These studies were designed to determine the potential cytotoxic activity of MeOH extracts of 65 crude drugs against leukemia L1210 and $P388D_1$, cell line in vitro, of which 25 samples were selected. The n-BuOH extracts of 25 samples were measured using the same method and nine were selected. These samples were measured for the potential antitumor activity against $P388D_1$, for life span in vivo, and against sarcoma 180 for tumor weight. On the basis of results, Notoginseng Radix, Anemarrhenae Rhizoma, Albizziae Cortex, Portulacae Herba, Bupleuri Radix were evaluated the effective plant on the antitumor activity. In addition, the mixture of Notoginseng Radix and Albizziae Cortex was evaluated to enhance the antitumor activity in vivo.
하천수에서 Campylobacter jejuni 의 오염도와 그 생존율에 관한 연구

Campylobacter jejuni isolated from river and lake waters and chicken samples were examined for their contamination and survival in surface waters and for inactivation by chlorination. The isolation rates of C. jejuni from chicken samples were 50 to 100%, while only one out of 48 studies was positive from the surface water. When the organisms suspended in sterile river water were incubated at 4℃, they survived upto 20 days, but they died off after 1 day incubation at 30℃ or above. The in situ survival studies of C. jejuni in the waters of Dsecheong Dam and Han-river (1st bridge) revealed that they survived upto 14 days in winter, but for shorter period of time in spring and summer. The survival rates of the organism in the deeper water were a little longer than in the surface water, but the rates were not different between the both water environments. When the organisms were treated with 0. 5, 1. 0 and 2. 0 ㎎/ℓ chlorine solution at pH 7. 0 and 15℃ for 60 minutes, they were inactivated by about 1, 4, and 6 logs, respectively, The inactivation rate was increasingly affected by lowering the reaction pH and by increasing the reaction temperature.
한국 Streptomyces sp. 로 부터 분리한 방향족 화합물과 지질 화합물의 세포독성 연구

In an effort to screen new selective antitumor agents from the broth of soil microorganism, cytotoxicity oriented screening was performed against tumor cells and 3 compounds (Compound 1, 2 and 3) were isolated from Streptomyces parvullus ISP 5048 and their chemical structures were determined. Among these compounds, Compound 2 showed the highest cytotoxicity against P388D1 and L1210. While the IC_(50) values of compound 2 against P388D1 and L1210 were 0.073 ㎍/㎖ and 0.07 ㎍/㎖, respectively, and the IC_(50) value of Compound 3 was 0.17 ㎍/㎖ against human lung cancer cells, A549, the cytotoxicity of Compound 2 and 3 against normal cell line, Vero E6 cell was about 4- and 8-fold lower than that of adriamycin. Based on the chemical analysis data, Compound 3 was octacosamicine A, a known antibiotic, which was reported by Dobasih et al. (1988). Taken together the results demonstrated that Compound 2 and Compound 3 has the possibility to be developed as antitumor agent because of its potent cytotoxicity as well as high selectivity against various cancer cell lines.

An Actinomycete strain isolated from Mt. Dea-Dun had a strong antifungal activity. The culture brith produced by isolated strain showed only antifungal activity against fungi with the exception of yeast and bacteria. It was heat stable, dissolved in ehtylacetate. The concentrated antifungal agent showed cytotoxicity against HEP-2 and HeLa as tumor cell line, and showed weak cytotoxicity against VERO 36 as normal cell line. Morphological and physiological characteristics were tested with isolated strain. The spore color of isolated strain was gray. It had a short chain and produced brown colored lytic substance in yeast extract-malt agar. The cell wall of isolated strain was composed of meso-DAP, and we suggested it as genus Actinomadura. In the existing of chemical inhibitor, isolated strain grew on the condition of 0.0001% crystal violet, 0.1% phenol, 0.01% sodium azide and 10% sodium chloride. Carbon utilization of isolated strain was shown that glucose, sucrose, manitol and sodium citrate were well utilized.
약수에서 분리한 Yersinia pseudotuberculosis의 병원성과 16S rDNA 분석에 의한 분자학적 분류

