      • 혈액투석요법 시행 중인 만성 신부전환자에서 혈청 갑상선 홀몬농도에 관한 연구

        To investigate the thyroid hormonal changes and their mechanisms in patients with chronic renal failure, we measured serum levels of the thyroid hormones using rediommunoassay and calculated each ratio of serum thyroid hormones in 20 patients with chronic renal failure who were on hemodialysis therapy. The following results were obtained. 1) The serum T_3 concentrations was significantly lower in patients with chronic renal failure(0.75±0.23㎍/l) than in normal subjects(1.18±0.37㎍/l). 2) The serum T_4 concentrations was significantly lower in patients with CRF(4.96±1.20㎍/l) than in normal subjects(8.45±1.55㎍/dl). 3) The serum FT_4 concentrations was significantly lower in patients with CRF(0.69±0.22ηg/ml) than in normal subjects(1.18±0.15ηg/ml). 4) The serum TSH concentrations in patients with CRF (2.08±2.38μμ/ml) tends to be greater than normal subjects(1.65±0.79μu/ml). But it was not significant statistically. 5) The serum rT_3 concentrations in patients with CRF (21.95±14.79ηg/ml) tends to be greater than normal subjects(18.5±2.80ηg/ml). But it was not significant statistically. 6) The calculated serum concentration ratios of T_3/T_4 and rT_3/T_3 in patients with CRF(0.17±0.09 and 38.30±43.31 : presented as Mean±SEM) dis not differ significantly that of normal subjects(0.14±0.04 and 17.04±5.71). But the serum concentration ratio of rT_3, T_3 in patients with CRF(4.53±2.73) was significantly greater than in normal subjects(2.25±0.50) 7) There was no significant difference in thyroid functions between male & female patients with CRF. 8) The serum T_3, T_4, TSH and rT_3 concentrations not related to the duration of hemodialysis, but serum FT_4 concentrations was lower in patients treated with hemodialysis for more than 36monts(0.49±0.10ηg/ml) than in patients treated with hemodialysis for less than 36months(0.74±0.22ηg/ml). It was concluded that the serum T_3, T_4 & FT_4 concentrations were significantly decreased, while calculated serum concentration ratios of rT_3/T_4 was significantly increased in patients with CRF. Thus we estimates that this decreased thyroid function in patients with CRF may reflect the alteration of serum rT_3 metabolism or increased peripheral conversion of T_4 to rT_3. It is also estimated that chronic hemodialysis leads to a propotional decline of thyroid hormone.

        벼, 콩 및 채소류에 대한 Mn-54, Co-60, Zn-65, Cs-137의 토양-작물체간 전이계수

        벼, 콩, 상치, 당근, 호박의 가식부위에 대하여 몇가지 토양에 있어서 토양-작물체간 Mn-54, Co-60, Zn-65, Cs-137의 전이계수를 포트 재배에 의한 방사성 추적자 흡수실험을 통하여 조사하였다. 핵종간 전이계수는 거의 모든 경우 Zn-65>Mn-54>Cs-137>Co-60의 순이었다. 콩이 벼보다 전반적으로 한 자리 정도 높은 값을 보였고 채소류의 경우에는 대체로 상치에서 가장 높고 호박에서 가장 낮은 값을 보였다. 강산성 토양에서는 약산성 토양에서보다 전이계수가 훨씬 높았다. 본 조사결과에 입각하여 한국인의 섭식경로 피폭선량 평가에 이용하기 위한 각 핵종의 전이계수치가 작물별로 제안되었다. Soil-to plant transfer coefficients of Mn-54, Co-60, Zn-65 and Cs-137 were estimated for the edible parts of the rice, soybean, lettuce, carrot, and squash grown in different soils by radiotracer uptake experiments using pot cultures. The transfer coefficients of radionuclides were n the order of Zn-65>Mn-54>Cs-137>Co-60 in most of the cases studied. The coefficients for soybean were roughly and order of magnitude higher than those for rice. Among vegetables, lettuce mostly showed the highest value and squash, the lowest. In the strongly acidic soils. From the data obtained, crop-specific transfer coefficients of the four nuclides were proposed for the use in Korean food-radiation dose assessment.

