        어성초 전탕액이 흰쥐 장기내 카드뮴 축적에 미치는 영향(I)

        This study was conducted to investigate the antitoxic component in aqueous extract of Houttuynia cordata $T_{HUNB}$. The results were as follows: Generally, detoxication effects by Houttuynia cordata $T_{HUNB}$ extract increased in proportion to the extract concentrations. When 40mg/kg dosage of Houttuynia cordata $T_{HUNB}$ extract was administered, it showed the highest antitoxic effects in metallothionein induction. After the extract treatment, body weights increased in proportion to the extract concentration. However, after 3 weeks, the body weight decreased insignificantly. From the above results, Houttuynia cordata $T_{HUNB}$ extract increased metallothionein concentration and decreased the toxicity of cadmium in rats. 흰쥐에게 카드뮴 경구투여시, 어성초 전탕액의 경구 투여한 후 카드뮴의 독성에 대한 해독경감효과에 대하여 다음과 같은 결과를 얻었다. 1. 흰쥐에게 어성초 전탕액의 경구 투여 용량이 증가할수록 카드뮴의 독성에 대한 해독경감효과는 증가하였으며, 신장에서보다 간장에서 해독경감효과가 좋았다. 2. 흰쥐 신장 및 간장의 MT의 농도는 간장에서 보다 신장에서 우수한 결과를 보였으며, 신장에서는 어성초 전탕액의 경구 투여 농도가 10mg/kg 이상이 되면 MT 형성은 어성초 전탕액의 투여 농도가 증가하여도 크게 증가하지는 않았다. 3. 흰쥐의 체중변화는 어성초 전탕액의 경구 투여 용량이 증가할수록 흰쥐의 체중변화도 증가하는 경향을 보였으나, 3주 이후에는 체중의 증가가 적게 나타났다. 이상과 같이 어성초 전탕액의 경구투여를 종합해 보면, 어성초 전탕액의 경구 투여 시켰을 경우 카드뮴의 독성에 대한 해독경감효과가 있는 것으로 판단된다.

        In the present study, we have evaluated cytotoxic effects of Taraxaci herba extract on human skin melanoma cells. The light microscopic study showed morphological changes of the treated cells. Disruptions in cell organelles were determined by calorimetric methods: MTT (3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazoliumbromide), NR (Neutral red) and SRB (Sulforhodamine B protein) assay. These results suggest that Taraxaci herba retains a potential antitumor activity.

        L1210 및 $P388D_1$ 세포에 대한 고삼 에틸 아세테이트 추출물의 세포독성에 관한 연구 (III)

        This study was carried out to evaluate cytotoxic effects of the roots of Sophora flavescens Ait. extracts on murine leukemia tumor cells lines $(P388D_1\;and\;L1210)$. Disruptions in cell organelles were determined by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazolium bromide (MTT) assay. The comparison of $IC_{50}$ values of the ethyl acetate of Sophora flavescens Ait. extract in leukemia cell lines showed that their susceptibility to these extracts decreased in the following order : Adriamycin>Fr.4>Fr.5>Fr.3>Fr.1>Fr.2 by the MTT assay. These results suggest that the fraction 4 of the ethyl acetate soluble extract of Sophora flavescens Ait. may be a valuable choice for the studies on the treatment of murine leukemia cell lines.

        This study was carried out to evaluate cytotoxicity of the extracts from Sophora flavescens Ait. against L1210 (lymphocytic leukemia) and $P388D_1$ (lymphoid neoplasms) cells in vitro. We have determined cytotoxicity by MTT {3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazo-liumbromide} assay. The order of cytotoxicity of Sophora flavescens Ait. extracts against L1210 and $P388D_1$ cells in vitro is as follows: AM> EASF > CFSF > MTSF > WSF > HXSF and AM> EASF> CFSF> MTSF> HXSF> WSF. These results suggest that the ethyl acetate soluble extract of Sophora flavescens Ait may be a valuable choice for the development of antitumor agents.

