Effects of Adipose-Derived Stem Cells on Keloid Fibroblasts Based on Paracrine Function

Adipose-derived stem cells (ASCs) possess multipotent differentiation capabilities, including the ability to repair and regenerate injured tissues. Recent studies have reported that ASCs have effects on fibroblasts that are related to wound healing. On the other hand, keloid is a result of pathologic wound healing, and keloid fibroblasts are known to have higher growth potential and an abnormal balance between proliferation and apoptosis. This study aimed to explore the paracrine effects of ASCs on normal fibroblasts (NF) and keloid fibroblasts (KF). NF and KF were cultured alone and in combination with ASCs using a trans-well cell-contact-independent cell culture system. Cell morphology was observed, cell proliferation was assessed using a CCK-8 kit, and apoptosis was assessed by live-dead-cell staining on days 1, 3, and 7. Western blot analysis at 24 hours after culture was used to investigate the production of type 1 collagen and alphasmooth muscle actin (α-SMA). Co-culture with ASCs resulted in increased proliferation of both NF and KF. In the co-culture with ASCs, collagen production decreased and expression of α-SMA increased in both the NF and KF. Comparison between the four groups revealed a significant difference in the expression of both type I collagen and α-SMA (p=0.016 and 0.022; Kruskal-Wallis test). Our findings are different from previous studies that used ASCs for normal dermal fibroblast and normal wound healing. However, considering pathologic mechanisms of keloid, ASCs may have the different effect on keloid fibroblasts, which could be another effect of ASCs modulating keloid during healing process.

Purpose: To validate and update a nomogram for predicting ductal carcinoma in situ (DCIS) upstaging in preoperative biopsy. Materials and Methods: Medical records of 444 preoperative DCIS patients were evaluated and used to validate a previous versionof the Severance nomogram for predicting DCIS upstaging in preoperative biopsy. Patients were divided into two groups accordingto the final postoperative pathology. Univariate and multivariate analyses with the chi-square test, Student’s t-test, andbinary logistic regression method identified new significant variables. The updated nomogram was evaluated with the C-index andHosmer—Lemeshow goodness of fit test. Results: The area under a receiver operating characteristic curve for comparison with the previous nomogram was 0.48. In postoperativepathology, the pure DCIS and invasive cancer groups comprised 345 and 99 cases, respectively. Approximately 22.3% ofpatients preoperatively diagnosed with DCIS were upstaged to invasive cancer. Significant variables in the univariate analysis wereoperation type, human epidermal growth factor receptor 2 overexpression, comedo necrosis, sonographic mass, mammographicmass, preoperative biopsy method, and suspicious microinvasion in preoperative biopsy. In multivariate analysis, operation type,sonographic mass, mammographic mass, and suspicious microinvasion were risk factors for upstaging. The updated model withthese variables showed moderate discrimination and was appropriate in the calibration test. Conclusion: The previous nomogram did not effectively discriminate upstaging of preoperative DCIS in an independent cohort. An updated version of the nomogram appears to provide more accurate information for predicting preoperative DCIS upstaging.

Purpose: Secondary correction of unilateral cleft lip and nose deformity is necessary in number of patients despite advanced techniques and overall treatment philosophy. Various techniques and modifications have been reported by many centers. But the definite procedure for secondary unilateral cleft lip nasal deformity is the question under debate. Methods: Secondary cleft lip and nosede formity has its own anatomical abnormalities, including incomplete release of nasal lining, incomplete alar cinching, and progressive septal deviation. In order to correct these anatomical abnormalities, we have performed the procedures of complete release of congenital cicatricial contracture of nasal lining, extended cinching of alar, suspension of orbicularis oris muscle, and anteriorly limited open rhinoplasty. Results: Between June of 2001 and Feb of 2006, 45 patients with secondary cleft lip and nose deformity underwent correction according to our procedures. During 36 month period of follow up, there were no significant complications. Esthetic and functional improvement was identified. Conclusion: Consequently, total mobilization of all the displaced anatomical structures and placing them in a normal position are the basic and essential in correction of secondary cleft lip and nose deformity

Although Companies are forming cooperation and networks with various companies such as technology and R&D, but it is urgently necessary to secure multiple competencies of companies that can effectively adapt. This study investigated the effects of corporate strategic orientation, technological innovation capability, and organizational capability on corporate performance targeting SMEs located in the Daegu area. The survey was conducted from September 1 to November 30, 2021 for employees of Daegu companies, and a total of 183 cases were used for the final analysis. Looking at the results of the analysis, as a result of multiple regression analysis on Daegu companies, it was found that the strategic orientation, technological innovation capability, and organizational capability of managers, all independent variables, had a significant positive (+) effect on corporate performance. It can be seen that efforts should be made to improve education and R&D according to various competency bases and innovations because of the large impact on the manager's mind, the company's core technical competency, and the company's organizational performance. This study aimed to understand the importance of corporate strategic orientation, technological innovation capability, and organizational capability on corporate performance targeting relevant SMEs that can have a significant impact on national and regional economies.
