제주지역 양식넙치의 연쇄구균증 발생동향 및 원인균에 대한 분자적 동정

제주지역 양식넙치의 주요 세균성 질병의 일종인 연쇄구균증 발병동향과 특성을 분석하였다. 제주지역 양식넙치의 연쇄구균증은 연중 발생하는 양상을 보였으나 상대적으로 고수온기에 발생율이 높았으며, 30cm 이상의 개체에서 주로 관찰되었다. 2003년 6월부터 2005년 5월 사이에 분리된 연쇄구균 균주를 multiplex PCR assay 기법을 이용하여 종 조성을 분리한 결과 Streptococcus iniae와 S. parauberis로 동정되었으며 그 비율은 각각 46%와 54%이었다. S. iniae는 상대적으로 9월과 10월에 높은 분리율을 나타내었으나 S. parauberissms 3월과 4월에 분리율이 높게 나타났다. S. iniae와 S. parauberis 감염개체의 병리학적 중상은 일부분 유사하였으나 S. iniae 분리개체는 복부팽만, 탈장, 복수저류증상이 주로 관찰되는 반면, S. parauberis 분리개체는 체색흑화, 무안측 발적 중상이 두드러지게 관찰되었다. 이 두 종의 병원체 모두 ampicillin과 amoxicillin에 감수성이 높았으며, S. iniae 균주들은 S. parauberis 균주들보다 상대적으로 doxycycline, erythromycin, oxytetracycline에 감수성이 높았다. Streptococcosis of olive flounder(Paralichthys olivaceus) is an important bacterial disease in Jeju island. In this study, we investigated monthly infection pattern of this disease in different size of the flounder fish. Even though the disease occurred throughout the year, the infection ratio was relatively higher in the months with warm water season. The infection was more prevalent in adult flounder over 30 cm total length compare to these of small size fish. Two infectious species of streptococcosis pathogens were detected by multiplex PCR assay. Detection ratios of Streptococcus iniae and S. parauberis reached up to 46% and 54%, respectively, from June 2003 to May 2005 in Jeju island. S. iniae occurred intensively from September to October, whereas S. parauberis reported from March to May. S. iniae and S. parauberis infections of cultured flounder share some common features, but clinical findings showed considerable differences between two diseases. Distended abdomen, protruded anus and ascitic fluid in the peritoneal cavity are evident lesions detected in S. iniae infection, whereas, flounders infected by S. parauberis showed prominent lesions such as darkened surface and haemorrhaging in the non-ocular side. Both streptococcosis pathogens were sensitive to antibiotics, such as ampicillin and amoxicillin. However, S. iniae strains were more sensitive to doxycycline, erythromycin and oxytetracycline than S. parauberis strains.

Streptococcus iniae의 유전학적 다양성과 RAPD fingerprint profile의 비교

2004년부터 2005년까지 제주도에서 전형적인 연쇄구균증상을 나타내는 양식넙치로부터 Streptococcus sp.를 분리하였다. 생화학적 성상 조사와 multiplex PCR assay법을 통해 S. iniae 94 균주를 동정하였다. RAPD fingerprint profile을 조사해 본 결과 A, B, C-type 총 3가지의 genotype을 확인하였으며, 동일종에 대해 외국에서 보고된 선행 연구결과와 비교하였을 경우, 지리적으로 다른 유전적 다형현상을 나타내는 것으로 추정되었다. 근래에 제주도에 우점하는 genotype은 A-type이었고, RAPD fingerprint profile은 약 2,000 bp, 1,300 bp, 1,000 bp, 850 bp, 550 bp의 밴드 양상을 나타내었다. S. iniae의 serotype 동정에 유효한 것으로 보고되어진 P14 random primer는 유전적 다형현상에 의해 제주도에서 분리된 S. iniae의 serotype 동정에 적합하지 않은 것으로 판단되었다. Streptococcus sp. were isolated from cultured flounder (Paralichthys olivaceus) having Streptococcosis during 2004 to 2005 in Jeju Island. Ninety four Streptococcus iniae strains were isolated using biochemical test and multiplex PCR assay. Three genotypes (A, B, C-type) of S. iniae were appeared in the RAPD analysis and they showed international or local genetic polymorphism. Presently, S. iniae having A-type is a dominant S. iniae genotype in Jeju and showed band patterns at about 550, 850, 1000, 1300 and 2000 base pares. In this study, the reported P14 random primer, that used to distinguish serotypes of S. iniae could not be applied to distinguish Jeju island S. iniae's genetic polymorphism.

