        한국, 중국, 일본에서 자생하는 ‘ 마 ’ 속 식물의 화학 성분과 활성

        Plants of genus Dioscorea have long been served as important carbohydrate-stuffed foods in the tropical and subtropical regions, and utilized as traditional herb medicines to enhance digestive function, improve anorexia, and treat diarrhe a in oriental countries. It is known that around 600 species of Dioscorea are distributed in the world including 107 species in Asia, but actually utilized Dioscorea species are restricted to small numbers. Phytochemical investigations for Dioscorea species have revealed a number of chemical components such as sapogenins, saponins, phenanthrenes, stilbenes, diterpenes and purine derivatives. According to recent pharmacological studies, Dioscorea species possess significant antioxidant, antibacterial and anti-inflammatory activities as well as anticancer, antidiabetic, cholesterol-lowering and hypolipidemic effects. Here, sev en Dioscorea species (D. batatas, D. japonica, D. bulbifera, D. opposita, D. tokoro, D. nipponica and D. alata), mainly distributed and used in Korea, China, and Japan, are reviewed to provide their botanical, phytochemical and pharmacological properties were described.

        Determination of Allantoin in Dioscorea Rhizoma by High Performance Liquid Chromatography Using Cyano Columns

        An easy and reliable HPLC method was developed to determine allantoin in Dioscorea Rhizoma using cyano columns. Qualitative and quantitative analyses of allantoin were performed successfully by cyano columns (YMC-Pack CN column, Zorbax SB-CN column and Discovery® Cyano column). The intraday precision were 0.58 - 3.33% for YMC-Pack CN, 0.41 - 2.20 for Zorbax SB-CN and 0.45 - 1.93% Discovery® Cyano columns, while interday variations were 0.09 - 1.84%, 0.04 - 2.59% and 0.87 - 5.18% for YMC-Pack CN, Zorbax SB-CN and Discovery® Cyano columns. The recoveries of allantoin were in the range at 98.8 - 102.6% (RSD 1.1 - 1.6%) for YMC-Pack CN column, 99.7 - 110.5% (RSD 1.3 - 4.9%) for Zorbax SB-CN column, and 97.2 - 110.1% (RSD 1.8 - 5.7%) for Discovery® Cyano column. The contents of allantoin in four Dioscorea Rhizoma samples were determined by cyano columms and ranged at 4.1-7.1 mg/g dry weight. The present study indicated that HPLC method using cyano column for determining allantoin is a reliable method and this method can be applied to verify allantoin in Dioscorea Rhizoma.

        황칠나무 추출물의 항산화 및 간세포보호효과

        Dendropanax morbifera Leveille, an endemic species in Korea, is best known as a tree that produces a resinous sap. Although D. morbifera is used in folk medicine for various diseases, its active ingredients are largely unknown. In this study, we investigated antioxidative activities of ethanolic extracts of three parts of this plant including leaves, debarked stems, and roots. The root extracts exhibited strong 1,1-diphenyl-2-picrylhydrazyl radical (DPPH) scavenging activity compared with leaf or stem extracts. The root extracts showed hepatoprotective activity against t-butyl hydroperoxide-induced HepG2 cells, and reduced the ROS level in the cells. The root fractions lowered the mRNA level of COX-2 on lipopolysaccharide-stimulated Raw246.7 cells. These results suggest that ethanolic root extracts of D. morbifera are a source of antioxidant and hepatoprotective compounds, which indicate a potential for a botanical drug.

        Dihydrobenzofuran Neolignans Isolated from Euonymus alatus Leaves and Twigs Attenuated Inflammatory Responses in the Activated RAW264.7 Macrophage Cells

        Anti-inflammatory effects of dihydrobenzofuran neolignans isolated from Euonymus alatus leaves and twigs were evaluated in lipopolysaccharide (LPS)-stimulated RAW264.7 macrophage cells. Six neolignans, (+)-simulanol (1), (+)-dehydrodiconiferyl alcohol (2), (-)-simulanol (3), (-)-dehydrodiconiferyl alcohol (4), (+)-dihydrodehyrodiconiferyl alcohol (5), threo-buddlenol B (6) effectively inhibited the production of nitric oxide (NO) induced by LPS, and the activity of iNOS. (-)-dehydrodiconiferyl alcohol (4), which showed the most potent inhibitory activity, attenuated the activity of iNOS enzyme and also the expression of iNOS and COX-2 proteins. The subsequent production of pro-inflammatory cytokines, interleukin-1b, interleukin-6, tumor necrosis factor-a and prostaglandin E2 were also inhibited by the pretreatment of RAW264.7 cells with (-)-dehydrodiconiferyl alcohol (4). These neolignans are thought to contribute to anti-inflammatory effects of E. alatus, and expected to be potential candidates to prevent/treat inflammation-related diseases.