      • 화살나무 추출물에 의한 Doxorubicin-유도 조직손상의 억제

        Doxorubicin(DR), a potent antineoplastic drug, is effective in adult and childhood solid tumors and hematologic cancer. However, it has minor(mucositis, nausea, vomiting and alopecia) and major(myelosuppression, cumulative dose-related cardiomyopathy, and skin and subcutaneous ulcers) clinical toxic effects. Accidental subcutaneous DR-extravasation may provoke a prolonged and severe cutaneous inflammatory reaction for which there is currently no simple and effective treatment method. Because of this, apart from measures to prevent paravenous extravasation, many methods of preventing these ulcers have been tried clinically or experimentally, but the efficacy is uncertain. The present study was undertaken to exploit Euonymus alatus extract(EA) as an antidote for DR-induced skin ulceration, using animal model, and to evaluate the effects of EA on the pharmacological activity of DR. Sprague-Dawley rats and ICR mice were intradermally injected DR(0.5 mg/rat and 0.2 mg/ mouse) with or without EA(10 mg/rat and 5 mg/mouse). In rat model, intradermal injection of DR alone produced predictable uniform skin necrosis and ulceration involving the deep dermis. These ulcers developed within 3-7 days, remained stable for 2-4 weeks, and all rats died in 55 days of observation. However, intradermal injection of DR plus EA into rat developed only minor transient edema, erythema and mild inflammation involving the superficial dermis, which healed quickly, and all rats survived throughout the experimental periods. In mouse model, intradermal injection of DR alone produced an ulcer approximately 15.2 mm2 at 5 days. This ulcer was progressively aggravated in size until 15 days, and regressed and healed after that. But, when mice were intradermally injected DR plus EA, the ulcers were reduced in size and healed quickly. These results suggest significant efficacy for EA as a DR-extravasation antidote. B16 cells, 3LL cells and human tonsillar lymphocytes were cultured in the presence of DR with or without EA, and the proliferation responses of these cells were evaluated. DR alone de-creased remarkably the proliferation of malignant cells as well as normal cells, in a dose-dependent fashion. On the other hand, the EA itself reduced weakly the responses of tumor cells but increased those of normal cells. And EA also slightly enhanced the DR- induced inhibition of the tumor cell-proliferation, but it restored the DR-derived decrease of the lymphocyte-prol-iferation. These results suggest that EA itself has no side effects, and also suggest that EA may possibly diminish the systemic toxic effects of DR-administration. Taken together, the present study reveals that EA has an antidotic activity against DR-ex-travasations and systemic DR. And these results suggest the possibility that EA could be developed and used in a form of DR-annex as an antidotic and reinforcing agent of DR.