        조개나물 추출물의 세포독성과 항균효과

        This study was carried out to evaluate cytotoxic effects of Ajuga multiflora Bunge extracts on murine leukemia tumor $(P388D_1)$ cell lines. Disruptions in cell organelles were determined by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazoliumbromide (MTT) assay. The comparison of $IC_{50}$ values of Ajuga multiflora Bunge extracts in L1210 and $P388D_1$ cell lines showed that their susceptibility to these extracts decreased in the following order: Adriamycin>methanol extract>chloroform extract>ethyl acetate extract>hexane extract>water extract by the MTT assay. In order to develop an antimicrobial agent, dried Ajuga multiflora Bunge was extracted with several solvents, and then antimicrobial activity was investigated. The minimal inhibitory concentration (MIC) of the extract against microorganisms were also examined. Antimicrobial activities of amocla and ketoconazole as references were compared to those of extracts of $H_2O$, n-hexane, chloroform, ethyl acetate and methanol. The antimicrobial activity of all extracts from the sample had growth inhibition activity against gram-negative bacteria, gram-positive bacteria and fungi $(MIC>200\;{\mu}g/ml)$. These results suggest that the methanol soluble extract of Ajuga multiflora Bunge may be a valuable choice for the studies on the treatment of murine leukemia tumor cell lines.

        한국산 생약으로부터 항암물질의 개발 (제13보). -농길리 추출물의 세포독성 및 항암작용에 관한 연구-

        The cytotoxic and antitumor activity of Herba cratalariae sessiliflorae on cultured NIH 3T3 fibroblast and human oral epitheloid carcinoma cells were evaluated by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazoliumbromide (MTT) colorimetric method. The light microscopic study was carried out to observe morphological changes of cultured mouse fibroblast and human oral epitheloid carcinoma cells (KB). These results were obtained as follows; Ethyl acetate, chloroform and hexane extracts showed a significant cytotoxicity in NIH 3T3 fibroblast, but the other extracts did not show. All extracts exhibited a significant antitumor activity in human oral epitheloid carcinoma cells, but ethanol extract did not show a antitumor activity. Hexane extract showed low cytotoxic effect, but exhibited the most antitumor activity. The MTT absorbance in NIH 3T3 fibroblast was significantly decreased by treatment with chloroform, ethyl acetate and hexane extracts respectively. Human oral epitheloid carcinoma cells was significantly decreasd by treatment with all extracts with the exception of ethanol extract. The difference in MTT absorbance in two cell Types was most remarkable when treated with water and hexane extracts. Cholroform and hexane extracts showed the strongest effect in growth inhibition of human oral epitheloid carcinoma cells. These results indicated that water extract possessed no cytotoxicity and a strong antitumor activity.

        팔선 양조용수의 세포독성 및 항암활성

        In the present study, we have evaluated cytotoxic effects, antitumor activities and metastasis inhibitory effects of Palsun brewing water NIH 3T3 cells, human oral epitheloid carcinoma cells, and human skin melanoma cells. The light microscopic study showed morphological changes, AG-NOR (argyrophylic nucleolar organizer region) by silver chloride stain, and glycoprotein by PAS (periodic acid stain) reaction of the treated cells. Disruptions in cell organelles were deteminied by colorimetric methods;MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazoliumbromide) and SRB (sulforhodamine B protein) assays. These results suggest that Palsun Brewing Water retains no cytotoxic effects in NIH 3T3 cells and a growth-inhibitory activity in cancer cell lines.

        고삼추출물의 항균효과 (Ⅰ)

        In order to develop a antimicrobial agent, dried Sophora flavescens Ait. was extracted with several solvents, and then antimicrobial activity was investigated. The minimal inhibitory concentration(MIC) of the extracted substance against microorganisms were also examined. Antimicrobial activity of the initial methanol extract from the sample was the strongest compared to those of other solvent extracts such as H2O, n-hexane, chloroform, ethyl acetate and methanol. The methanol extract had strong growth inhibition activity against gram-postive bacteria (MIC, 25~50mcg/ml) such as B. subtilis and S. aureus. Among gram positive bacteria tested, B. subitlis was the most susceptible to the extracted substance. The antimicrobial activity of the methanol extract from the sample had strong growth inhibition activity against gram-negative bacteria such as P. aeruginosa (MIC, 25mcg/ml).

