        낙동강 하구 생태계 식물성 플랑크톤의 일차생산성

        Primary production of phrtoplankton was investigated in Naktong estuarine ecosystem monthly from October 1991 to September 1992 using carbon-14 uptake method. In spite of large spatial and temporal variation, there was a primary peak of net phytoplankton production in spring through early summer. The productivity was minimum of 543mg C·m^-2.day^-1 in September and maximum of 4112mg C·m^-2·day^-1 in April. The seasonal changes in factors regulating production in Naktong estuarine area were proposed based on the annual patterns of physical variables and nutrient concentrations. The late spring and early summer flowering of biomass and net production of phytoplankton was explained as the result of the balance between light and temperature. The high level of Production terminated in summer as the result of water clarity. Nutrient limitation may not be important, because of high nutrient level year around in this eutrophicated estuarine area.

        洛東江 하류 수계의 毒性 Microcystis 生態 연구

        The standing crop of genus Microcystis, microcystin concentrations and environmental factors were monitored at stations of the lower reaches of the Nakdong River in 1998 and 1999 during the periods of its occurrence. The Microcystis were observed from May to Octorber, and the cell density was highest at Station Seonam up to 250,000 cells${\cdot}ml^{-1}$ forming scum over the water surface. There were signigicant relationships between the standing crop of Microcystis and nitrate nitrogen, total phosphorus concentrations and Ph. Presumably these parameters were important in the succession to Microcystis dominated phytoplankton community in the summer period in the river. However, Ammonium nitrogen, phosphate phosphorus concentrations and N/P ratio were not critical factors. The Microcystis bloom was notable above $25^{\circ}C$ of surface water temperature. Microcystins were detected from May to November in the algal materials from the river. The 84.2% of algal materials with Microcystis exhibited the microcystin with the maximum of 1711.8 ${\mu}g{\cdot}g^{-1}$ dw. The microcystin concentrations in the algal materials were significantly related to the stading crop of Microcystis, which was the primary determinant factor in the toxin levle of algal materials. The concentrations were also significantly related to pH of the water column in the positive pattern.

        도시근교 하천 조만강의 (潮滿江) 식물플랑크톤 일생산성 추정

        Hourly photosynthetic P-I curves were used to estimate the daily production from in situ C-14 data. P^B_(max) were ranged from 1.4 to 7.5(㎍ C/㎍ chl-a/hr) and α ranged from 3.6 to 14.8[㎍ C/㎍ chl-a/(mol/m^2/hr)]. P^B_(max) and α were simultaneously high in the growing season and low in winter. α and I_k were good indicators for seasonally low-irradiance adaptation in algal photosynthesis. At surface high irradiance, photoinhibition didn`t occur and algal assimilation showed a linear or logarithmic decrease along the depth. As water transparency dropped, primary productivity per unit area didn`t increase as production per unit volume did at water surface. Daily productions were estimated from 1.5(g C/m^2/day) at January to 4.0 (g C/m^2/day) at July. The photoadaptation and lack of photoinhibition were reflected by the role of irradiance and water temperature as most significant predictors of daily algal production. Carbon contents of phytoplankton comprised of 33% of particulate organic carbon of water, turn-over rate (P/B) of phytoplankton was 0.61/day and turn-over time was 1.6 day. Choman River, a stagnant and fertilized river near Kimhae city, is characterized by high nutrient loading from the surrounding area and high autochthonous production.

