        Effect of Nitrogen Deficiency on Recombinant Protein Production and Dimerization and Growth in Transgenic Plants

        This study investigated the effects of nitrogen (NH4+) in the soil on the expression of prostatic acid phosphatase (PAP)-IgA Fc and PAP-IgA FcK recombinant proteins in transgenic plants, and on plant growth in the greenhouse. Plant height and leaf length were greater in soil A (NH4+: 51 mg·L-1) than in soil B (NH4+: 1 mg·L-1). In soil B, the plant leaves turned yellow, with chlorophyll concentrations that were significantly lower than the chlorophyll concentration of leaves from plants grown in soil A. The expression level of the PAP-IgA Fc and PAP-IgA FcK proteins and the protein dimerization rates were lower in soil B when compared with soil A. In addition, PAP-IgA Fc and PAP-IgA FcK expression and dimerization levels were significantly lower in transgenic plants grown in soil B, when compared with transgenic plants grown in soil A. Soil characteristics such as hydrogen ion concentration (pH), electric conductivity (EC), and bulk density did not differ between soil A and B. Thus, appropriate soil NH4+ concentrations are essential for optimized plant growth and enhanced expression of dimerized PAP-IgA Fc and PAP-IgA FcK recombinant proteins in transgenic plants.

        피부판을 이용한 비결손의 재건

        The nose is the most prominent visible and central fearure of the human face. Its size, shape and configuration, therefore, play a significant role in the determination of man's appearance. Real or imaged deformity of the nose is highly significant in the indivisual's conception of his own self image. Also its position make it vulnerable to trauma. The degree of loss and deformity usually dictate the type and quantity of tissue required for correction and for the most satisfactory reconstruction, the surgeon must consider all of the alternatives to reach the end point with the fewest number of the procedures in the most predictable manner and leave the least amount of donor deformity. Of course, the tissues closest to the nose will give the best result with most natural appearence in color and texture. We had experienced 7 cases of nasal reconstruction: using midline forehead flap for the defect of nasal tip, midline island forehead flap with a subcutaneous pedicle for the defect of cover of the nose, midline forehead flap and scalping flap for subtoral defect of the nose and nasolabial flap for the defect of ala nasi. So we could report them with reviews of the relating references.

        Immune response of heterologous recombinant antigenic protein of viral hemorrhagic septicemia virus (VHSV) in mice

        Viral hemorrhagic septicemia (VHS) is an important infectious disease in fish worldwide caused by viral hemorrhagic septicemia virus (VHSV). VHSV is the causative agent of serious systemic diseases in fish, affecting a number of teleost fish species. In this study, VHSV glycoprotein (G), including its epitope, as a subunit vaccine candidate, was expressed in tobacco plant (Nicotiana tabacum). The recombinant gene, VHSVG, was fused to the immunoglobulin Fc fragment and extended with the endoplasmic reticulum (ER) retention signal (KDEL) to generate VHSVG-FcK. The recombinant expression vector for VHSVG-FcK was transferred into Agrobacterium tumefaciens (LBA4404), and plant transformation was conducted N. tabacum. Polymerase chain reaction (PCR) was performed to confirm gene insertion and VHSVG-FcK protein expression was confirmed by immunoblot analysis. VHSVG-FcK protein was successfully purified from tobacco plant leaves. Furthermore, ELISA analysis showed that mice serum immunized with the plant-derived VHSVG-FcK (VHSVGPFcK) had a high absorbance against VHSVG-FcK, indicating that the plant-derived recombinant subunit vaccine protein VHSVG-FcK can induce immune response. Taken together, this recombinant vaccine protein can be expressed in plant expression systems and can be appropriately assembled to be functional in immunogenicity.

        수부 구축성 화상반흔 환자에서 술후 관절운동범위의 개선에 관한 임상적 고찰

        Burns of the hand are common in patients with thermal injury. But, because the treatment of burn is usually directed toward the life saving and due to the shortage of the knowledge about the postburn deformities, many patients with hand burn are found after development of severe functional impairment, In department of plastic surgery of Inje College, Baik Hospital, 59 cases of burn scar contractured hands were treated with free skin graft and aggressive physiotherapy from January of 1984 to December of 1986. The results are follow; 1) Among the 59 cases of burn contracted hands, 48 cases (81%) were excellent in results. 2) The other 11 cases of fair or poor end results were mainly due to the third degree burn and inadequate splinting after split thickness skin grafting. 3) Age of the patient had close relation to the postoperative functional recovery, the younger the better. 4) The postoperative end results in contracture after electric or flame burn were worse than in contracture after the others. 5) The results of full thickness skin graft were better than that of split thickness skin graft. 6) The time duration from burn injury to skin grafting has an important effect on postoperative end results, the longer the worse. 7) The degree of preoperative limitation of motion has close relationship with postoperative end results, the severer limitation of motion, the worse end results.

        Expression and in vitro function of anti-cancer mAbs in transgenic Arabidopsis thaliana

        The anti-colorectal cancer monoclonal antibody CO17-1A (mAb CO), which recognizes the tumor-associated antigen EpCAM, was expressed in transgenic Arabidopsis plants. PCR and western blot analyses showed the insertion and expression of heavy chain (HC)/HC fused to the KDEL ER retention modif (HCK) and light chain (LC) of mAb CO and mAb CO with HCK (mAb COK) in Arabidopsis transformants. Both plantderived mAbP CO and mAbP COK were purified from a biomass of approximately 1,000 seedlings grown in a greenhouse. In sandwich ELISA, both mAbP CO showed a slightly higher binding affinity for the target, EpCAM, compared to mAbM CO. In cell ELISA, both mAbsP COs showed binding affinity to the human colorectal cancer cell line SW480. Furthermore, mAbM CO, mAbP CO, and mAbP COK exhibited dose and timedependent regression effects on SW480 cells in vitro. In summation, both mAbP CO and mAbP COK, expressed in Arabidopsis, recognized the target antigen EpCAM and showed anti-proliferative activity against human colorectal cancer cells.
