Spider silks have great potential as biomaterials with extraordinary properties. Here, we report the cloning and characterization of the major ampullate silk protein gene from the spider Araneus ventricosus. A cDNA encoding the partial major ampullate silk protein (AvMaSp) was cloned from A. ventricosus. An analysis of the cDNA sequence shows that AvMaSp consists of a 240 amino acid repetitive region and a 99 amino acid C-terminal non-repetitive domain. The peptide motifs that were found in the spider major ampullate silk proteins, (A)n, (GA)n, and (GGX)n, were conserved in the repetitive region of AvMaSp. Phylogenetic analysis further confirmed that AvMaSp belongs to the spider major ampullate spidroin family of proteins. The AvMaSp-R cDNA, which encodes the 240 amino acid repetitive domain, was expressed as a soluble 22 kDa polypeptide in baculovirus-infected insect cells. Recombinant AvMaSp-R was degraded abruptly by trypsin. However, AvMaSp-R was stable at 100 °C for at least 30 min. Additionally, the AvMaSp-R was stable at pH values from 2 to 12 for at least 1 h. Taken together, our findings describe the molecular structure and biochemical properties of the A. ventricosus major ampullate silk protein and demonstrate its potential as a biomaterial.

Accumulation of unfolded proteins in the endoplasmic reticulum (ER) triggers the transcriptional induction of molecular chaperones and folding enzymes localized in the ER. Thus, eukaryotic cells possess an intracellular signaling pathway from the ER to the nucleus, called the unfolded protein response (UPR) pathway. To obtain genes related to UPR from Bombyx mori Bm5 cell, the cDNA library was constructed with mRNA isolated from Bm5 cell line in which N-glycosylation was inhibited by tunicamycin treatment (5 ㎍/㎖). From the cDNA library, we selected 40 clones that differentially expressed when cells treated with tunicamycin, and produced expressed sequence tags (ESTs). Among these clones, we have isolated TmInc329 clone showing high similarity with ATF (encodes a bZIP transcription factor) of M. muscululs. Basic-luecine zipper (bZIP) domain in amino acid sequences of TmInc329 shared homology with several transcription factors, yeast Haclp, human CREB and mouse ATF. Also, TmInc329 clone is up-regulated when N-glycosylation of newly synthesized proteins in the ER is inhibited by tunicamycin treatment. Therefore we suggest that TmInc329 clone is a gene responding to accumulation of unfolded proteins in the ER.
전력용 XLPE Cable의 부분방전과 방사전자파 특성

본 논문은 XLPE(cross-linked polyethylene insulated power Cables)을 절연물로 사용하는 22.9[kV]용 전력케이블에 외부 손상에 의해 발생되는 부분 방전을 모의하였으며, 절연 진단방법 중 하나인 UHF법을 이용하여 부분 방전시 공간으로 전파되는 방사전자파을 EMI, EMC 측정용 BiconiLog 안테나(EMCO 3142)와 스펙트럼 아날라이저를 이용해 측정하는 UHF법을 이용하였다. 본 연구 결과 방전진전과정의 평가가 가능함을 확인하였다. This paper simulated partial discharge caused by 22.9[kV] power cable using XLPE(Peroxide Crosslinkable Polyethylene) insulation having the outside damage. As one of the insulation diagnostic method a radiated electromagnetic waves were measured by an UHF method using a BiconiLog antenna(EMCO-3142) and a spectrum analyzer used to measure EMI, EMC. From results of this study, It was confirmed that discharge Ogress were possible to be estimated by the proposed method.

Sr-Nd-Pb isotopic results are reported for the Ogcheon amphibolites from the central part of the Ogcheon Belt. Rb-Sr and Pb-Pb whole rock isotope data plot greatly scattered in the isochron diagrams due to later alteration or metamorphism, whereas the Sm-Nd whole rock isotope data define a linear array with an age of $1270{\pm}220$ Ma ($1{\sigma}$). Considering several geochemical features of the amphibolites, the 1270 Ma linear array may be not a true but an apparent mixing isochron due to source heterogeneity.