        분자 모니터링을 이용한 서낙동강과 남해 연안 플랑크톤 군집 분석

        플랑크톤의 종다양성은 특정 지역의 수계환경 변화 모니터링에 있어 중요 생태지표로써, 환경 평가에 유용한 정보로 사용되고 있다. 기존의 종다양성 평가는 주로 형태학적 형질에 근거한 종동정을 통해 이루어졌으나, 많은 시간과 전문성을 필요로 하고 연구자의 주관적 판단에 의존하는 단점이 있다. 따라서, 본 연구에서는 채수된 환경시료에 대해 보다 빠르고 정확한 플랑크톤 종다양성을 파악하기 위하여 분자마커를 활용한 분자모니터링 기법을 도입하였다. 서낙동강(김해교)과 남해 연얀(남해도) 정점에서 각각 채수된 환경시료에서 DNA를 추출한 후 18S nuclear ribosomal RNA 유전자를 대상으로 중합효소연쇄반응을 수행하였다. 클로닝 과정을 통해 만들어진 각각의 클론 라이브러리에서 클론을 무작위로 선택하여 제한효소절편다형성 패턴분석을 한 후 특이성을 가지는 클론을 선별하였다. 김해교에서는 60개 블론을 대상으로 44개의 특이적 클론을 선별하였고 남해에서는 150개 클론을 대상으토 27개의 클론을 선별하였다. 이틀 클론틀에 대한 염기서열 분석결과 다양한 계통분류군에 속승하는 플랑크톤의 종조성 결과를 보여주었다(김해교: Heterokontophyta(7), Ciliophora(23), Dinophyta(l), Chytridiomycota(l), Rotifera(I), Arthropoda (11), 남해: Ciliophora( 4), Dinophyta(3), Crγptophyta(l),Arthropoda(19)). 본 연구를 통하여 분자마커를 활용한 분자모니터링 기법이 기존 형태학적 형질에 근거한 분석이 가지는 한계를 보완하여 채수된 환경시료의 종조성 분석에 효율적으로 사용될 수 있다고 판단된다. The biodiversity of eukaryotic plankton has commonly been used to evaluate the status of aquatic ecosystems. Therefore, an accurate and rapid method for species identification is needed to reveal the biodiversity of environmental water samples. To date, molecular methods have provided a great deal of information that has enabled identification of the hidden biodiversity in environmental samples. In this study, we utilized environmental polymerase chain reaction (PCR) and constructed the 18S nuclear ribosomal RNA clone library from environmental water samples in order to develop more efficient methods for species identification. For the molecular analysis, water samples were collected from the Seonakdong River (Gimhae Bridge) and the coast of Namhae,(Namhaedo). Colony PCR and restriction fragment length polymorphism of PCR (PCR-RFLP) were then adopted to isolate unique clones from the 18S rDNA clone library. Restriction fragment length polymorphism pattern analysis of the Gimhae Bridge sample revealed 44 unique clones from a total of 60 randomly selected clones, while analysis of the Namhae sample revealed 27 unique clones from 150 clones selected at random. A BLAST search and subsequent phylogenetic analysis conducted using the sequences of these clones revealed hidden biodiversity containing a wide range of taxonomic groups (Heterokontophyta (7), Ciliophora (23), Dinophyta (1), Chytridiomycota (1), Rotifera (1) and Arthropoda (11) in the Gimhae Bridge samples Ciliophora (4), Dinophyta (3), Cryptophyta (1), Arthropoda (19) in the Namhae samples). Therefore, the molecular monitoring method developed here can provide additional information regarding the biodiversity and community structure of eukaryotic plankton in environmental samples and helps construct a useful database of biodiversity for aquatic ecosystems.

        세균에 의한 남조 Anabaena cylindrica의 분해에 대한 연구

        A Gram (-), rod-shaped bacterium in size of 1.6-2.8 $\times$ 0.4 μm was isolated from a eutrophic reservoir, which exhibited growth-inhibiting effect against a bluegreen alga (Anabaena cylindrica). This isolate showed positive reactions for catalase and oxidase, and optimal conditions of 35-40°C and pH 9.0. This isolate was designated AC-1 in this manuscript. In a mixed-culture of A. cylindrica and AC-1, their growth patterns were inversely correlated and the bluegreen algal vegetative cells completely disappeared within 24-36 hours. AC-1 showed similar lytic activity in natural water as in an artificial medium. The lytic activity of AC-1 was dependent on the photosynthetic activity of A. cylindrica. When observed under phase contrast microscope, the isolate lysed vegetative cells of A. cylindrica in scattered state in a liquid medium, whereas heterocysts have not been lysed.

