Two cases of lingual masses were observed among 500 Fischer 344 (F344) rats which were used as control and treated animals in a 2 year carcinogenicity study in Toxicology Research Center, Korea Research Institute of Chemical Technology. The masses grossly appeared as tan, pedunculated, fungiform on the dorsal aspect of the base of the tongues. They were approximately $1.5{\times}1.2{\times}0.3cm$ in size. The microscopic features consisted of acanthosis, hyperkeratosis, papillary projection with connective tissue cores and multifocal chronic active inflammation with hair shafts. The results observed support the epigenetic mechanism of tumorigenesis which is caused by Physical stimuli of foreign bodies. Both of the masses were diagnosed as papillomas with the incidence rate of 0.4%(1/250) in each sex on the basis of the gross and microscopic features.

We present background data of Sprague-Dawley (SD) rats from the control groups of 2 weeks and 4 weeks repeated-dose toxicity studies performed in our laboratory during 2002-2003. These include hematology, blood biochemistry and organ weights. These data can be used as background data for the interpretation of data from repeated-dose toxicity studies of drugs or chemicals in SD rats.

This experiment was carried out to study the toxic effect of solublized methylcellulose (MC). Sprague-Dawley rats were dosed with 1%(w/v) MC in 0.9% saline by gavage at a dose of 10ml/kg b.w/day or by intravenous injection at a dose of 5ml/kg b.w/day for 28 days. Clinical signs were observed once a day. Body weights, water and food consumptions were measured and urinalysis was performed several times during the experiment. Rats were sacrificed on days 3, 7, 15 and 28 for hematology, blood chemistry, organ weights and histopathology. The relative weight of the spleen and foamy cells of the spleen were increased in the gavage group. Body weight gain, food consumptions, the values of RBC, Hb, MCH, Hct, serum proteins, glucose, bilirubin, AST, and ALP were decreased in I.V. treatment group. On the other hand, water consumptions, the values of serum cholesterol, creatinine, and BUN were increased. Microscopic findings were granulomas, distended sinusoids, and hypertrophy of Kupffer cells with vacuoles in the liver. Spleen exhibited granuloma, increased extramedullary hematopoiesis, and congestion. Kidney exhibited foamy cells in the glomeruli, distension of the tubules. The findings appeared more severe when the treatment was extended. In conclusion, MC solution is not a safe vehicle for intravenous administration because of the toxic effects on the liver, kidney and spleen. In addition, a long-term and large dosage of oral administration of MC appears to be unsafe also and needs to be investigated further.

Establishment of the basic data line is very important in the evaluation of the result of clinical and/or nonclinical experiments, in the improvement the confidence of test results. A reference range data base containing serum chemistry and hematology values in 5 weeks old BALB/c and C57BL/6 mice which were purchased from the different companies is described. Hematological parameters are white blood cell count, red blood cell count, hemoglobin concentration, hematocrit, mean corpuscular volume, mean corpuscular hemoglobin, mean corpuscular hemoglobin concentration, platelet count. In the BALB/c mice, there are significant differences observed between lots, sexes and strains. In the C57BL/6 mice, there are significant differences observed between lots and strains but no significant differences observed between sexes. Serum biochemistry parameters are aspartate aminotransferase, alanine aminotransferase, alkaline phosphatase, blood urea nitrogen, creatinine, glucose, total cholesterol, total bilirubin, total protein, albumin, triglyceride, calcium, inorganic phosphate, phospholipid, high density lipoprotein cholesterol, low density lipoprotein cholesterol, albumin globulin ratio, sodium, potassium, chloride. In both stains of mice, there are significant differences observed between lots, sexes and strains. Data from the present study will support in the proper interpretation and validation of these results.

A reference range data base containing organ weight values on animals used in 4 week and 13 week toxicity tests is described. Data listed include the values of minimum, maximum, mean and standard deviation for each of the following organ weights. Organs: Briain, heart, liver, spleen, kidney, adrerial gland, testis, ovary. This study was undertaken to determine organ weight reference values of Ktc: SD rats.

Two Beagle dogs were examined by ultrasonography to get detailed informations about the normal size and texture of abdominal organs. Ultrasonographic examination of liver, gallbladder, stomach, duodenum, spleen, kidney and urinary bladder was performed on the abdomen in dorsal recumbency. The ultrasonographic values in this study can be used as reference for the diagnosis of changes in abdominal organs of Beagle dog.