        Terpenoid constituents from the aerial parts of Asplenium scolopendrium

        Dioscorea japonica Thunb. Ethanolic Extract Attenuated Oxazolone-Induced Atopic Dermatitis-like Skin Lesions in BALB/c Mice

        The rhizomes of Dioscorea japonica Thunb. are widely consumed as food and also used to treat diabetes and polyuria in Korea. This study was undertaken to study the anti-atopic dermatitis effects of a 95% ethanolic extract (DJE) of D. japonica in an oxazolone-stimulated murine model of atopic dermatitis (AD). The therapeutic effects of DJE on AD-like skin lesions were assessed on both ears. DJE (1%) or dexamethasone (0.5%; the positive control) were applied to skin lesions for three weeks. Serum levels of IgE and IL-4 were assessed by ELISA (enzyme-linked immunosorbent assay). Histopathological examinations were performed by hematoxylin and eosin (H&E) and toluidine blue staining and revealed DJE significantly reduced dermal thickness and inflammatory cell infiltration when applied to oxazolone-treated ear skin. DJE-treated AD mice also showed lower serum levels of IgE and IL-4 than oxazolone-stimulated controls. Our findings demonstrate DJE might be a useful safe, topical agent for the treatment of atopic diseases.

        Development of Gradient Centrifugal Partition Chromatography Method and Its Application for the Isolation of 3,5-Dimethoxyphenanthrene-2,7-diol and Batatasin-I from Dioscorea opposita

        Gradient centrifugal partition chromatography (GCPC) method was developed and applied to isolate 3,5-dimethoxyphenanthrene-2,7-diol (DMP) and batatasin-I (BA-I) from the dichloromethane soluble extract of Dioscorea opposita. In this method, the lower phase of n-hexane-methanol-water system (HMW, 10 : 9 : 1, v/v) was used as a mobile phase A (MpA) and water was used as a mobile phase B (MpB). This gradient CPC method is comparable to that of reversed-phase HPLC method in that the stationary upper-phase of HMW (10 : 9 : 1 v/v) works as if it were reversed-phase silica gel due to its hydrophobic property, while the lower phase (MpA) and water (MpB) functioned as hydrophilic mobile phases. The initial condition of the mobile phase was 20% MpA/ 80% MpB and maintained for 150 min to obtain DMP (1.2 mg), and then MpA was increased up to 50% to elute BA-I (1.7 mg). The purities of DMP and BA-I were 94.1% and 98.3% with the recovery yields of 83% and 86%, respectively. Similar results were obtained by linear-gradient CPC. The CPC peak fractions were identified by comparing their retention time to those of authentic samples of DMP and BA-I and their spectroscopic data (1H NMR and 13C NMR) to those of literature values.

        Complete NMR assignments of undegraded asterosaponins from Asterias amurensis

        Four asterosaponins, thornasteroside A (1),versicoside A (2), anasteroside B (3), and asteronylpentaglycosidesulfate (4), were isolated from the predatorystarfish Asterias amurensis Lu¨tken. Unlike previous studiesfocusing on structure elucidation by degradation of thecomplex saponin molecules, complete nuclear magneticresonance (NMR) assignment for the intact molecules wasaccomplished using 600 MHz high magnetic field NMR. The complete set of NMR assignments can help in thestructure elucidation of asterosaponins isolated in lowyields without resorting to chemical degradation. Furthermore,this approach can be extended to other complexsteroidal saponins, which may accelerate the discovery ofbioactive secondary metabolites from this invasive starfishspecies.

