        24개월 이상 선천 코눈물관막힘 환아의 단계적 탐침술을 동반한 단, 상하누소관 실리콘관 효과

        Purpose: The clinical effectiveness of monocanalicular or bicanalicular intubation with sequential probing was evaluated in patients over the age of 24 months with congenital nasolacrimal duct obstruction. Methods: Patients over 24 months of age with congenital nasolacrimal duct obstruction who underwent monocanalicular intubation with sequential probing (19 patients, 20 eyes) or bicanalicular intubation with sequential probing (22 patients, 22 eyes) were studied. Success rates and complications were evaluated. Silicone tube was removed 6 months after surgery. Success was defined as no epiphora and no retention on fluorescein dye disappearance test. Results: The success rate was 95.0% (19 eyes / 20 eyes) in the monocanalicular intubation group and complications included 7 cases of early tube dislodgement, which achieved successful outcome. The success rate was 82.6% (19 eyes / 22 eyes) in the bicanalicular intubation group and complications included 4 cases of punctal slitting, and 3 cases of tube extrusion. The success and complication rates were not significantly different between the 2 groups (p = 0.608, p = 1.000, respectively). Conclusions: In congenital nasolacrimal duct obstruction, the monocanalicular tube intubation group had similar success and complication rates to the bicanalicular tube intubation group. Silicone tube maintenance for 2 months in the monocanalicular group and for 3 months in the bicanalicular group was sufficient. J Korean Ophthalmol Soc 2013;54(11):1635-1640

        Effects of Argon Laser Iridotomy on the Corneal Endothelium of Pigmented Rabbit Eyes

        Purpose: In Asian countries, laser iridotomy for the treatment of angle-closure glaucoma is a common cause ofbullous keratopathy, which may be associated with a shallow anterior chamber and dark iris pigmentation inAsians. Several cases of corneal decompensation after argon laser iridotomy have been reported. In the presentstudy, we evaluated the harmful effects of argon laser iridotomy on the corneal endothelium. Methods: Argon laser iridotomy was performed on the right eyes of pigmented rabbits. Changes in cornealthickness and endothelial cell density after laser iridotomy were evaluated. Terminal deoxynucleotidyl transferasedUTP nick end labeling (TUNEL) was performed for assessment of corneal endothelial cell apoptosis. Combined staining with alizarin red and trypan blue, as well as a live/dead cell assay, were performed for evaluationof damage to the corneal endothelium induced by laser iridotomy. Results: Corneal thickness did not change immediately after laser iridotomy; however, a significant increasewas observed 24 hours after iridotomy (p = 0.001). The endothelial cell density of laser-treated eyes four daysafter laser iridotomy was significantly decreased compared with control eyes (p < 0.001). TUNEL stainingshowed many TUNEL-positive cells in the corneal endothelium and corneal stroma. No endothelial trypanblue-stained cell nuclei were observed after laser iridotomy; however, several large endothelial cells with damagedmembrane integrity were observed. The live/dead cell assay clearly showed a large number of deadcells stained red in several areas throughout the entire corneal button 24 hours after iridotomy. Conclusions: Argon laser iridotomy induces corneal endothelial cell apoptosis in pigmented rabbit eyes, resultingin decreased endothelial cell density.

        CDISC 표준에 기반한 임상시험 데이터베이스의 설계

        In this paper, a database for clinical trials which conforms to the standards developed by CDISCis proposed. The proposed database consists of CDISC metadata and CRF database. The CDISCmetadata database allows mapping CDASH standard variables of CRFs to SDTM ones of rawdatasets. The CRF database decomposes CRFs into data items specified in ODM, and stores theitems. By utilizing the databases, it is possible to maintain CRFs and their raw datasets to generateODM XML that describes CRFs. The ODM XML will be transformed to HTML or PDF files byapplying XSLT. 본 논문에서는 CDISC에서 개발한 국제표준을 준수하는 임상시험 데이터베이스를 설계한다. 설계한 임상시험데이터베이스는 CDISC 메타데이터 데이터베이스와 증례기록지 데이터베이스로 구성된다. CDISC 메타데이터 데이터베이스는 CDASH 표준과 SDTM 표준을 통합하여, 증례기록지의 데이터 항목을 원시 데이터셑의 변수에 매핑한다. 증례기록지 데이터베이스는 ODM 표준에서 정의한 구조를 이용하여, 증례기록지의 내용을 분해하여 저장한다. 설계한 임상시험 데이터베이스는 CDISC 표준에 따른 증례기록지와 원시 데이터셑을 동시에 유지할 수 있다. 또한,증례기록지를 ODM XML 파일로 생성 가능하며, 이 파일은 XSLT 변환을 거쳐 HTML 파일로 화면에 디스플레이되거나 PDF 파일로 출력될 수 있다.

