        태반형 알칼라인 포스파타아제 유전형에 대한 단일클론 항체

        A total of nine monoclonal antibodies (MAbs) against common phonotypes of human placental alkaline phosphatase (PLAP) was produced by hybridoma using a mixture of PLAP subtypes (type 1+2+3) as antigen. Six of these reacted only with PLAP, but not with intestinal and liver alkaline phosphatases. Three out of these six MAbs have been found to discriminate at least one of the product of three common alleles from others, while the other three MAbs reacted with the six common allelic varients. The results suggest that anti-PLAP MAbs can be used as probes to discriminate among allelic varients of PLAP.

        Regulation of Early Steps of Chondrogenesis in the Developing Limb

        In the developing limb, chondrogenesis is animportant prerequisite for the formation of cartilage whosetemplate is required for bone formation. Chondrogenesis isa tightly regulated multi-step process, including mesenchymalcell recruitment/migration, prechondrogenic condensation ofthe mesenchymal cells, commitment to the chondrogeniclineage, and differentiation into chondrocytes. This processis controlled exquisitely by cellular interactions with thesurrounding matrix and regulating factors that initiate orspecific genes in a temporal-spatial manner. Understandingthe cellular and molecular mechanisms of chondrogenesisis important not only in the context of establishing basicprinciple of developmental biology but also in providingresearch direction toward preventive and/or regenerativemedicine. Here, I wil overview the current understanding ofcellular and molecular mechanisms contributing toprechondrogenic condensation processes, the crucial stepsinteractions.

        Study on the Viability of a Midsize Hybrid Electric Aircraft via Initial Sizing Methodology

        Electric propulsion is an environment-friendly technology. However, it has been more frequently applied to small aircraft because electric energy sources have lower specific power and energy compared with conventional fossil fuels. To examine the viability of electric propulsion for large aircraft, we assessed the advantages and disadvantages of a midsize hybrid electric aircraft in terms of the fuel savings and weight increase. In this study, we employed a parallel hybrid electric propulsion system that consists of a turboprop engine and an electric motor that runs on fossil fuel and liquid hydrogen fuel cells, respectively. With the hybrid electric propulsion system, we carried out an initial sizing study using an ATR 72 nominal midsize aircraft to evaluate the fuel savings and weight increase compared with the original ATR 72 with a turboprop. For the initial sizing of the hybrid electric ATR 72, the total power of the hybrid electric propulsion system was optimally distributed between the turboprop engine and electric motor to achieve maximum fuel savings with minimum weight increase. The results showed that the hybrid electric ATR 72 was barely viable with current technology. The hybrid electric ATR 72 could be more practical with the advancement of fuel cell technology.

        하이브리도마 방법에 의한 항 α - Proteinase Inhibitor 모노클론항체의 생산과 분석

        Monoclonal antibodies (MAbs) against human α₁-PI was produced by hybridizing SP 2/0 -Ag 14 mouse myeloma cells with spleen cells of Balb/c mice immunized with a α₁-PI. The resulting hybridoma clones were screened for their ability to bind α₁-PI by ELISA. Seven positive stable monoclones were obtained by cloning three times in serial dilutions, and each antibody was identified to be homogeneous by several criterial. By western bloting analysis using CNBr-cleavage peptide fragments (Fr I: amino acid residues 64-220; Fr II: 243-351; Fr III : 1-63), four of these monoclones were found to synthesize MAb specific to Fr I, and one clone to Fr II. But MAbs of the other two clones showed reactivity only to intact α₁-PI. Five of our monoclones were appeared to synthesize IgG₁(k) antibodies having affinity constant in the range 0.1-1.1×10^9 M^(-1)

      • Production and Characterization of Monoclonal Antibodies against Human alpha-1-Proteinase Inhibitor by Hybridoma

        Alpha-1-proteinase inhibitor(${\alpha}_1-PI$)로 면역시킨 Balb/c 마우스 비장세포와 SP 2/0-Ag 14 마우스 미에로마 세포를 융합시키는 하이브리도마 기법으로 모노클론 항체 생산을 실시하였다. 하이브리드클론들이 생산하는 항체들과 ${\alpha}_1-PI$와의 반응성은 ELISA로 분석하였으며, 양성클론을 제한희석법으로 클로닝하여 7클론의 항-${\alpha}_1-PI$ 특이 클론세포를 얻었고, 이들의 단일클론성을 확인하였다. 7클론 중 5클론이 생산하는 모노클론항체는 항원에 대한 결합상수가 $0.1-1.1{\times}10^9M^{-1}$의 $IgG_1(k$) 이었고, 이들 항체의 항원 특이성을 ${\alpha}_1-PI$의 CNBr-peptide 분획을 이용한 Western blot를 실시한 결과 FrI(아미노산 잔기 64-220)에 특이한 것이 4클론, Fr II (243-351)에 특이한 것이 1클론이었고, 나머지 2클론의 특이성은 결정할 수 없었다.       • Phosphatidylcholine 의 분해에 의한 HeLa 세포와 Gelatin 기질과의 상호작용의 유도

        HeLa cells, a transformed human epithelial cell line, attach to various substrata but subsequent spreading is specific to collagen or gelatin. The spreading is initiated by the activation of phospholipase A₂ (PLA₂) which produces arachidonic acid (AA) as a consequence of cell surface collagen receptor clustering. This study examines the mechanism of PLA₂ activation and which phospholipids are hydrolyzed by PLA₂ to release AA in response to HeLa cell adhesion to a gelatin substratum. The levels of phosphatidylcholine decreases, among various phospholipids, during attachment and spreading of HeLa cells. Lysophosphatidylcholine is the only lysophospholipids formed during HeLa cell adhesion indicating that clustered collagen receptors activate PLA₂ to hydrolyze posphatidylcholine to AA and lysophosphatidylcholine. Among various molecular entities which are known to regulate PLA₂ activation, we have previously shown that PLA₂ activation is not mediated by either changes in Ca^(2+) levels, alkalinization of cytoplasmic pH, or activation of protein kinase C. It is also likely that PLA₂ activation is not mediated by either pertussis or cholera toxinsensitive G proteins as those toxins do not affect both AA release and cell spreading.

        포병 표적탐지 레이더 운용의 계량적 효과 분석

        In the future warfare, the importance of the counter-fire operation is increasing. The counter-fire operation is divided into offensive counter-fire operation and defensive counter-fire operation. Reviewing the researches done so far, the detection asset of offensive counter-fire operation called UAV(Unmanned Aerial Vehicle) and its operational effectiveness analysis is continually progressing. However, the analysis of the detection asset of defensive counterfire called Target Acquisition Radar(TAR) and its quantitative operational effectiveness are not studied yet. Therefore, in this paper, we studied operational effectiveness of TAR using C2 Theory & MANA Simulation model, and showed clear quantitative analysis results by comparing both cases of using TAR and not using TAR.

      • 흡착 크로마토그라피법에 의한 사람 Alpha-Fetoprotein 의 분리

        For the preliminary step to make and characterize the monoclonal antibodies of human alpha-fetoprotein (AFP) was purified from 534 g of human fetal tissues through the procedures of tissue extraction, DEAE-cellulose, concanavalin A-Sepharose, Cibacron Blue F3GA-agarose and immunoadsorbent affinity chromatography. The isolated AFP preparation showed a single band on polyacrylamide gel electrophoresis and a single precipitin arc against rabbit anti-human cord serum and anti-human AFP on immunoelectrophoresis. Our AFP also displayed a single band on SDS-polyacrylamide gel electrophoresis. The recovery of AFP was 8.76㎎ total.
