도축 폐혈액 단백질로부터의 Angiotensin I Converting Enzyme 저해 펩타이드의 생산

본 연구는 도축 폐기물인 가축혈액을 이용하여 항고혈압 기능성 식품소재로서의 angiotensin I converting enzyme 저해 펩타이드 분획을 생산하기 위한 조건과 가능성을 조사하기 위하여 수행되었다. 산업적으로 이용 가능한 단백분해효소 중 Alcalase가 혈장 원액 및 그로부터 분리된 albumin에 대하여 가장 높은 활성의 가수분해물을 생성하였다. 특히 albumin의 Alcalase 가수분해물과 이를 gel chromatography를 통새 분획하여 얻은 고활성 분획의 $IC_50$값은 각각 0.5 및 0.02 mg/mL로서 지금까지 보고된 식품단백질 유래 펩타이드 혼합물들과 비교할 때 활성이 매우 높은 것에 속함을 알 수 있었다. 또한 이 고활성 펩타이드 분획은 혈장 원액으로부터 단순한 한외여과만을 거쳐도 얻을 수 있음을 확인함으로써 산업적 실용화 가능성이 높은 공정임을 알게 되었다. For the production of angiotensin I converting enzyme inhibitory peptides as a material for antihypertensive functional foods from animal blood produced in slaughterhouse, the optimum condition for enzymatic hydrolysis to yield a peptide fraction of the highest activity were investigated with a respect of industrial production. Among several industrially-usable enzymes tested, $Alcalase^?$ produced hydrolysates of the highest activity from total plasma and purified albumin. $IC_50$ values of albumin hydrolysate and its third fraction separated by gel chromatography were 0.5 and 0.02 mg/mL, respectively. The fraction was found to be obtained by a simple ultrafiltration using a membrane of MW cutoff 1,000. The possibility for the industrial production of antihypertensive peptides from animal blood plasma protein was suggested.

Central pontine and extrapontine myelinolysis(CPM) is clinically charaterized by quadriplegia, pseudobulbar palsy, and consciousness change, usually following rapid correction of hyponatremia, but the exact mechanism of the pathophysiology remains obscure. Pathologic study reveals well-circumscribed areas of myelin loss sparing neurons and axons in pons and extrapontine region. Vascular changes and inflammation are absent. In the past, the diagnosis of CPM has generally been made on the basis of autopsy findings, but at the present, magnetic resonance imaging has made it possible to make the diagnosis before autopsy for the detection of pontine and extrapontine lesions. We report three cases of central pontine and extrapontine myelinolysis associated with typical clinical symptoms following rapid correction of hyponatremia. MRI showed high-signal areas in basis pontis, bilateral thalamus, caudate nucleus, and putamen.

To find out the urokinase concentration at which free form plasmin begin to form in plasma, we measured plasmin from the normal pooled plasma of various concentration of urokinase between 300 unit/ml and 2.4 unit/ml, by chromogenic method (S-2251) and SDS-PAGE with fibrin-agar zymography. Free form plasmin begin to be detected by both chromogenic method and SDS-PAGE with fibrin-agar zymography between 5∼10 unit/ml of urokinase. This finding suggest that the urokinase concentration should be around 10 unit/ml for the effective fibrinolysis during intravenous administration.
