Daunomycin의 흰쥐 태아의 경골 근위골단 연골판의 기질합성에 미치는 영향

Daunomycin, an anthracycline antibiotic anticancer drug, was isolated from the culture of Streptomyces peucetius. It is well known that daunomycin intercalates between DNA base pairs to inhibits DNA and RNA synthesis by above mentioned mechanisms. Although daunomycin is very effective against many tumors, it may also damage to proliferating normal cells. In this experiment, the author pursued the effect of daunomycin on the matrix of tibial epiphyseal plate in rat fetus. The pregnant albino rats, Spraque-Dawley strain, were used as experimental animals. Triple doses of 5mg of daunomycin per kilogram body weight were administrated to experimental animals at 8th, 10th and 12th days of pregnancy. The fetal rats were delivered at 15th, 17th, 19th and 21th days of pregnancy by Cesareansection method. The specimens, obtained from the proximal tibia, were stained with hematoxylin-eosin to observe general structure of epiphyseal plate and with several specific method to observe the nature of the specific mucopolysaccharide. The alcian blue (pH 2.5) and alcian blue (pH2.5)-PAS for detecting the distribution of acid and neutral mucopolysaccharide, alcian blue (pH 1.0)-PAS, alcian blue (pH 0.4) and aldehyde fuchsin (pH 1.7)-alcian blue (pH 2.5) for observing the distribution and intensity of sulfated and nonsulfated mucin. The rsults obtained were as follows. 1. In experimental animals treated with daunomycin, the reserving zone of proximal tibial epiphyseal plate was expanded and proliferating zone was narrowed. 2. In experimental animals treated with daunomycin, acidic and neutral mucopolysaccharide in proximal tibial epiphyseal plate was decreased. 3. In experimental animals treated with daunomycin, the content of sulfated mucin was increased in proximal tibial epiphyseal plate. In according to these results it is consequently assumed that daunomycin inhibits the differentiation of the chondrocyte in proximal tibial epiphyseal plate and synthesis of mucopolysaccharide in proximal tibial epiphyseal plate. But the sulfation of mucopolysaccharide in proximal epiphyseal plate is accelerated.
