      • 방광암 환자의 혈청 Neutral RNase의 활성도와 특징에 대한 연구

        In order to investigate whether clinical values of serum ribonucleases (RNase) could be used as markers for bladder cancer, activities and positive rates of neutral and acid RNases in serum of patients with bladder cancer were determined and compared using normal healthy persons as control subjects. Two peaks each for RNase were isolated from serum in both patients with bladder cancer and normal controls. The two peak enzymes were subject to analyse substrate specificity and protein patterns to find out whether cancer specific RNase was present in the serum of patients with bladder cancer. 1) Activities of neutral and acid RNases measured using pyrimidine polyribonucleotide or RNA as substrate were significantly increased in serum of patients with bladder cancer compared with normal controls. 2) When serum neutral and acid RNase activities were measured in the superficial and invasive types of bladder cancer, activities of the two enzymes were significantly higher in the invasive type than in the superficial type. The positive rate of the enzyme as a marker for bladder cancer was relatively higher in the activity of neutral RNase measured with poly C as substrate. These results indicated the possibility that the activity of serum neutral RNase measured with poly C could be used as a diagnostic parameter for bladder cancer. 3) Two peaks for neutral RNase were obtained in serum of patients with bladder cancer by DEAE-cellulose column chromatography. Of the two enzymes, the peak Ⅱ neutral RNase in bladder cancer serum appeared to be different from that in normal in terms of the substrate specificity and electrophoretic protein patterns. The results suggested that the peak Ⅱ neutral RNase in bladder cancer serum might be different from normal and specific for bladder cancer.

        Trichophyton rubrum으로부터 세포외 단백분해효소의 분리 정제 및 특성 연구

        Background: Trichophyton rubrum is the most common dermatophyte isolated from human and has ability to invade the tissues such as stratum corneum, nail and hair. The potential role of proteinases as virulence factors of T. rubrum has been discussed at length. Objective: As a first step towards assessing its virulence role, we report on the purification and characterization of proteinase from T. rubrum isolate culture filtrates. Methods: An extracellular serine proteinase has been purified from culture filtrates of Trichophyton rubrum HP-9 by ultrafiltration, gel filtration chromatography, and affinity column chromatography. Azocoll and keratin azure were employed as the substrates of enzyme activities. Peak of proteolytic activity was analyzed by gelatin co-polymerized gel electrophoresis. Results: The molecular weight of the purified enzyme was approximately exhibited to 14.0 kDa on sodium dodecyl sulfate polyarcylamide gel electrophoresis (SDS-PAGE). The optimum pH and molality of 14.0 kDa proteinase activity was 6.0 and 100mM, respectively. The activity was inhibited by serine proteinase inhibitor, phenylmethylsulfonyl fluoride (PMSF). The proteinase degraded gelatin, collagen type VI, and keratin from human epidermis but not hemoglobin. Conclusion: The 14,000 Mr extracellular serine proteinase purified from T. rubrum NP-9 culture filtrates has neutral pH optimum 6.0 and activities against gelatin, collagen type VI, and keratin.

      • 양념류를 첨가한 전통고추장의 숙성 및 저장 중의 특성 변화

        Microbial and physicochemical characteristics of traditional kochujang with addition of garlic or onion, were investigated for 52 weeks of aging to information on shelf-life of kochujang. Viable cells of yeast in the kochujang decreased during aging with the increasing ratio of garlic or onion. However, aerobic and anaerobic bacterial counts did not show any remarkable differences in the garlic or onion added kochujang , they increased slowly until 37 weeks of aging. The enzyme activities in kochujang decreased slowly during aging, but those showed the highest activities for garlic or onion added. The pH and titratable acidity of kochujang changed a little by addition of garlic or onion. Reducing sugar contents of kochujang decreased slowly during aging, but alcohol contents increased. As the ratio of garlic or onion increased, reducing sugar and alcohol contents showing highest value. Amino nitrogen contents of kochujang were higher in garlic or onion added, but ammonia nitrogen contents were low during aging, Water activities of kochujang decreased during aging with the increasing ratio of garlic or onion, but consistency of 4~6% garlic added group were higher than that of others. The color values of kochujang tended to decrease during aging in a and b values, and those of high values in the case of garlic or onion added kochujang.

        계획지원을 위한 공간 빅데이터의 분석과 활용 : 수도권 지역의 건물에너지 데이터를 대상으로

        The advancement and spread of information and communication technology (ICT) has changed the way we live and act today. Computers and ICT devices have become smaller and almost invisible, and they are now virtually everywhere in the world. Most socio-economic activities are now subject to the use of computers and ICT devices, although we do not really recognize it due to pervasive computing technologies. Nevertheless, the activities supported by digital devices leave digital records, and a myriad of these records becomes what we call “big data.” Big data differs from conventional data that we have collected and managed in that it holds detailed information of people’s lives and activities. Thus, it offers new insight into our society, and hence a new opportunity for innovation. Not surprisingly, experts and scholars forecast that big data will transform various socio-economic activities as well as our societies. The goals of this research are twofold: First, it aims to understand the nature and characteristics of spatial big data. Then, it seeks to suggest ways of utilizing such spatial big data for planning support. In this regard, this study defines the concept of “planning a support system for big data,” which consists of modules and techniques for the visualization, analysis, and simulation of spatial big data. To achieve these goals, this study conducts an experimental case study with energy usage data for the Seoul Metropolitan Area. Then, the study concludes with future studies and policy measures to promote the use of spatial big data in the territorial and urban planning fields. 정보통신기술의 발전으로 국민과 기업의 각종 활동에 대한 방대한 빅데이터가 실시간 축적되고 있다. 빅데이터는 국민과 기업의 활동에 관한 세부적인 정보를 포괄하고 있어, 국토˙도시 분야에서는 국민체감형 정책을 수립하고 집행하는 데 이를 활용할 수 있다. 따라서 국토˙도시 분야에서의 빅데이터 활용 가능성과 활용방법 등을 살펴보고 이를 통해 국토˙도시 분야의 발전을 도모함과 동시에 공간 빅데이터 분야의 발전방향을 제시할 필요성이 있다. 이와 같은 배경하에 본 논문은 공간 빅데이터의 개념과 범위를 구체화하고, 국토˙도시 및 공간계획 분야에서 이를 보다 구체적으로 활용하기 위한 방안을 모색하고자 한다. 이를 위해 먼저 빅데이터와 공간 빅데이터의 개념을 고찰한 뒤, 계획 분야에서의 빅데이터의 의미와 활용방향을 모색한다. 그리고 공간빅데이터를 활용하기 위한 계획지원체계의 개념적 틀을 정립한 뒤, 이를 기반으로 시각화-분석-시뮬레이션 모형을 개발하고 건물에너지 공간 빅데이터를 활용하여 실험 연구를 수행한다. 그리고 시사점을 도출하여 국토˙도시 및 공간계획 분야에서 공간 빅데이터의 구축을 촉진하고 이를 활용하여 국민이 공감할 수 있는 국토˙도시 정책을 개발하기 위한 향후 과제를 제시하였다.
