Under the background of Chengdu Chongqing double city economic circle, through the analysis of the current situation and problems of the ideological and political construction and development of Chengdu Chongqing universities, combined with the advantages of Sichuan and Chongqing universities, this paper seeks measures to improve the ideological and political construction and development of Chengdu Chongqing universities. It is found that the ideological and political construction and development of colleges and universities in Chengdu and Chongqing can be improved mainly by strengthening the ideological and political education model and reforming and innovating, integrating and optimizing relevant educational resources; Specific measures include integrating online media, strengthening practical teaching, optimizing teachers and improving infrastructure construction.

成渝双城经济圈作为国家重要的协同发展战略, 对成渝地区教育、人才培养发展、人才联育方面, 提供了历史性的机遇, 人才作为社会、国家重要的发展动力, 对于实现民族振兴、赢得国际竞争主动权的战略资源具有重要作用。本文立足于成渝双城经济圈政策实行的大背景下, 深入探讨实现人才创新发展的路径与基础, 在切实分析成渝双城经济圈高校人才培养模式现状后, 通过结合成渝地区重点高校报告和相关数据进行比较研究, 提出所面临的缺陷与不足等相关问题的合理可行的建议。 As an important national cooperative development strategy, Chengdu Chongqing double city economic circle provides a historic opportunity for education, talent training and development, and Talent Association in Chengdu-Chongqing area. As an important driving force for social and national development, talent plays an important role in realizing national revitalization and winning the strategic resources of international competition initiative. Based on the background of the implementation of the policy of Chengdu Chongqing double city economic circle, this paper deeply explores the path and basis for the development of talent innovation. After the actual analysis of the current situation of the talent training mode in Chengdu Chongqing double city economic circle, through the comparison of the report of key universities in Chengdu-Chongqing area and related data, it puts forward reasonable and feasible suggestions on the defects and deficiencies faced and other related issues.

The purpose of this study is to prevent the stick, scuffing, scratch between piston and cylinder in advance, to obtain data for duration test in actual engine operation. Therefore, temperature gradient in peripheral and axial distribution of cylinder bore were measured in use of 800cc class gasoline engine. The results obtained are summarized as follows ;<br/> 1) The maximum reaction force by overlap is 101.97kg. The skirt length at that position is 10mm from skirt upper.<br/> 2) The rear side temperature of top dead center is 142℃ in peripheral distribution, it is about 20℃higher than thrust, anti-thrust, front side temperature.<br/> 3) The rear side temperature of bottom dead center is 125℃ in axial distribution, whereas the rear side of midway position temperature is 120℃.<br/> 4) The temperature of cylinder wall is about 20-30℃ higher than temperature of cooling water.<br/> 5) The temperature of cylinder wall at top dead center in use of SAE 30^* lubricating oil is 16℃ higher than SAE 20^# at 6000rpm.<br/>
HlyA 유전자 Primer를 이용한 PCR에 의한 식품으로부터 Listeria monocytogenes의 신속 검출 방법

본 연구는 식품에 존재하는 L. monocytogenes균의 신속한 검출방법을 조사하기 위하여 hlyA 유전자 primer를 사용하여 PCR 기법으로 행하였으며 primer의 특이성과 PCR의 민감성, L. monocytogenes균의 검출을 위한 최적조건 및 우유 및 소고기의 적용시험을 각각 조사하였다. PCR 특이성 실험에서는 L. monocytogenes 20주에 대한 PCR 분석결과 모두 713 bp 크기의 PCR products를 확인할 수 있었고, Listeria spp. 및 다른 세균에서는 동일한 poducts가 증폭되지 않으므로써 hlyA based primer의 L. monocytogenes에 대한 PCR 특이성이 관찰되었다. PCR 분석법의 민감도는 L. monocytogenes ATCC 19111의 1 pg DNA와 2.4×10⁴ cell 수준에서 target DNA가 증폭되었다. Tailing의 제거와 민감도를 높이기 위하여 PCR cycle 수에 따른 최적조건을 조사한 결과 20~30cycle 정도의 PCR clcyle 수가 좋은 것으로 나타났으며 민감도는 20 cycle PCR amplification을 행한 후 한번더 10~15 cycle로 처리했을 때 훨씬 증가하였다. 한편, 우유(10 mL)와 쇠고기 절편(10 g)에 0~10^7 CFU/mL 또는 g 수준으로 L. monocytogenes를 신속하게 검출하기 위하여 PCR을 적용한 결과, 우유시료에서는 2번 PCR을 반복함으로써 2 cells까지 검출할 수 있었고, 쇠고기 절편은 LEB로 35℃에서 24시간 증균하여 PCR함으로써 2.6×10² cell까지 검출할 수 있었다. The study was conducted to develop a rapid method for the detection of Listeria monocytogenes in foods via polymerase chain reaction (PCR) technique using hemolysin gene (hlyA) primers. Specificity and sensitivity of PCR, optimal conditions for PCR and application of hlyA gene primers for the detection of L. monocytogenes from milk and beef were investigeted. Each of the 20 L. monocytogenes strains gave a single 713 bp band, but other Listeria spp. and other bacteria did not show any bands. As few as 1 pg of L. monocytogenes DNA or 2.4×10⁴ L. monocytogenes cells could be detected with hlyA gene primers. PCR product was most improved at 20~30 cycle in terms of removal of tailing and sensitivity. Also, the sensitivity was significantly improved by the further 10~15 cycle after 20 cycle PCR amplication. Milk (10 mL) and beef (10 g) samples were inoculated with L. monocytogenes at the concentrations ranging from 0 to 10^7 CFU/mL or g to determine the best sensitivity of PCR for the rapid detection of L. monocytogenes. PCR assay could detect 2 cells in milk with repeating PCR amplication and 2.6×10² cells in beef sample after 24 hr enrichment growth at 35℃ in LEB.
