Escherichia coli bacterioferritin comigratory protein (BCP) is a putative bacterial member of the thiol-specific antioxidant protein family that are widely expressed in various pathogenic bacteria and show distant yet significant sequence homology with peroxiredoxins. Two of the protein structure of this family has been solved by the X-ray crystallography. One of that is Escherichia coli p20 and other is alkyl hydroperoxide peroxidase C (AhpC). The three-dimensional structure of BCP is predicted by homology modeling using these two crystal structures. The predicted structure revealed a same peroxiredoxin architecture with minor differences in loop structures. Based on the predicted structure, it is hypothesized that BCP is involved in atypical 2-cys type mechanism using unique intra helical disulfide formation.

      • 지렁이에서 추출한 Lumbrokinase의 X-선 회절 예비실험 및 homology modelling

        The earthworm has been used as a fibrinolytic agent in East Asia for several thousand years. Fibrinolytic activities by the earthworm have been tested since 1983 and proteolytic enzyme, named Lumbrokinase, has been extracted and purified from Lumbricus rubellus. Lumbrokinase shows an ability of direct degradation of fibrin to minimize side effect, and it shows greater stability compared with other fibrinolytic agents. Recently, it is immobilized in a Korean type total artificial heart polyurethane valve, and it lead to decreased thrombus formation and better biocompatibility. We have investigated the three dimensional structure of this enzyme by the X-ray crystallography method. The microcrystals of Lumbrokinase Ⅲ-2 has been obtained in the condition of 18% PEG8000, 0.1M Na cacodylate buffer(pH6.5), and 0.2M magnesium acetate. The preliminary X-ray diffraction experiment shows that this crystal has a space group of P21 and three molecules in asymmetric unit. For the further structural analysis, the homology model of Lumbrokinase has been built from the structure of porcine beta trypsin. It is expected that the crystal structure of Lumbrokinase can be obtained in a short period by using molecular-replacement method and the high resolution three-dimensional stucture of Lumbrokinase will provide an crucial information about the protein engineering and new fibrinolytic agent development.

        임플랜트의 표면처리 방법이 골유착에 미치는 영향에 관한 연구

        The purpose of this study was to compare the effects of various surface treatments by measuring removal torque on bone healing around titanium implants. 40 Screw-shaped cp titanium implants with length of 4mm, outer diameter of 3.75mm, and pitch-height of 0.5mm were used Group 1 was left as machined(control), Group 2 was blasted with $50{\mu}m\;Al_2O_3$, group 3 was blasted and etched in etching solution($NH_4OH : H_2O_2:H_2O= 1 : 1 : 5$) at $90^{\circ}C$ for 1 minute group 4 was blasted and oxidated under pure oxygen at $800^{\circ}C$. The implant surface roughness was analyzed with SEM and CLSM(Confocal Laser Scanning Microscope) and implants were placed in proximal tibial metaphysis of 10 New Zealand White rabbits. After 3 months of healing period, removal torque of each implant was measured to compare bone healing around implant. The results obtained were as follows 1. In SEM view, blasting increased the roughness of the surface, but etching of that rough surface decreased the roughness due to the removal of the tip of the peak. Oxidation also decreased the roughness due to formation of needle-like oxide grains on the implant surface. 2. The Sa value from CLSM was least in the machined group($0.47{\mu}m$), greatest in blasted group($1.25{\mu}m$), and the value decreased after etching($0.91{\mu}m$) and oxidation($0.94{\mu}m$). 3. The removal torque of etched group(24.5Ncm) was greater than that of machined group(16.7Ncm) (P<0.05), and was greatest in the oxidated group(40.3Ncm) and the blasted group(34.7Ncm).

      • 분자모델링에 의한 Nuclear Thiol Peroxidase의 삼차원 구조예측

        Yeast nucleus-localized thiol peroxidase(nTPx) is a functional peroxidase with two cysteine residues. This enzyme is homologous with Ahpc on 180 residues at the C terminal. while 60 residues at the N terminal has no homology with any protein. Traditional homology modeling technique is used for the prediction of nTPx protein. The predicted structure showed the typical thioredoxin fold with the topology which is similar with AhpC peroxiredoxin family. Two cysteine residues can not be close for the disulfide formation in this structure, which indicates that nTPx should have a typical 2-Cys redox mechanism. Possible homodimer structure also could be predicted and these interactions can be a part of large assemblies such as decamer.