      • 고삼 메탄올 추출물의 향균효과에 관한 연구

        고삼(Sophora flavescens Ait)뿌리의 메탄올 추출물을 C_18 frash chromatography fraction중 항균력을 타진하기 위한 방법의 일환으로, 각종 미생물에 대한 균종별 항균력을 비교, 검토한 결과를 요약하면 다음과 같다. 1. 그림 양성균에 대해서 fraction 1,5,6,8에서 50㎍/㎖, 25㎍/㎖, 12.5㎍/㎖, 6.25㎍/㎖이상으로 항균효과를 나타냈으며, 특히 fraction6은 S. aureus 6538P와 B. subtilis에서 최소억제농도6.25㎍/㎖로 가장 강한 항균효과를 보여주었다. 2. 고삼 메탄올 추출물 C_18 frash chromatography fraction 1,5,6,8은 E. coli에서 200㎍/㎖이상으로 항균효과를 나타냈으나, P. aeruginosa에 대해서는 50㎍/㎖, 25㎍/㎖, 12.5㎍/㎖, 6.25㎍/㎖이상 항균효과가 나타났으며, 이중 fraction 6에서 6.25㎍/㎖로 가장 강한 항균효과를 보여주었다. 3. 고삼 메탄올 추출물 C_18 frash chromatography fraction의 진균에 대한 최소억제농도는 200㎍/㎖이상으로 약한 항균효과를 나타내었다. 4. 고삼 뿌리의 메탄올 추출물을 C_18 frash chromatography fraction중 Fr. 5는 12.5㎍/㎖과 6.25㎍/㎖로 가장 강한 항균물질이 포함되어있다고 사료된다. The minmum inhibitory concentration (MIC) the of extract was the strongest compared to those of other fractions. The fraction 6 had strong growth inhibitory activity against gram-positive bacteria (MIC,6.25~12.5㎍/㎖)such as S. aureus, B. subtilis and M.luteus. Among gram-positive bacteria tested S. aureus and B. subtklis were the most susceptible to the extracted substance. The antimicrobial activity of the fraction 6 from the hot methanolic extract had strong growth inhibition activity gram-negative bacteria such P.aeruginosa(MIC, 6.25㎍/㎖).

      • L1210 및 P388D_1에 대한 고삼 메탄올 추출물의 세포독성에 관한 연구

        고삼(Sophora flavescens Ait. S. angustifola S. et Z)뿌리의 메탄올 추출물을 C_18 frash chromatography fraction중 MTT정량분석법으로, 마우스 백혈병 세포인 L1210 및 P388D_1 세포에 대하여 세포독성효과를 평가하였다. 이들 추출물은 마이크로로 그람 농도의 범위에 대하여 세포독성을 나타내었으며, 이들 추출물에 대한 세포독성은 Table 1에 나타난 바와 같이 Fr. 5 추출물이 가장 강한 세포독성을 보였다. 즉 L1210 암세포에 대하여 메탄올 C_18 frash chromatography fraction중 IC_50값은 AM>Fr. 5>Fr. 4>Fr. 10>Fr. 6>Fr. 8>Fr. 2>Fr. 7>Fr. 9>Fr. 3>Fr. 1로 Fr. 5에서 가장 강한 세포독성을 보였으며, P388D_1 암세포에 대한 세포독성은 실험결과에서도 Fr. 5 추출물이 가장 강한 세포독성을 보였으며, 다음과 같은 순서로 세포독성이 감소하였다. MTT정량분석법에 의하면, AM>Fr. 5>Fr. 4>Fr. 6>Fr. 7>Fr. 10>Fr. 8>Fr. 3>Fr. 2>Fr. 1>Fr. 9 정량분석값을 얻을 수 있었다. 결론적으로 고삼 메탄 추출물 C_18 frash chromatography fraction에서, Fr. 5에 마우스의 백혈병 세포인 L1210 및 P388D_1 암세포에 대한 가장 강한 항암활성을 나타내었다. 따라서 Fr. 5 추출물에 보다 항암활성이 높은 화합물이 함유되어 있을 것으로 사료된다.
