Residues of ampicillin and amoxicillin in the muscles of olive flounder Paralichthys olivaceus were investigated after oral doses of 20 mg/kg body weight/day of ampicillin and 40 mg/kg body weight/day of amoxicillin in a first examination and 40 mg/kg body weight/day of ampicillin and 80 mg/kg body weight/day of amoxicillin in a second examination for 5-6 days. The ranges of seawater temperatures in the 1^(st) and 2^(nd) examinations were 13.4-15.6℃and 16.8-21.4℃, respectively. Ampicillin and amoxicillin concentrations were analyzed by high performance liquid chromatography (HPLC). The average recovery rates of ampicillin and amoxicillin in fish samples ranged from 84.8-95.2% and 100.8-103.8%, respectively. Residual concentrations in the olive flounder peaked on the 5th day, with average concentrations of 0.075 mg/kg and 0.311 mg/kg in the 1st examination, and 0.098 mg/kg and 0.630 mg/kg in the 2^(nd) examination for ampicillin and amoxicillin, respectively, with maximum concentrations in muscle. In the 1^(st) examination, ampicillin concentrations in olive flounder after withdrawals of 1 and 3 days were 0.041 mg/kg and 0.023 mg/kg, respectively, and amoxicillin concentrations after withdrawals of 1 day and 10 days were 0.172 mg/kg and 0.023 mg/kg, respectively. In the 2^(nd) examination, the ampicillin concentrations in olive flounder after withdrawals of 1 and 3 days were 0.041 mg/kg and 0.023 mg/kg, respectively, and amoxicillin concentration after withdrawals of 1 day and 10 days were 0.172 mg/kg and 0.023 mg/kg, respectively. We suggest that the recommended withdrawal periods should be 3 days for ampicillin and 10 days for amoxicillin in the olive flounder.

A lateral flow immunoassay kit based on antigen-antibody interactions was developed to detect residues of beta-lactams, quinolones, tetracyclines, and sulfonamides in farmed fish. Group-specific antibodies showing cross-reactivity with other antibiotics in the same group were produced in rabbits. The rabbits were immunized eight times to obtain the maximum titers. Antibodies were extracted from the antisera collected from the immunized rabbits and produced group-specific reactions with antibiotics from the four groups. A kit was prepared that optimize conditions for the antigen-antibody reaction, using colloidal gold conjugated antibodies, and was designed to detect the four groups of antibiotics simultaneously. The kit enabled the detection of antibiotics in the four groups at below maximum residue limits (MRLs), which were 200 µg/kg for tetracyclines, 100 µg/kg for sulfonamides, 50 µg/kg for beta-lactams, and 100 µg/kg for quinolones. The cross-reactivity of the antibodies ranged from 10-80% for the sulfonamides, 20100% for tetracyclines, 38-100% for quinolones, and 20-100% for the beta-lactams, confirming that the antibodies were group specific. The test kit was used 30 times to examine spiked antibiotics at the limits of detection (LODs) and all produced positive results, indicating high sensitivity. The LODs for the assay ranged from 4-20 ng/mL for beta-lactams, 25-50 ng/mL for sulfonamides, 20-100 ng/mL for tetracyclines, and 30-80 ng/mL for quinolones, and there were no false negative reactions at above these LODs. In addition, all of the LODs of the developed kit were correlated with high-performance liquid chromatography (HPLC) data. Our lateral flow immunoassay kit can simultaneously detect antibiotic residues from a large number of fish samples rapidly, strengthening the safety of domestic farmed and imported fish.

This study investigated the concentration of total mercury in 17 species of commonly consumed fishes and assessed the risk to human health from provisional tolerable weekly intakes PTWI% as affected by migratory characterization, such as migration and vagility. The measured mean concentrations of total mercury in these 17 species of commonly consumed fishes suggest that mean concentrations of total mercury in 10 species of migratory fishes (largehead hairtail, chub mackerel, Pacific saury, skipjack tuna, Pacific cod, anchovy, Alaska pollack, brown croaker, Japanese Spanish mackerel, yellow croaker and Pacific herring) were low compared to those in 7 species of demersal fishes (red stingray, brown sole, bastard halibut, conger eel, blackmouth angler, rockfish and filefish). Based on the mean concentrations, the PTWI% of total mercury among commonly consumed migratory fishes was 3.393%, which was higher than that of commonly consumed sedentary fishes (2.710%).
Total Mercury Contents of Antarctic Toothfish Dissostichus mawsoni Caught in the Antarctic Sea

Mercury is an element of special concern for human health. Measurements of total mercury levels in fish have been taken into consideration to assess risk. In this study, the Antarctic toothfish Dissostichus mawsoni was evaluated as a potential safe food source through measurement of total mercury contents. Total mercury concentrations in Antarctic toothfish ranged from 0.101 ± 0.047 mg/kg to 0.139 ± 0.075 mg/kg. The total mercury concentration was significantly correlated with macroscopic values including total fish length, weight, gonadosomatic index, and maturity (P < 0.01 or 0.05). Furthermore, according to the risk assessment, the total mercury body exposure rate from Antarctic toothfish ranged from 2.125% to 2.847% of the Provisional Tolerable Weekly Intake. Therefore, the Antarctic toothfish could be used as a potential safe seafood source.
유통 마른 김(Porphyra sp.)의 중금속(Hg, Pb, Cd) 함량과 위해성 평가