        국제표준에 기반한 전자증례기록지의 생성

        In this paper, we develop a system that generates electronic case report forms (CRFs) conforming to the CDASH, SDTM and ODM standards developed by the CDISC consortium. With the system, users edits CRFs using the CDISC metadata database which seamlessly integrates metadata provided by both the CDASH and SDTM standards, and stores the edited CRFs into the CRF database based on the ODM standard. The proposed system also generates SDTM raw datasets without user intervention. The system provides user interfaces with which users can fast and easily edit CRFs without prior knowledge or understanding of the CDISC standards. This will greatly reduce setup times of clinical trials. 본 논문에서는 CDISC에서 개발한 CDASH, SDTM 및 ODM 국제표준을 준수하는 전자증례기록지를 생성하는 시스템을 개발한다. 개발한 시스템은 CDASH와 SDTM 표준을 통합한 CDISC 메타데이터 데이터베이스를 사용하여 증례기록지를 편집하고, ODM 표준에 기반한 증례기록지 데이터베이스에 편집한 내용을 저장한다. 개발한 시스템은 편집한 증례기록지뿐만 아니라 원시 데이터셋도 CDASH 및 SDTM 표준에 따라 데이터를 유지한다. 또한 본 시스템은 증례기록지의 콘텐츠 및 양식을 쉽고 편하게 편집하는 사용자 인터페이스를 제공한다. 사용자는 CDISC 표준에 대한 지식이 없어도 CDISC 표준에 기반한 증례기록지를 편집하고 생성할 수 있으며, 이를 통해 임상시험 셑업 기간을 크게 단축할 수 있다.

        Intracellular delivery of recombinant proteins via gold nanoparticle– DNA aptamer composites is independent of the protein physicochemical properties and cell type

        Here, we report that the gold nanoparticle–DNA aptamer (AuNP–Apt) conjugate-based system canefficiently deliver recombinant proteins into mammalian cells in a manner independent of their size,isoelectric point, and cellular localization. Additionally, AuNP–Apt system-assisted protein delivery canbe effective on primary and stem cells, indicating that its use is not limited to fast-dividing cells. Wefurther show that the intravenously administered AuNP–Apt system can deliver proteins into rat organs. Ourfindings show that this system can serve as a simple, efficient, and versatile platform for the deliveryof recombinant proteins into mammalian living systems.

        SH-SY5Y Human Neuroblastoma Cell에서 에스트로겐의 신경세포 보호효과에 대한 기전

        Apoptosis is one major mechanism underlying neuronal cell death in degenerative diseases in the central nervous system. Previous studies have shown that estrogen has neuroprotective effects in several neuronal cell death model systems. In the present study, we established a staurosporine-induced apoptotic neuronal cell death system with human SH-SY5Y cell line. 17b-estradiol(E2) blocked staurosporine-induced apoptosis of SH-SY5Y cells as revealed by MTT as well as LDH assay. The cells showed typical DNA fragmentation at 4 h and 8 h following staurosporine treatment, which was attenuated by E2 pretreatment. Staurosporine-induced chromatin condensation was also reduced by E2 treatment. Because apoptotic stimuli are known to induce caspase-3 activation and PARP cleavage, we examined whether E2 blocks these processes. E2 markedly attenuated staurosporine-induced casapase-3 activation and PARP cleavage. This study shows that E2 blocks staurosporine-induced apoptosis in SH-SY5Y human neuroblastoma cells, suggesting that E2 is a neuroprotective agent that may be used for the treatment of neurodegenerative diseases such as Alzheimer's disease and Parkinson's disease.

        본 논문은 MPEG-7 BiM 규격에 따라 XML 스키마 정의를 기반으로 바이너리 형태로 압축하고 복원하는 시스템의 구현에 관한 것이다. MPEG-7 BiM 압축기 및 복원기의 세부 모듈과 기능을 서술하고, 설계 및 구현방법을 제안한다. 구현된 MPEG-7 BiM 압축기 및 복원기는 대역폭의 제약이 심한 방송 분야에서 메타데이터 전송을 위한 핵심 모듈로 사용 될 수 있다.