      • 신장세포암에서 WT1과 IGF2 유전자의 발현

        IGF2 유전자는 제 11번 염색체 단완 (11p15.5)에 위치해 있으며 아버지로부터 유전된 대립형질만 기능을 하는 genomic imprinting되는 유전자이다. 이 유전자는 정상세포에서는 세포증식을 촉진시키는 작용을 하고 몇가지 암에서는 LOI에 기인한 과발현이 있는 것으로 알려져 있다. 한편 WT1 유전자는 제 11번 염색체 단완 (11p13)에 위치해 있으며 어머니로부터 유전된 대립형질만 기능을 하는 즉 IGF2 유전자와는 정반대의 genomic imprinting 되는 유전자이다. 이 유전자는 비뇨생식기관의 발생에 매우 중요한 유전자이며 암화과정에서는 암억제 유전자로서 기능한다. 소아신장에서 발생되는 Wilms 종양에서는 IGF2 유전자와 WT1 유전자의 고장이 주원인으로 생각하고 있다. 이에 근거하여 성인에서 발생되는 신장세포암에서도 Wilms 종양에서의 결과와 같을 것인지의 여부를 알기 위하여 12명의 신장세포암 환자 ( 남자 6명, 여자 6명)로부터 얻은 조직을 이용하여 이들 유전자의 발현정도를 조사하였다. 환자의 연령은 남자에서는 43세에서 56세까지였고 여자에서는 23세에서 61세까지였다. 신장세포암에서 IGF2 유전자와 WT1 유전자의 발현정도는 다같이 성별이나 연령과는 무관하였다. IGF2 유전자는 50%에서 발현되었으며, 그 발현정도는 저발현에서부터 중등도 그리고 과발현까지 다양했으며 주로 신세뇨관 부위에 있는 암세포에서 발현되었다. WT1은 56%에서 발현되었으며 과발현과 중등도 발현이 대부분이었다. IGF2 유전저의 발현과 WT1 유전자의 발현은 각 예에서 상이하게 나타나서 두 유전자간의 작용은 일치하지 않았다. IGF2 gene which was located on chromosome 11p15.5 is expressed by the paternal allele. It stimulates the cell proliferation in normal cells, however, it is over expressed in some cancers due to LOI (loss of genomic imprinting). In contrast, WT1 gene which was located on chromosome 11p13 shows maternal specific monoallelic expression. It is a very much important gene in developing urogenital system, besides, it acts as tumor suppressor gene in some cancers. IGF2 and WT1 genes are the main causative genes in Wilms tumor which is developed in pediatric kidney. To understand the status of IGF2 and WT1 expressions in renal cell carcinoma which is developed in adult life, tissues from 12 cases patients (male 6 cases ; female 6 cases) were detected by immunohistochemistry with antibodies from IGF2 and WT1 genes. To distinguish the different ages between Wilms tumor and renal cell carcinoma, the patients age in renal cell carcinoma was ranged in 43-56 years old in man and 23-61 years old in female. No direct correlation was, in general, between gene expression of IGF2 and WT1 and age or sex of patients in renal cell carcinoma. Expression of IGF2 gene revealed six of twelve (560%) and expression of WT1 gene showed seven of twelve (56%) in renal cell carcinoma, respectively. The status of IGF2 gene expression was varied from low to high expression. In contrast, the status of WT1 gene expression was higher than in IGF2 gene expression. However, there was no apparent correlation between expressions in each cases. In conclusion, IGF2 and WT1 genes are very important in carcinogenesis of renal cell carcinoma, however interaction between two genes was obscure.

        지방척수수막류 14례의 임상 분석 : Clinical Analysis of 14 Cases

        Lipomyelomeningocele is the most common malformation leading to spinal cord tethering. Nonetheless, these lesion continues to pose a challenge in patient management. The purpose of this study was to identify the clinical feature, candidate for surgical repair, and outcome of lipomyelomeningocele in young children and adults. We studied the magnetic resonance images and/or lumbar myelo-CT of lipomyelomeningocele performed between January 1987 and September 1995 at our institutions and reviewed the pertinent medical records and radiologic studies to determine clinical feature, surgical candidate, and outcomes of these patients. Of the 14 cases(11 female and 3 males) studied, the most common factor that caused these patients to seek help was the cosmetic effect of the mass on their back, followed by urinary incontinence, and weakness of lower extremities. The patients' age ranged from 2 months to 21 years(mean, 5.0 years). All the patients underwent immediate surgical management that consisted of removal of lipoma, untethering of the cord, and complete dural sac repair. Follow up for these patients ranged from 6 to 82 months(median, 54 mo). None of the patients that underwent surgical correction before the emergence of neurological deficit had ever developed neurological deficits at the end of the follow up. However a few of the patients, despite presented with neurological deficits or urinary incontinence at the time of surgery, improved significantly. In conclusion, early detection and prophylactic surgery is imperative to prevent permanent neurological deficits and urinary dysfunction.

        This is the continuous study about "A Study of Facade Characters of Building in City Street and Their Effects on Characters of Street Environment (Ⅰ)." In this report, the formal characters of the building-such as a commercial or an office at about the same time when Taegu is raised to the statue of a special city were studied. So, these results will offer the design criteria to design better form of the building and will be the usefull information to research the change of city street in the future.