        국내 남해안에 발생한 적조원인생물들의 24S rRNA 유전자 염기서열분석

        국내 남해안의 적조발생지역에서 채집한 Prorocentrum minimum, P. micans, P. triestinum, P. balticum, Gymnodinium sanguineum, Alexandrium catenella, Scrippsiella trochoidea, Heterosigma akashiwo를 순수배양하고 이로부터 PCR을 이용해 약 700 bp에 해당되는 24S rRNA 유전자의 D1과 D2 변이부위를 증폭하고 클로닝한 후 염기서열을 분석하였다. 분석된 염기서열들을 이미 밝혀진 적조원인종들의 것과 ClustalW 프로그램을 사용하여 배열하고 계통수를 구성한 결과 Prorocentrum과 Alexandrium 속의 것들은 형태적인 분류결과와 종수준까지 거의 일치함을 보여주었다. 이러한 연구결과는 그 24S rRNA 유전자의 염기서열분석이 국내에서 발생하는 적조원인종들에 대한 종 수준에서의 신속한 확인에 이용될 수 있음을 제시한다. Cultured isolates of Prorocentrum minimum, P. micans, P. triestinum, P. balticum, Gymnodinium sanguineum, Alexandrium catenella, Scrippsiella trochoidea, and Heterosigma akashiwo from red tides in southern coast of Korea were phylogenetically compared on the basis of their 24S rRNA genes. For each isolate approximately 700 bp of 24S rDNA, encompassing D1 and D2 hypervariable domains, was amplified using the polymerase chain reaction and sequenced. The sequences were aligned with those reported in Genbank by using ClustalW program and phylogenetic tree was generated to show that morphospecies designations in the Alexandrium and Prorocentrum species are congruent with terminal taxa defined by phylogenetic analysis of the 24S rRNA fragment. Our results suggest that the molecular phylogeny approach could facilitate identification of domestic dinoflagellates which have ambiguous morphologies.

        부산 연안역의 진핵플랑크톤 종다양성에 대한 메타게놈 분석 연구

        플랑크톤의 종조성에 대한 정보는 해양 생태계 내에서 물질과 에너지 순환을 이해 하는데 필수적이다. 또한 수층의 물리화학적 특성과 변동에 민감하기 때문에 해양환경 특성을 이해하는데 중요한 정보를 제공한다. 본 연구는 메타게놈 분석 기법(metagenomics)을 적용하여 낙동강 유출수, 장강 희석수, 남해 연안수, 쓰시마 난류수 등의 혼합으로 복잡한 환경특성을 가질 것으로 예상되는 부산 연안역의 수괴 내에 존재하는 플랑크톤 종다양성을 분석하였다. 표층 해수에서 18S rDNA 클론라이브러리를 구축하였고 세 정점에서 분석된 370개의 클론들 중에서 94개의 phylotype들을 발굴하였다. 계통분석 결과 phylotype들은 Dinophyceae(42개), Ciliophora(15개), Bacillariophyta(7개), Chlorophyta(2개), Haptophyceae(1개), Metazoa(Arthropoda(17개), Chaetognatha(1개), Cnidaria(2개), Chordata(1개)), Rhizaria(Acantharea(2개),Polycystinea(1개)), Telonemida(1개), Fungi(2개) 등의 다양한 계통군들에 속하는 것으로 나타났다. 근접하게 위치한 세 정점에서 나타난 플랑크톤 종조성 차이는 이들 정점들이 여러 기원의 수괴(water mass) 혼합에 따른 다양한 물리화학적 환경요인의 영향에 기인한 것으로 판단된다. 향후 메타게놈 분석 기법을 통한 플랑크톤 종조성 연구는 다양한 물리화학적 특성을 가진 연안 해양생태계를 이해하는데 유용할 것으로 판단된다. The species composition of plankton is essential to understand the material and energy cycling within marine ecosystem. It also provides the useful information for understanding the properties of marine environments due to its sensitivity to the physicochemical characteristics and variability of water masses. In this study we adopted metagenomics to evaluate eukaryotic plankton species diversity from coastal waters off Busan. Characteristics of water masses at sampling sites is expected to be very complex due to the mixing of various water masses; Nakdong River runoff, Changjiang diluted water (CDW), South Sea coastal water, and Tsushima warm current. 18S rDNA clone libraries were constructed from surface waters at the three sites off Busan. Clone libraries revealed 94 unique phylotypes from 370 clones; Dinophyceae(42 phylotypes), Ciliophora(15 phylotypes), Bacillariophyta(7 phylotypes), Chlorophyta(2 phylotypes), Haptophyceae(1 phylotype), Metazoa(Arthropoda( 17 phylotypes), Chaetognatha(1 phylotypes), Cnidaria(2 phylotypes), Chordata(1 phylotype)), Rhizaria (Acantharea(2 phylotypes), Polycystinea(1 phylotype)), Telonemida(1 phylotype), Fungi(2 phylotypes). The difference in species diversity at the closely located three sites off Busan may be attributed to the various physicochemical properties of water masses at these sites by the mixture of water masses of various origins. Metagenomic study of species composition may provide useful information for understanding marine ecosystem of coastal waters with various physicochemical properties in the near feature.