        베이커리 매장 내 구매촉진을 위한 사용자 경험 연구

        본 연구는 베이커리 매장 내 물리적 환경요소에 따른 감각 경험과 문화 체험 경험이 구매의사 및 브랜드 매력도에 미치는 영향 요인을 밝혀내어, 방문고객의 경험 만족 증진을 위한 개선점 도출 및 방향성 제시를 목적한다. 위 목적을 실현시키기 위한 문헌연구로서 구매의사에 영향을 미치는 매장 내 물리적 환경 요소와 사용자 및 브랜드 경험, 사용자 경험을 측정하는 방법론에 관한 이론적 토대를 마련하였으며, 이를 바탕으로 구매의사에 영향을 미치는 매장 내 물리적 환경소를 감각 경험 요소와 문화 체험 경험 요소로 구분 및 선정하였다. 연구의 객관적인 결과 도출을 위해 현재 업계 내 선두 브랜드인 파리바게트와 뚜레쥬르를 선정, 비교 평가를 실시하였다. 실증연구로서 물리적 환경요소를 기반으로 한 설문조사(questionnaire survey)와 포커스 그룹 인터뷰(focus group interview)를 병행하여 실시하였고, 섀도우 트랙킹(shadow tracking)을 통해 베이커리 매장 내에서의 고객 여정을 관찰하였다. 추가로 아이 트랙킹(eye tracking)을 통해 피 실험자의 실제 시선 산재도 및 집중도 등을 추적하여 인터뷰 응답내용에 따른 진위여부를 파악하는 등의 보완작업을 수행하였다. 위 실험을 통해 도출된 결론 및 제안 사항은 다음과 같다. 첫째, 고객 여정에 따른 경험요소, 즉 터치 포인트를 고려한 경험 스토리 구성이 필요하다. 둘째, 제품 자체의 매력도를 높이기 위해 물리적 환경요인이 종합적으로 고려되어야 한다. 셋째,사용자(고객) 시선에 따른 높이,각도 등이 고려된 효과적 디스플레이 진열방식이 이루어져야 한다. 넷째, 제품 정보와 관련된 시각물, 쟁반이나 집게, 의자, 테이블 등 고객의 편의성을 돕는 요소들의 사용성 관리가 중요하다. 본 연구의 궁극적 취지는 방문고객의 감각 경험에 근거한 구매요인 도출에 의의가 있으나, 매장 내 감각 경험 요소에만 주안점을 두었음에 연구의 한계점을 밝힌다. 따라서 향후 베이커리 매장의 물리적 환경 요소 중 문화 체험 경험 요소에 대한 추가적 보완연구가 이루어진다면 베이커리 업체의 충성고객을 확보하기 위한 전략 수립에 도움이 될 것이다. The objectives of this study are, to define in-store environment affects shoppers" sensory and cultural experiences can affect purchase decision making and brand preferences, and to develop to-be-direction of shoppers’ experience enhancement to increase their satisfaction in bakery industry. We divided the physical environment elements, influencing shoppers in their purchase decision making, into sensual elements and cultural experiences elements from the preliminary theory. And we validated those two category of elements by filed-validating Paris Baguette and Tous les Jour. For field research, we have tried to find shoppers insight through survey and focus group interview, observed shoppers" path in stores by shadow tracking, tracked down shoppers" eye-sight by eye tracking and validated research data by field visits. Based on implication from the research, we suggest the followings: To organize shoppers experience storyline considering touch points in shoppers’ path in stores. Physical environment should be strategically considered to increase product preference. Product display should be based on height and angle of shoppers" eye-sight. Usability maintenance of POP, plate, tongs, table and chair is also very important. However, it also shows some limits by focusing mainly in sensory experiences in stores. If it is followed by additional study of shoppers interactive and cultural experiences in stores, it will be more helpful to establish strategy of increasing loyal shoppers in bakery industry.

        방풍의 화학 성분

        The roots of Saposhnikovia divaricata Schischk. (Umbelliferae) have been known to possess analgesic, anti-inflammatory, anti-parasitic and anti-bacterial activities, and used for curing headaches, fever and arthralgia. In this study, we ai med to isolate active constituents to provide phytochemical data for the quality control of this plant. Nine coumarins, eight chromones, three sterols and a coumarolignan were isolated from EtOAc-soluble fraction of the roots of S. divaricata through repetive column chromatography method using silica gel, ODS gel, Sephadex-LH 20, MPLC and HPLC. By analyses of spectroscopic data and comparison of their data with those of published values, the compounds were identified as 3'-O-angeloylhamaudol (1), β-sitosterol (2), marmesin (3), phellopterin (4), anomalin (5), imperatorin (6), xanthotoxin (7), deltoin (8), bergapten (9), stigmasterol (10), ledebouriellol (11), hamaudol (12), 8'-epicleomiscosin A (13), xanthoarnol (14), cimifugin (15), 5-O-methylvisamminol (16), daucosterol (17), 4'-O-β-D-glucosyl-5-O-methylvisamminol (18), nodakenin (19), sec-O-glucosylhamaudol (20), prim-O-glucosylcimifugin (21). Among them, 8'-epicleomiscosin (13) was firstly reported from Umbelliferae family and xanthoarnol (14) and nodakenin (19) were isolated from this plant for the first time.
