Osteogenic effects of exosomes derived from human chorion membrane extracts

Objective: Human chorion membrane extracts (CME) are known to exhibit osteogenic effects when used for treating human osteoblast-like cells (MG63 cells), but the active compound in CME remains unknown. The aim of this study was to identify the presence of exosomes in CME and to determine the osteogenic effect of CME exosomes on MG63 cells. Methods: Exosomes were isolated from human placenta CME using the ExoQuick-TC solution and were characterized. The activity and deposition of alkaline phosphatase (ALP) on MG63 cells cultured with or without exosomes in osteogenic induction medium (OIM) were determined. Human amniotic membrane extracts (AME) were used as controls as they had not affected the osteogenic differentiation of MG63 cells in our previous study. Results: Transmission electron microscopy (TEM) revealed that exosomes isolated from CME and AME (CME-Exo and AME-Exo, respectively) had a cup-shaped structure. NanoSight™ particle tracking analysis (NTA) confirmed that the size of these exosomes was 100–150 nm. In vitro osteogenic experiments demonstrated that the exosomes from CME, but not those from AME, presented increased alkaline phosphatase (ALP) activity and resulted in the mineralization of MG63 cells in a dose-dependent manner. Conclusion: Exosomes were identified in CME and AME from the human placenta. Further, the exosomes from CME were found to be capable of promoting osteogenic differentiation, suggesting that exosomes are a key component of CME that stimulate the osteogenesis of human osteoblast-like cells. CME exosomes can be developed as promising therapeutic candidates for bone regeneration.

Electrodes for electric double layer capacitor were prepared with oxyfluorinated pitch based activated carbon fibers (ACFs) and its properties were also investigated. The oxyfluorination of pitch based ACFs were carried out with F<sub>2</sub>:O<sub>2</sub> ratios of 7:3, 5:5 and 3:7 at 30°C. It was confirmed by SEM images that diameter and morphology of oxyfluorinated pitch based ACFs. The textural and electrochemical properties of oxyfluorinated pitch based ACFs were also evaluated by N<sub>2</sub> adsorption at 77 k and cyclic voltammetry. The cyclic voltammetry of oxyfluorinated pitch based ACFs electrodes were performed in a potential range of 0 - 1 V at different scan rates of 5 and 50 mV/s in 1 M H<sub>2</sub>SO<sub>4</sub> electrolyte.
고정형 유료방송의 서비스 개선방향 도출을 위한 정성적 연구

본 연구는 고정형 유료방송 서비스의 개선과 수요 확대를 위한 시사점 도출에 도움을 주고자, 케이블TV, 위성TV, IPTV 등 유료방송 서비스의 가입자를 대상으로 심층인터뷰와 관찰법이 병행된 ‘민속지학 인터뷰(ethnographic interview)’를 실시하여 서비스 만족․불만족, 개선요구 사항 등에 대해 분석하고 시사점을 도출하고자 하였다. 본 연구는 유료방송 서비스에 대해 통합적으로 비교 분석하였고, 그동안 활성화 되지 않았던 IPTV에 대한 소비자 인식 조사를 시도하였으며, 결합상품 시장의 소비자 인식 조사를 다루었다. 연구 결과, 디지털케이블TV에서는 해약 의사가 거의 없고, 실시간이나 VOD 서비스를 잘 이용하고 있는 것으로 나타났다. 하지만 마케팅 활동이 활성화되지 못하여 소비자들에게 디지털케이블TV의 사용방법 및 인식이 부족하다는 점이 발견되었다. 이의 개선을 위해서는 케이블TV 설치기사가 방문 시 설명과 사용방법을 알려 주는 것이 케이블TV 이미지 쇄신 및 마케팅활동을 위해 필요하다고 생각된다. 또한 대대적인 광고 역시 중요한데, 무엇보다 중요한 것은 고객 만족이기 때문이다. 위성TV는 과거에 비해 가입자 수가 줄어들고 있는 추세이다. 현재 가입자는 약정기간이 종료되면 디지털케이블TV나 IPTV로 전환 가능성이 많은 것으로 조사되었다. 위성TV사업자는 프리미엄 채널 운영 등을 통해 케이블 및 IPTV와의 차별화 전략을 추구해야 할 것이다. IPTV 가입자들은 VOD 이용 요금에 대한 불만족이 나타났다. 케이블, 위성을 비롯한 타 유료방송에 비해 VOD 다운로드 속도, 리모컨 조작의 불편함에 대한 개선이 필요한 것으로 나타났다. This study addressed on the satisfaction, dissatisfaction, and improvement of fixed pay-TV services-cable TV, satellite TV, and IPTV in order to provide the implications for service improvement and demand expansion of pay-TV industry through ethnographic interview. This study focused on entire pay-TV companies' marketing strategies and conducted a in-depth interview of IPTV subscribers. Also, this study did in-depth interviews about consumers' perceptions in convergence market. Finally, we found several implications as followed. In the digital cable TV, subscribers make full use of TV such as watching live channels, or VoD. Although they are willing to watch digital cable TV continuously, they aren't handy with remote controls because of lack of efficient and customized marketing strategies. Thus, the engineers who visit the subscribers' home in order to set cable TV or set-top box need to show the effective way to control TV remote controls with full explanation. It's good to improve and renew the brand images of digital cable TV for subscribers. Also, extensive advertisements are needed, because customer satisfaction is of greatest importance. The subscribers of satellite TV are decreased. They want to change the platform from satellite to cable TV or IPTV when their contracts run out. The satellite company needs to fortify the positioning of premium content channel for the differentiation from cable TV or IPTV. The subscribers of IPTV are discontented with the cost of VoD. In comparison with other media such as cable TV or satellite TV, IPTV needs to upgrade their service including low speed to download VoD, complicated remote controls to deal with.