To investigate heavy metals (Hg, Pb and Cd) and their potential health risks in commercial dried laver (Porphyra sp.), we collected 45 samples from the major production areas on the western and southern coasts of Korea (Hwaseong, Seocheon, Gunsan, Muan, Shinan, Jindo, Haenam, Wando, Jangheung, Goheung and Busan). The Hg, Pb and Cd concentrations were measured using inductively coupled plasma spectrometry (ICP-MS) or a mercury analyzer. The average Hg, Pb and Cd concentrations in the dried laver were 0.006±0.0017, 0.196±0.0614 and 0.894±0.4882 mg/kg, respectively. Based on the 2007 Korean Public Nutrition Report, these levels are 0.02, 0.11 and 2.47% of the provisional tolerable weekly intake (PTWI) for Hg, Pb and Cd, respectively, established by the FAO/WHO. The hazard quotient (HQ) determined from the ratio of exposure and safe levels were less than 1.0. Therefore, the levels of overall exposure to Hg, Pb and Cd for dried laver were below the recommended JECFA (Joint FAO/WHO Expert Committee on Food Additives) levels, which indicate safe levels for public health.

This study evaluated the effect of the inland pollution sources on the bacteriological water quality in Narodo area after rainfall events. Following 16 mm of rainfall, the water discharged from Dohwa Stream and contaminants from Balpo village had a very limited effect on bacteriological water quality in adjacent area and the affected area did not extend to the designated area. In comparion, after 33 mm of rainfall, the density of fecal coliforms at stations located in the mouth of Dohwa stream and the discharg point of contamination sources located in Balpo village were lower than after 16 mm of rainfall. These results were obtained during an extreme high tide near the coast the contaminants discharged from Dohwa stream and Balpo village. Therefore, it is necessary to conduct a detailed survey to examine the dffects of tides on the diffusion characteristics of the contaminants discharged from Dohwa stream and Balpo village in order to manage of Narodo area efficiently.

The sanitary quality of a shellfish-growing area in Gangjinman, Korea, was assessed through a bacteriological examination of seawater form January 2007 to December 2009. The range of the geometric mean (GM) and the estimated 90th percentile for most-probable-number (MPN) values for fecal coliforms in samples collected in the designated area were 2.0-2.4 and 6.1-11.5 MPN/100 mL, respectively. Therefore, the bacteriological quality of seawater in the designated area in the Gangjinman area met the National Shellfish Sanitation Program (NSSP, USA) guidelines for approved areas. Also, the sanitary status of shellfish harvested from the designated area met the Korean Shellfish Sanitation Program (KSSP) fecal coliform criterion (<230 MPN/100 g). However, the degree of bacteriological contamination increased rapidly, and water quality exceeded the criteria for approved areas when large amounts of rain fell before the sampling date. Fortunately, fecal pollutants did not affect the bacteriological quality of a buffer zone established between the shoreline and the designated area. These results indicate that the current buffer zone established around the designated area protects the designated area properly.
연안 어시장에서 유통되는 별복(Arothron firmamentum)과 밀복류(Lagocephalus spp.)의 독성

The toxicity of five species of puffer fish, Arothron firmamentum (Byeolbok), Lagocephalus gloveri (Heukmilbok), L. wheeleri (Eunmilbok), L. inermis (Minmilbok) and L. lunaris (Milbok), collected from fish markets in Korea, was determined using a mouse bioassay. In A. firmamentum, the proportion of toxic specimens containing >10MU/g was 87.5% in the ovaries, and 10.0% in the skin; no toxicity was detected in the muscle, fin, liver, intestine and gallbladder using the mouse bioassay. The highest toxin levels were found to be 87MU/g in the ovaries, and 13MU/g in the skin. Toxic specimens containing >10MU/g were not detected from samples taken from any of the organs in L. wheeleri and L. inermis. In L. gloveri, most specimens were found to be non-toxic, but toxin levels of 11-72MU/g were detected from within the skin, fins, and intestines in one specimen. In L. lunaris, the proportion of toxic specimens was 50.0% in the ovaries, and 7.1% in the gallbladder; no toxicity was detected in the other organs by the mouse bioassay. The highest toxin levels were 75MU/g in the ovaries, and 14MU/g in the gallbladder. Therefore, the toxicities of edible muscle and skin in the five species of puffer fish marketed in Korea were found to be within acceptable levels for human consumption.

We investigated the effect of the discharged water from Bong Stream, which is located in the drainage area of Gangjinman area on the bacteriological water quality in the coastal area after rainfall events. Following 12.5 mm of rainfall, water discharged from Bong Stream had a very limited effect on bacteriological water quality in adjacent area and the affected area did not extend to the designated area. On the other hand, after 23 mm rainfall, the density of fecal coliform at stations located in the designated area was higher than at stations located in the adjacent area. The degree of bacteriological contamination at the surveyed stations in the coastal area after rainfall events did not show a relationship with distance from the shoreline. These results indicate that the direction of spread and the range of contaminants from the drainage area were affected by tides at the time of the survey. Therefore, a detailed survey of the effects of tides on the diffusion characteristics of the contaminants from Bong Stream is needed to establish a proper management plan for the surveyed area.
