Objectives: To circumvent the limitations of the current golden standard method, colony-forming unit (CFU) assay, for viability of Bacille Calmette–Guérin (BCG) vaccines, we developed a new method to rapidly and accurately determine the potency of BCG vaccines. Methods: Based on flow cytometry (FACS) and fluorescein diacetate (FDA) as the most appropriate fluorescent staining reagent, 17 lots of BCG vaccines for percutaneous administration and 5 lots of BCG vaccines for intradermal administration were analyzed in this study. The percentage of viable cells measured by flow cytometry along with the total number of organisms in BCG vaccines, as determined on a cell counter, was used to quantify the number of viable cells. Results: Pearson correlation coefficients of FACS and CFU assays for percutaneous and intradermal BCG vaccines were 0.6962 and 0.7428, respectively, indicating a high correlation. The coefficient of variation value of the FACS assay was less than 7%, which was 11 times lower than that of the CFU assay. Conclusion: This study contributes to the evaluation of new potency test method for FACS-based determination of viable cells in BCG vaccines. Accordingly, quality control of BCG vaccines can be significantly improved.
유아교사의 놀이민감성, 교육과정 운영 자율성 및 반성적 사고와 유아의 놀이몰입 간 구조 관계 분석

This study is to verifiedy the relationship among variables, empirically and integrally, by inquiring into the relative influence of early childhood teachers’ play sensitivity, autonomy in the implementation of early childhood curriculum, and reflective thinking on young children’s play flow, trying to parse the structural relationship as to what mediating role the reflective thinking plays. Four hundred and fifty-six early childhood teachers working at early childhood education institutions, and one child in each class participated in thise study. The results of this study, were as follows: First, early childhood teachers’ play sensitivity and curriculum operation autonomy had a significant impact on reflective thinking, which in turn had a significant impact on early childhood play commitment. Second, early childhood teachers’ play sensitivity and autonomy in implementing the early childhood curriculum appeared to have an indirect effect on young children’s play flow by mediating reflective thinking. The results of this study have educational implications for early childhood play and provide basic data for effective field support by examining the relationship between play sensitivity, curriculum operation autonomy, and reflective thinking, which are teachers’ personal variables that affect early childhood play commitment. 본 연구는 유아 놀이몰입에 영향을 미치는 유아교사의 놀이민감성, 교육과정 운영 자율성, 반성적 사고의 상대적 영향력을 규명하고 반성적 사고의 매개효과와 구조적 관계를 살펴 보고자 하였으며 현재 유아교육기관에 근무하고 있는 유아교사 456명과 각 해당 학급의 유아 1명이 연구대상이었다. 연구결과 첫째, 유아교사의 놀이민감성과 교육과정 운영 자율성은 반성적 사고에 유의한 영향을 미치는 것으로 나타났으며, 반성적 사고는 유아 놀이몰입에 유의한 영향을 미치는 것으로 나타났다. 둘째, 유아교사의 놀이민감성과 교육과정 운영 자율성이 반성적 사고를 매개로 하여 유아 놀이몰입에 간접적인 영향을 미치는 것으로 나타났다. 본 연구는 유아 놀이몰입에 영향을 미치는 교사의 개인변인인 놀이민감성, 교육과정 운영 자율성, 반성적 사고와의 관계를 살펴봄으로써 유아놀이에 대한 교육적 시사점을 제시하고 효과적인 현장지원의 기초자료를 제공하였다.