        낙동강 하구생태계의 세균 생물량과 이차생산성

        부영양화된 낙동강 하구역에서 세균에 의한 용존 유기물의 이용능을 조사하고 이에 미치는 환경요인의 영향을 파악하기 위하여 미생물학적 요인과 물리화학적 환경요인의 연중 변화를 분석하였다. 총세균수는 0.33 ~ $2.09{\times}10^7$ cells/ml의 변화폭을 나타냈으며, 오염부하량이 큰 정점에서 종속영양세균의 개체수와 비례하였다. 세균이 체적과 생물량은 각각 0.064~0.156 ${\mu}m^3$/cell, 0.163~1.036 ${\mu}g$-C/ml을 나타냈다. 세균의 이차생산성은 0.24~60.86 ${\mu}g$-C/l/h의 범위로 측정되었으며, 부영양화 지표로 사용되는 환경인자와 밀접한 관계를 보였다. 담수역에 속하는 정점에서의 세균생산성의 계절별 변화는 동계에 최고치를, 하계에 최저치를 나타내어 갈수기의 오염부하량 증가와 밀접한 상관성을 보였다. 해수역에 속하는 다대의 경우 동계에 낮은 값을, 하계에 높은 값을 나타내고, 환경요인 중 엽록소 a, 수은 등과 밀접한 상관성을 보였다 (r>0.5, p<0.05). 이로 미루어 낙동강 하구의 하구언 상류역에서는 주위에서 유입되는 유기물이, 하구언 하류역에서는 식물성 플랑크톤의 광합성 산무링 세균의 이차생산성에 미치는 주 영양물질임을 암시하고 있다. To investigate the bacterial potentials for utilizing dissolved organic matter in highly eutrophic estuary, the annual fluctuations of microbiological and physicochemical environmental parameters were analyzed in Naktong River Estuary. Total bacterial number ranged from 0.33 to $2.09{\times}10^7$ cells/ml, and correlated with the heterotrophic bacterial numbers in more eutrophic sites, especially. Bacterial biovolume and biomass varied between 0.064 and 0.156 2.09${\mu}m^3$/cell, 0.163 and 1.036 ${\mu}g$-C/ml, respectively. Bacterial secondary productivity ranged from 0.24 to 60.86 ${\mu}g$-C/l/h, and showed high correlations with the environmental parameters of pollution indicator. The seasonal variation pattern of bacterial productivity in freshwater sites was high in winter and low in summer, which was interpreted as the results of pollution loads varied with the amount of rainfall. In seawater site, the pattern was different from those of freshwater sites; high in summer and low in winter. In this site, the values of bacterial productivity showed positive correaltions with chlorophyll a, heterotrophic bacterial number, and temperature (r>0.5, p<0.05). These results suggested that the main source of organic matter which influences the bacterial productivity may be allochthonous materials in the upper freshwater zone of Naktong River Barrage, and autochthonous algal excretory products in the lower seawater zone of Naktong River Barrage.

