        Cyclodextrin 분해효소의 정제 및 그 특성

        Cyclodextrin hydrolase from Bacallus stearodhermophilus KFCC 21203 was purified and the properties of the purified enzyme were investigated. The enzyme was purified 15 folds with 77% recovery by ammonium sulfate fractionation, DEAE-cellulose chromatography, and Ultro AcA 34 gel filtration. The specific activity and the molecular weight of the enzyme were 1.30 units/㎎ protein and about 29,500, respectively. The maximum activity of the enzyme was shown at 55℃ and pH 5.5. However, stable temperature and pH were 40 ℃ and 5.0∼8.0, respectively. The Km value for γ-cyclodextrin was 3.78 × 10^(-3) M. The degradation activity of the enzyme was selectively high for γ-cyclodextrin, and very low for β-cyclodextrin, but not for α-cyclodextrin. The decomposed products of γ-cyclodextrin were mainly glucose and maltose, and a little mlatotriose. The activity of the enzyme was very high for amylose, potato starch, corn starch, amylopectin and maltooligomer, and relatively high for glycogen and dextrin. The decomposed products of them were mainly glucose and maltose.

      • 烏藥順氣散의 소염 진통작용에 관한 연구

        Ohyaksungisan. combined preparation of crude drugs, has been used for hemiplegia, arthralgia and paralysis in traditional Korean medicine. The anti-inflammatory activity of the aqueous extract from Ohyaksungisan(OSSE) was investigated on acetic acid-induced edema and adjuvant arthritis in rats. Acute toxicity and analgesic action in mice were also examined. Its anti-inflammatory activity on 5% acetic acid-induced edema and adjuvant arthritis was observed with oral administration. The acute toxicity showed 10% mortality at 2400㎎/㎏(p.o). but was not showed at 1200㎎/㎏(i.p). OSSE was showed to have significant analgesic action(P<0.05) at 150㎎/㎏ and this action was strengthened at 300, 600㎎/㎏. The anti-inflammatory effect was showed significant preventive effect on the hind paw edema from 90 min, and the adjuvant arthritis. when orally administered for 19 days. showed significant inhibitory effect on the hind paw edema from the 5th day.

      • NaCl 및 N.P.K. 濃度가 油菜種子 發芽에 미치는 影響

        油菜品種別 內監性程度와 N.P.K. 施用이 發芽에 미치는 影響을 究明하고자 試驗을 實施하여 얻은 結果를 要約하면 다음과 같다. 1. 油菜品種別 內監性程度는 木浦350號 >木浦179號 >木浦130號 >木浦132號 順으로 强하였다. 2. NaCl 1.0%以下에서는 發芽에 큰 影響을 주지 않았으나 NaCl 1.5%以上에서의 發芽率은 현저히 減少되었다. 3. 各 品種 共히 限界監分濃度는 NaCl 2.0%였다. 4. NaCl 濃度가 增加함에 따라 發芽勢는 減少되었으며 發芽期間은 연장되었다. 5. NaCl 0.5%以上에서 監分濃度가 增加함에 따라 生育은 현저히 沮害를 받았다. 6. 監分濃度가 增加함에 따라 T/R ratio는 현저히 增加되었다. 7. N.P.K. 施用에 따른 發芽率은 NaCl 0區에서는 肥種間에 差가 없었지만 NaCl 0.8%區에서는 urea 處理區에서 현저히 減少된 반면, ammonium sulphate區에서 현저히 높았다. The experiments were conducted to investigate the effects of the degree of salt tolerance of Rape varieties and N. P. K. application on germination. The results obtained are as follows; The degree of salt tolerance in Rape varieties was shown to be strung in the following order; Mokpa 350 > 179 > 130 > 132. Below 1.0% NaCl did not greatly influence germination but at over 1.5% NaCl germination rate decreased remarkably. For all varieties, the limit far salt concentration was 2.0% NaCl. In accordance with increased NaCl concentration: germination speed decreased and the germination period was extended. In accordance with increased salt concentration over 0.5% NaCl; growth and development was highly retarded. In accordance with increased salt concentration; T/R ratio was increased remarkably. In accordance with N. P. K. application; germination rate showed no difference difference among kinds of fertilizer at 0% NaCl but at 0.8% NaCI those ammonium sulfate plot showed a remarkably increase, while on the other hand, those with urea treatment showed a remarkably decrease.

      • 猪笭湯이 實驗的 賢贓障碍家兎에 미치는 影響

        This study was attempted to investigate the effect of Jueryeong Tang(JRT) on renal failure of the rabbit treated with folate. Serum creatinine, blood urea nitrogen and serum uric acid in folate-treated rabbits were markedly increased and creatinine clearance was decreased by comparing with the normal group. JRT(900㎎/㎏) given orally produced significant reduction in concentration of both serum creatinine and blood urea nitrogen by comparing with the corresponding folate-treated group. while did increased clearly in creatinine clearance. The morphological changes of arcuate arteries and glomeruli evoked by folate treatment were almost returned to normal state after administration of JRT. (900㎎/㎏). These experimental results suggest that JRT may greatly improve renal failure of the rabbit induced by folate treatment.

        『復古編』 「聯綿字」 고찰

        『復古編』是宋代張有編纂的字書。 第六卷「聯綿字」裏最早提出“聯綿字”, 而且收錄了58個詞。 雖然書裏沒提到“聯綿字”的槪念, 但通過對58條項目的分析可以確認「聯綿字」在聯綿詞硏究上的價値。在語音方面每條項目中說明每個字的反切, 所以可以對兩個字的音轉關係加以分析。 58個詞當中90%的詞屬於雙聲或疊韻。 這意味着張有已認識到聯綿詞的語音特徵。在字形方面「聯綿字」有很高的硏究價値。 它最早把聯綿詞的異形放在一個項目裏, 而且說明一個是正體, 另外一個是俗體。 根據這些資料我們可以判斷哪個是原來的形態, 同時還可以考察聯綿詞的字形特徵、 造詞方式, 從而可以進一步掌握中國人有關漢字的思惟體系。在語義方面「聯綿字」裏提到的詞義和各個字義爲考察聯綿詞整個詞義的形成特徵提供了資料。總而言之, 『復古編』中的「聯綿字」是聯綿詞硏究史上最早的著作, 可是它所提供的語音、字形、 語義資料以及這些資料的價値並不是初步的。 在本人看來結合元代編纂的『續復古編』「聯綿字」進行硏究的話, 對今後的聯綿詞硏究會有更大的幇助。

      • 탱자의 成分硏究 (Ⅰ) : Hesperidin의 分離 The isolation of Hesperidin

        The trifoliate orange(Poncirus Trifoliata Rafinesque: Rutaceae) is produced at the south of middle repion in Korea. The fruit has been used for the G-I tract at the name of Ji-Sil(枳實)from the chinese medicine. I found the needle crystal(A) by extracting the fruit of trifoliate orange with ethanol. This crystal(A) is the same as the standard with melting point 259℃ After I examined spot by radiating the Ultra-Violet rays with the developmental method of Wattman No2 of paper partition chromatography. I found that Rf is 0.86 in phenolsolution as the developmental fluid, Rf is 0.89∼0.90 in acetic acid: H₂O(6:4) and the melting point is 228∼229℃ by hydrolysis with HCl. As the result, I could find that Crystal(A) is Hesperidin.
