To improve the identification power of TLC/HPTLC the in situ reflectance spectra obtained directly from plates with commercial scanner are used. The spectrum normalization should be carried out prior to comparing and searching the spectra from library for the identification of compounds. Because the reflectance does not obey the Lambert-Beer''s law, there arise some problems in normalization. These problems could be solved to some extent by normalizing the spectra with regression methods. The spectra are manipulated with the regression function of a curve obtained from the correlation plot. When the parabola was used as the manipulating function, the spectra were identified with the accuracy of 97% and this result was better than that of conventionally used the point and area normalization method.

Methanol extracts of 94 Korean plants were screened for angiogenesis inhibitors using the tube-like formation assay of HUVEC (Human Umbilical Vein Endothelial Cell) and evaluated for growth inhibitory activity on A549 cells, human lung cancer cells. Extracts of Euphorbia sieboldiana, Adonis amurensis, and Anthriscus sylvestris showed antiangiogenic and growth inhibitory activity at $50\;{mu}g/ml$. Aristolochia manshuriens and Styrax obassia expressed antiangiogenic activity without growth inhibitory action.

The enantiomers of five amino acids (alanine, valine, threonine, leucine and phenylalanine) could be separated by gas chromatography with optically active (S)-5-isopropyl-N3-phenyl-2-thiohydantoinic stationary phase, which prepared from L-valine and phenylisothiocyanate. Gas chromatographic separations on methylesterificated and N-trifluoroacetylated amino acids have been conducted in isothermal at several column temperatures (180~190, 200, 210oC). The separation factors were 1.29 (alanine, 190oC), 1.35 (valine, 190oC), 1.33 (threonine, 190oC), 1.17 (leucine, 190oC) and 1.05 (phenylalanine, 190oC) and D-isomers eluted prior to L-isomers in every instance. The result of this experiment shows that this stationary phase can be used for the separation of the other amino acids enantiomers.

Quantitative analysis of (+)-catechin (C), (-)-epigallocatechin (EGC), (-)-epicatechin (EC), (-)-epigallocatechin gallate (EGCG), (-)-epicatechin gallate (ECG) and caffeine in commercial green tea was carried out by HPLC employing gradient elution of 0.1% acetic acid and acetonitrile on ODS column. The optimized HPLC method provided satisfactory linearity, accuracy and precision. The relationship between the concentration of the components and cultivated region of the commercial green tea was not significant, while the concentration of EGCG, ECG and caffeine decreased significantly in the later harvested green tea samples (p<0.01). Multivariate analysis of the components was performed in order to characterize and evaluate the cultivated region and harvest period-related variation. Hierarchical clustering and discriminant analysis were applied to classify the geographical and seasonal origins of the green tea samples. The classification accuracy of the cultivated region and harvest period by discriminant analysis was 95% and 91%, respectively, indicating that this method could be reliable and convenient for the quality control of herbal products with different origin.
전탕용수의 미네랄 함량이 두충의 유효성분 추출에 미치는 영향 분석

Objective : Eucommiae Cortex is bark of Eucommia ulmoides Oliver, a deciduous tree native to China. Today, teas or herbal drugs made from E. ulmoides leaves are widely consumed as a healthy food especially in Eastern Asia including Japan, China and Korea. This study was aimed to verify the extraction efficiency of bioactive compounds from Eucommiae Cortex depending on characteristics of decoction water. Method : Six types of decoction water with various origin were prepared and the contents of minerals in water were analyzed by ICP-AES and ICP-MS. The contents of extracted bioactive compounds were analyzed by HPLC and the relationship between mineral contents and extracted bioactive compounds in the decoction water were analysed by multiple factor analysis. The decoction waters could be classified in three groups by hierarchical cluster analysis. Result : Extracted amounts of compounds in Eucommiae Cortex showed a positive relationship with Na, Si, K concentration in decoction water, while a negative relationship with Ca, Mg concentration. Conclusion : The study proved that the minerals in water influenced the extraction efficiency of bioactive compounds from Eucommiae Cortex.