8주간 순환운동 프로그램이 지적장애 아동의 운동수행능력에 미치는 영향

이 연구의 목적은 8주간 규칙적인 순환운동 프로그램이 지적장애 아동의 운동수행능력에 미치는 영향을 파악하는 것이었다. 연구 참여에 동의한 총 20명의 지적장애아동들은 실험집단(10명), 통제집단(10명)에 단순무선표출로 할당하여 배정하였다. 두 집단의 비교를 위해 TGMD-Ⅱ(이동기술 물체조작기술)를 사용하였으며 SPSS Ver 23.0K 통계프로그램을 이용하여 집단별로 평균과 표준편차를 산출하기 위해 독립변인 T-test를 실시하였다. 순환운동 참여를 통해 이동기술에서 갤롭, 슬라이드, 홉에서 집단에 따라 유의한 차이가 나타났으며 물체조작 기술은 모든 요인에서 유의한 차이가 나타나지 않았다. 순환운동 참여를 통한 지적장애 아동들의 운동수행 자료로는 사용이 가능하지만 8주간의 실험기간이 짧고 프로그램 적응 시간이 부족하며 지적장애인 뿐만 아니라 비장애인을 포함하여 다각적인 연구가 이루어져야 할 것으로 사료된다. 또한 순환운동 프로그램은 지적장애 아동의 이동기술의 하위요인 중 갤롭, 슬라이드, 홉에서 운동수행능력에 긍정적인 영향을 미친다는 결론에 도달하였다. The purpose of this study was to investigate the effect of the 8-week regular cycling program on the development of the athletic performance ability of mental retardation students. A total of 20 children with intellectual disabilities were randomly assigned to two groups (i.e., 10 students for each group), using simple random sampling. Independent t-test was conducted to calculate the mean and standard deviation for each group using SPSS 23, and TGMD-II (Test of Gross Motor Development-2) was used to compare the two groups. The results indicate that there were significant differences between the two groups in gallops, slides, and hops through circular motion, but no significant differences were found through object manipulation skills. The results can be used as data of circular motion participation of children with intellectual disabilities. However, since the length of the experiment period and the time of adjustment is short (i.e., 8 weeks) to identify the effects, further experimental investigations will be needed including children with intellectual disabilities and non-intellectual disabilities. It was concluded that the circular motion program had a positive effect on exercise performance, in terms of gallops, slides, and hops.

기술현황분석 - 트랙터용 기계-유압식 무단변속기(DMT) 연구현황

기계-유압식 무단변속기인 DMT는 농촌의 고령화의 추세에 맞물려 중 대형 트랙터의 핵심기술로 부각되고 있다. DMT는 기계동력과 유압동력인 두 개의 전달 구조를 조합하고 서로의 단점을 보완함으로써 기동력이 우수하고, 변속충격이 거의 없고, 전달효율이 향상되고 있다. 트랙터용 DMT는 유압동력을 전달하는 HST, 기계동력을 분배 뽀는 합성하는 유성기어 장치, 전진과 후진을 선택하는 전 후진 클러치, 출력축을 정지하는 브레이크 등으로 구성되어 있다. 본 연구에서는 DMT의 핵심기술인 HST와 유성기어장치에 대한 내부구조 및 동력전달을 해석하고 시제품의 실험결과를 제시하여 국내에서 진행되고 있는 DMT의 연구현황을 소개하고자 한다.