In order to investigate the pathogenicity and development of differential identification technique in the Yersinia species and other entericbacteria, we isolated 5 strains of Y.pseudotuberculosis from spring water sites in Seoul. The biochemical characteristics of isolated strains revealed that indole, VP($25^{\circ}C$, $37^{\circ}C$), $H_2S$, phenylalanine, lysine, arginine, ornithine, gas from glucose, lactose, sucrose, sorbitol, oxidase and motility($37^{\circ}C$) were all negative and urease, glucose, mannitol, salicin, catalase and motility($25^{\circ}C$) were all positive. To detect the causative agent of pseudotuberculosis(Y.pseudotuberculosis), we carried out a study using a PCR with inv primers complementary to the pathogenic region and found that all strains were positive, this revealed that strains from spring waters were pathogenic. Also 16S rDNA for total 5 strains of Y. pseudotuberculosis were amplified and a stretch of approximately 1,450 nucleotides were sequenced and analyzed. The 16S rDNA nucleotide sequence homologies among Yersinia species ranged 97.5% to 100% and between Y.pseudotuberculosis and other entericbacteria they ranged 93.0% to 95.1%. The Phylogenetic tree generated from the sequence analysis of the 16S rDNA gene showed 3 coherent clusters that could be separated into Y.pseudotuberculsis strains, some Yersinia species strains and other entericbacteria strains. 서울시내 25개 자치구에 산재한 약수터에서 5주의 Yersinia pseudotuberculosis를 분리하여 생화학적 특성, 병원성의 유무 및 16S rDNA 분석을 실시하였다. 분리된 Y. pseudotuberculosis는 모두 병원성 유전자인 inv를 소유하고 있었으며, 16S rDNA를 증폭한 후 염기서열을 분석하여 NCBI Genbank에 등록된 다른 Yersinia 속 및 장내세균 등과 비교해 본 결과 Yersinia 속 등과는 97.5%에서 100%의 높은 상동성(Similarity)을 나타내었고, 다른 장내세균 등과는 93.0%에서 95.1%의 낮은 상동성을 나타내었다. 165 rDNA 염기서열을 기초로 계통수를 작성한 결과 크게 3개의 cluster를 형성하였는데 특히 Y.enterocolitica (Z49830)은 Y.pseudotuberculasis (Z21939)와의 상동성(97.7%)보다 Y.intermedia (X75279)와의 상동성(97.9%)이 더 높게 나타났으며, E. coli (Z83205)는 Proteus vulgaris (AJ233425) 와의 상동성(93.2%)보다 Salmonella enteritidis (U90318)와의 상동성(97.7%)이 더 밀접한 연관성을 나타내었다.

An Actinomycetes strain JB isolated from Mt. Hanla had a strong antimicrobial activity against gram positive bacteria and tumor cell growth inhibition. Especially, it couldn''t degrade starch and casein as organic compounds. It was resist on lincomycin and rifampicin. The spore mass of strain JB which was arethrospore was white. DAP of the cell wall was L,L-DAP. Antimicrobial material was heat stable, dissolved in ethyl acetate, and not dissolved in butanol. In the pressnce of 0.1% phenol and 4% sodium chloride, strain JB could grow, but it didn''t growth at below 10oC . Strain JB didn''t use dextran, sodium acetate and sodium citrate as sole carbon source and L-cystein and L-thereonine as nitrogen source. The filtered broth of strain JB had the antimicrobial activity against gram positive bacteria, especially Staphylococcus aureus (ATCC 65389) and the growth inhibition of tumor cell line.
Streptomyces 속 야생균주들이 생산하는 세포독성물질과 plasmid 와의 연관성에 관한 연구

We isolated Streptomyces spp. from Korean soil, which showed high cytotoxicity against tumor cell lines, L1210 and P388D1. Among 30 strains, three strains (DKM 104, DKM 128, DKM 409) were appeared to possess plasmid. Strain DKM 104 and DKM 128 had two CCC(Covalently Closed Circular) form plasmid, about 20 Kb in size and about 1 Kb compared with λ Hind III DNA size marker. And strain DKM 409 had three plasmids, among which two plasmid were CCC form about 1 Kb and about 20 Kb compared with same size marker. To find out whether plasmid involved in production of antitumor agent or not, we performed to curing experiment. Comparing cytotoxicity between culture filtrate of plasmid-containing strains and cured strains, we knew that only the cytotoxic activity of the strain DKM 128 was involved in plasmid.
