        췌장 베타세포에서 인터루킨-1beta로 유도한 인슐린 의존형 당뇨병 실험 모델

        To establish prediabetes in vitro/ model concerning the etiology of Insulin Dependent Diabetes Mellitus (IDDM) in cellular level we have designed experimental prediabefic model in pancreatic beta cells. RINm5F, HIT-T15 and isolated rat islets were chosen as pancreatic beta cells. Since interleukin-1beta-induced beta cell cytotoxicity has been implicated in the autoimmune cytotoxicity of IDDM, we used inteleukin-1beta as diabetogenic agent. For establishment of prediabetic in vitro model, the degree of beta cell deterioration was determined by cell proliferation, insulin release and morphological appearance. Cell proliferation, insulin release and morphology were changed dose-dependently in condition that inteleuldn-1beta was exposured to pancreatic beta cells. The concentration and exposure time of interleukin-1beta to set up prediabetic model in beta cell lines and isolated rat islets were 100-1000U/ml, 48hr. And 25-100U/ml, 48hr, respectively.

        췌장소도세포와 면역세포에 미치는 팔미원의 영향

        In order to investigate the usability of PALMIWON as antidiabetic immuno-modulating prescription for Insulin-dependent diabetes, we studied the effects of PALMIWON on immune cell and beta-cell. U937 was used as the model of immune cell and RINm5F as the model of beta-cell. The effects of PALMIWON was measured by cell viability in terms of MTT assay. As a result, PALMIWON and the compositional drugs showed the different effects on immune cell and beta-cell. Cell viability of U937 was significantly decreased whereas that of RINm5F was not significantly different from drug treated group and control, or significantly less reduction compared with U937. It implies that PALMIWON is useful as immunotherapeutic agents in the prevention and therapy of type 1 diabetes.

        췌장 베타세포에서 스트렙토조토신으로 유도한 인슐린 의존형 당뇨병 실험 모델

        To establish prediabetes in vitro model concerning the etiology of IDDM(Insulin Dependent Diabetes Mellitus) in cellular level we have designed prediabetes in vitro models in pancreatic beta cells. HIT-T15, RINm5F and isolated rat islets were chosen as pancreatic beta cells, and streptozotocin (STZ) used as diabetogenic agent. Degree of beta cell destruction to establish prediabetic in vitro model was determined by cell proliferation and insulin release using thymidine uptake and radio immuno assay. When HIT-T15 and RINm5F cells were treated with STZ, the degree of cell deterioration was dependent upon the origin and passage number of beta cells, and in the case of isolated islets STZ showed the more sensitivity than above two beta cell lines. The concentration and exposure time of STZ treatment to establish prediabetes in vitro model in beta cell lines and isolated rat islets were 2 ~ 10mM, 30 min. and 1 ~ 5mM, 30 min., respectively.

        팔미원의 in vitro 면역조절 작용

        PALMI\ON is composed of 8 oriental herbs which has been known to show some pharmacological effects in kidney, blood vessels and immune systems, and used for the treatment of kidney disease, hypertension, nervous disease and diabetic mellitus in the Orient for a long time. Based on our previous report that PALMI\ON showed different effects on immune cells and 0-cells, the immunoreactivity of ICSA (Islet Cell Surface Antibody) with beta-cell (RINm5F) and the cell proliferation and function of interleukin-1beta damaged beta-cells in the presence of PALMIWON were examined. It was observed that PALMIWON significantly inhibited the immunoreactivity of ICSA with beta-cell, and markedly increased cell proliferation and insulin release of interleukin-1beta damaged beta-cells.

        The aim of the present study was to investigate the effect of Palmiwon on the type 1-prediabetic models induced by streptozotocin (STZ) in RINmSF cells and HIT-T15 cells. Palmiwon increased the cell proliferation and insulin release when pre- and post-treated for the STZ-exposed pancreatic beta cells. The cell proliferation and insulin release of these beta cells were measured by ³H-thymidine uptake and RIA, respectively. We also analyzed nutrients such as sugars, fatty acid and amino acids and minerals containing in Palmiwon using by gas chromatography, amino acid analyzer and AA spectrometer, respectively. Palmiwon seems to have protective and recovery properties on the prediabetic model in cellular level, which were ascribe to various nutrients and minerals containing in Paliniwon. From these results, it could be suggested that Palmiwon may be available as preventive and therapeutic prescription of type 1 diabetes mellitus.

      • 담즙성 간경변증시에 발생한 신사구체 및 신세뇨관의 초미형태학적 변화에 관현 연구

        It has been well known that biliary cirrhosis with concomirtant impairment of renal functions were occured after prolonged obstruction of biliary tract. In the present study, an attempt was made to investigate the mechanism of impaired function and the ultrastructural change of the kidney in rats, ligated the common bile duct for 28 days days order to induce secondary biliary cirrhosis. The results obtained were as follows; 1. Secondary cirrhosis was produced evidently 28 days after ligation of common bile duct. Prominent proliferations of bile ducts and were seen in periportal area. 2. There were electron lucent materials forming lump in the subendothelium. 3. Indistinct three layers and focal widening of basement membranes were observed. 4. Focal fusions, edema and bleb formations of foot processes were seen, Electron dense materials with variable size and global shape were found in edematous foot processes. 5. Multiple myelin figures with varible size were evoked in the cytoplasm of the proximal convoluted tubule. 6. Ovoid materials with variable size were found in the mesangial matrix.