        낙동강 하류 물금과 을숙도 수환경의 진핵 플랑크톤 종조성에 대한 분자모니터링

        본 연구는 메타게놈 분석법을 기초로 낙동강 하류 담수 환경의 물금과 기수 환경의 을숙도대교 정점에서 채수된 환경 시료내의 진핵 플랑크톤 종다양성 및 군집 구조를 비교 분석하고자 하였다. 수환경 시료에서 추출된 DNA에 대한 environmental Polymerase Chain Reaction(PCR)을 수행하여 18S rDNA 클론라이브러리를 구축하였고, colony PCR, PCR-Restriction Fragment Length Polymorphism(RFLP), 염기서열 결정 및 유사도 분석을 통하여 종다양성을 분석하였다. 물금 및 을숙도대교 정점에서 338개의 클론들을 분석하였고(170 clones, 물금; 168 clones, 을숙도대교), 그 결과 총 74개의 phylotype을 발굴하였다(49개, 물금; 25개, 을숙도대교). 발굴된 phylotype에 대한 계통 분석 결과, Stramenopiles, Cryptophyta, Viridiplantae, Alveolata, Rhizaria, Metazoa 및 Fungi 등의 분류군에 속하는 다양한 생물종이 발굴되었으며, 국내 미기록종 및 신종 후보 가능 생물종과 속(genus)이상의 새로운 분류학적 처리가 필요한 생물종의 존재를 확인하였다. 특히 Stramenopiles의 Pirsonia 및 Alveolata의 Perkinsea에 속하는 phylotypes 등 국내 미기록 생물종을 포함한 숨은 종다양성(cryptic species diversity)의 발굴은 분자모니터링 기법이 낙동강 하구역 수생태계 변화 모니터링을 위한 새로운 유용한 생물학적 정보를 제공할 수 있음을 제시하고 있다. We have studied the eukaryotic plankton species diversity to compare the community structure of fresh and brackish waters in the lower reaches of the Nakdong River using metagenomic methods. We constructed 18S rDNA clone libraries of total DNAs extracted from environmental water samples collected at Mulgeum (MG100929, fresh) and Eulsukdo bridge (ES, brackish). Through the steps of colony PCR, PCR-RFLP, sequencing and similarity analysis, we discovered the diverse species composition of eukaryotic plankton. Total 338 clones (170 at MG100929 and 168 at ES) were analyzed, and then we found 74 phylotypes (49 for MG100929 and 25 for ES). From the phylogenetic analysis, we confirmed various eukaryotic plankton of broad range of taxonomic groups, including Stramenopiles, Cryptophyta, Viridiplantae, Alveolata, Rhizaria, Metazoa, and Fungi. We also found several unreported species in Korea and candidates of new taxonomic entities at levels higher than genus. Especially, the cryptic species diversity including unreported phylotypes of Pirsonia (Stramenopiles) and Perkinsea (Alveolata) suggests that the molecular monitoring method can produce new informative biological data in monitoring the changes in the Nakdong River Mouth ecosystem.

        수종 담수적조 원인종들의 형광특성과 적용연구

        The freshwater blooms mainly blue-green algal blooms occur frequently in the lower Naktong River in summer, which provoke many socio-economical problems; therefore, the early detection of bloom events are demanding through the quantitative and qualitative analyses of blue green algal species. The in vivo fluorescence properties of cultured strains of Microcystis aeruginosa, M. viridis, M. wesenbergii, M. ichthyoblabe, Anabaena cylindrica, A. flos-aquae, and Synedra sp. were investigated. Wild phytoplankton communities of the lower Naktong River were also monitored at four stations in terms of their standing stocks, biomass and fluorescence properties compared with its absorption spectram. The 77K fluorescence emission spectra of each cultured strains normalized at 620 nm was very specific and enabled to detect of blue green algal biomass qualitatively and quantitatively. The relative chlorophyll a concentration determined by chlorophyll fluorescence analysis method showed significant relationship with chlorophyll a concentration determined by solvent extraction method ($R^2$ = 0.906), and the blue-green algal cell number determined by microscopic observation ($R^2$ = 0.588), which gives insight into applications to early detection of blue green algal bloom.
