Methoxy poly(ethyleneglycol) (MPEG)-b-poly(ε-caprolactone) (PCL) diblock copolymer was synthesized by ring-opening of ε-caprolactone (ε-CL) in the presence of HCl · Et2O as a monomer activator with the terminal alcohol of methoxy poly(ethyleneglycol) (MPEG) as an initiator. The temperature sensitive behavior of the prepared MPEG-PCL diblock copolymer was examined in aqueous. The MPEG-PCL diblock copolymer was dissolved in water at 80 oC. The polymer solutions formed translucent sol around at the room temperature. The formed sol became gel at 31 oC as the temperature increased. On further increase of the temperature, the gel changed to sol at 52 oC. This indicates the diblock copolymer solutions at room temperature can form gel at body temperature. Albumin or brain derived neurotrophic factor (BDNF) loaded MPEG-PCL diblock copolymer solution was prepared to observe the release behavior of protein or growth factor. The release profile of albumin or BDNF in MPEG-PCL gel showed the sustained zero-order release for 20 days. In conclusion, we confirmed that MPEG-PCL diblock copolymer solutions is useful as protein or BDNF carriers.

Adipose derived stromal cells (ADSCs) are very similar to MSCs and can differentiate into mesenchymal lineage and nonmesenchymal lineage. ADSCs are very easily obtained than MSCs and no pain to acquire cells. Recently researches revealed that MSCs have nonspecific immune suppressive effect on lymphocyte proliferation. We studied about whether nonself ADSCs have lymphocyte suppressive effect on mixed lymphocyte culture (MLR) like MSCs. First we obtained 1st passaged ADSCs and tested whether these cells have stem cell characteristics. We confirmed induced cell by ALP and Oil red O staining. For immune suppressive effect test, ADSC were co-cultured with reaction lymphocyte and stimulator lymphocyte. We defined nonself ADSCs could suppression lymphocyte proliferation in MLR and two more cell number treatment could suppression lymphocyte proliferation 5-fold. Accordingly we found out ADSCs can lymphocyte suppression by dose-dependent pattern. In nonspecific immune suppression test by PHA as non specific lymphocyte stimulator, ADSCs can suppression PHA induced lymphocyte proliferation. In conclusion, it is our belief that ADSCs cells can alternative sources of MSCs in clinically usage and also remain carefully treatment.
섬유아세포에서의 DNA/폴리에틸렌이민 나노복합체를 이용한 유전자 전달

The aim of this study is to evaluate transfection efficiency in NIH/3T3cell using DNA/polyethyleneimine (PEI) complexes. The formation of complexes from DNA and PEI was performed by the adding of the PEI solution to the DNA solution. Gel retardation assay showed the optimal N/P charge ratio where PEI completely binds DNA. The particle size of DNA/PEI complexes measured by dynamic light scattering showed diameters of up to 500 nm. The viability of NIH/3T3 fibroblast cells in the presence of DNA/PEI complexes was evaluated by the MTT assay. After gene transfection, enhanced green fluorescence protein was monitored by flow cytometry. From AFM measurement, DNA/PEI complexes showed round-shape. These results indicated that DNA was sufficiently condensed by PEI to give DNA/PEI complexes. As the relative amount of PEI was increased, its cytotoxicity to NIH/3T3cells increased. By increasing culture time, fluorescence microscopy showed a more green fluorescence cells. Naked DNA showed no EGFP expression, whereas delivery of DNA/PEI complexes to cells showed EGFP expression and resulted in 2-15% transfection. Transfection efficiencies of DNA/PEI complexes increased with increasing N/P charge ratio.
장애인 건강보건관리사업의 인식도와 중요도 연구: 보건소 지역사회중심재활사업 지원 활성화를 중심으로

연구목적 : 본 연구는 경상남도 지역장애인보건의료센터에서 운영하는 장애인 건강보건관리사업의 인식도와 중요도를 조사하고, 보건소 지역사회중심재활사업(Community Based Rehabilitation, CBR)의 지원 활성화 방안을 모색하고자 하였다. 연구방법 : 경상남도 전체 시⋅군 20개 보건소의 CBR사업 담당자 20명을 대상으로 설문조사를 하였다. 통계분석은 SPSS 21.0 프로그램을 사용하였다. 연구결과 : 경상남도 지역장애인보건의료센터의 장애인 건강보건관리사업에 대한 인식도는 보건소 CBR사업연계 및 지원, 의료서비스 연계 및 지원이 가장 높았고, 중요도는 지역사회기반 보건의료-복지 전달체계 구축, 의료서비스 연계 및 지원이 가장 높았다. 장애인 건강보건관리사업의 인식도와 중요도가 대체로 긍정적으로 나타났지만 아직까지 연계현황은 인적⋅물적 자원의 부재, 기관 간 유기적 연계체계 부족 등의 이유로 미흡한 것으로 나타났다. 경상남도 지역장애인보건의료센터의 보건소 CBR사업 지원 활성화를 위해서는 기관 간 협력을 위한 업무 매뉴얼 개발과 관련기관이나 조직 등 공식 적인 연계망 형성이 필요한 것으로 나타났다. 결론 : 경상남도 지역장애인보건의료센터의 장애인 건강보건관리사업 일환으로 기관의 역할을 정비하고, 기관 간 연계 활성화에 대한 근거를 처음으로 제시하였다. Objective : This study investigates the awareness and importance of the healthcare program operated by the Gyeongnam Regional Health & Medical Center for Persons with Disabilities(GDC), and explores options for effectively supporting Community Based Rehabilitation(CBR) in public health centers. Methods : With a questionnaire, we surveyed 20 participants who managed the CBR in Gyeongnam’s public health centers. All analyses were performed using SPSS 21.0. Results : The awareness of the healthcare program in GDC was high in theses areas: linkage and support of CBR, linkage and support medical services. The importance were perceived as high in these areas: development of community based healthcare-welfare delivery system, linkage and support medical services. Although the awareness and importance of GDC was positive, there was not enough linkage status due to the absence of human⋅material resources, the lack of linkage system among institutions. For the GDC to support the CBR more effectively, formal linkage systems for cooperation such as work manuals and organizational networks are needed. Conclusion : This study provides evidence for the first time to support defining roles clearly and increasing the linkage status among institutions of healthcare program in GDC.
