PCR과 SDS-PAGE를 이용한 Bifidobacteria의 동정

SDS-PAGE를 이용한 단백질 패턴의 분석결과, 같은 종의 Bifidobacterium은 동일한 단백질 패턴을 보이며, 다른 종의 Bifidobacterium은 서로 다른 패턴을 보이고 있는 것을 확인하였다. 따라서 SDS-PAGE를 이용한 whole cell protein 패턴 분석으로 Bifidobacterium을 간편하고 빠르게 동정할 수 있는 가능성을 보였다. DNA homology 98% 이상으로 PCR에 의해 동정에 어려움이 많았던 B. catenulatum, B. Pseudocatenulatum group, B. animalis을 SDS-PAGE로 확인한 결과, B. catenulatum과 B. pseudocatenulatum, B. animalis는 뚜렷한 단백질 패턴의 차이를 보임으로써 구별이 가능했다. PCR 수행 시 primer제작은 기존에 보고된 것과 NCBI blast program을 통하여 제작을 하였으며, 그 결과 B. longum, B. bifidum, B. adolescentis 3종류의 종에서만 특이적으로 band가 나타나는 것을 확인할 수가 있었다. 본 실험에서는 SDS-PAGE와 PCR을 이용한 균주 동정법으로 신속하고 정확하게 Bifidobacterium을 탐색할 수 있었으며, 발효유 제조나 의약품 제조 등 산업적으로 이용 될 수 있을 것이다 (Mitsuoka, 1984). Bifidobacterium is a Gram-positive obligate anaerobic bacterium which is able to change cellular shape by nutritional conditions and dominant species in intestinal microflora of human. In order to identify Bifidobacteria spp. in human intestine, PCR and SDS-PAGE of whole cell proteins were used. Species-specific primer sets based on the 16S rRNA, 16S-23S intergenic spacer and heat shock protein genes of three Bifidobacteria spp. were constructed using NCBI blast program (Bifidobacteria adolescentis, B. longum and B. bifidum). PCR products using each primer sets showed the same sizes of expected target genes. From the SDS-PAGE of whole cell proteins in bifidobacteria, profiles of SDS-PAGE between 13 type strains showed distinct differences. These methods will be useful for monitoring and evaluating Bifidobacteria spp. in human intestine microbial population.
Is the shock index a useful tool in trauma patients with alcohol ingestion?

Objective: Alcohol consumption is a frequent risk factor for trauma. The shock index is widely used to predict the prognosis of trauma, and alcohol can influence the shock index in several ways. This study investigated the usefulness of the shock index in trauma patients who had ingested alcohol. Methods: This was a retrospective, observational, single-center study. We performed a logistic regression analysis to assess the association between alcohol consumption and massive transfusions. A receiver operating characteristic (ROC) curve was constructed to determine the predictive value of the shock index for patients who had ingested alcohol. Results: A total of 5,128 patients were included in the study. The alcohol-positive group had lower systolic blood pressure and higher heart rate; consequently, the shock index in this group was higher. There was no significant difference between the proportion of the alcohol-positive and alcohol-negative groups who underwent massive transfusions and suffered hospital mortality compared to the overall proportion of patients who underwent massive transfusion based on the shock index. In the logistic regression analysis, the alcohol-negative group showed higher odds ratios for massive transfusions compared to the alcohol-positive group. The area under the ROC curve for predicting massive transfusion was 0.831 for the alcohol-positive group and 0.825 for the alcohol-negative group. However, when a cutoff value of 1 was used, the false positive rate was significantly higher in the alcohol-positive group. Conclusion: The shock index is a useful tool for predicting outcomes in patients with trauma. However, in patients who have ingested alcohol, the shock index should be interpreted with caution.

식중독 병원성 미생물은 전 세계적으로 많은 질병을 발생시카고 있기 때문에 전염병, 생화학적 테러방지,식품 산업률 그리고 건강유지를 위해서 빠르고 신속한 검출이 필요하다병원성균을 빠르게 검출하기 위해서는 이러한 미생물의 종 특이적인 유전자와 프로브를 발굴해야 한다. 각각의 식중독균의 전체 염기서열과 현재 완료되었거나 진행 중인 생물체의 염기서열을 NCBI 의 non-redundant( nr) 데이터베이스와 BLAST 프로그램을 사용하여 비교한다. 결과를 바탕으로 특이적인 유전자와 프로브를 개발 후, PCR과 microarray와 같은 분자생물학적인 방법을 통해서 각각의 특이성을 확인 한다.
농기계용 디젤엔진에서 연료분사시기가 매연발생에 미치는 영향

A three-cylinder agricultural diesel engine was used in this work to investigate the combustion processes and smoke density with varying fuel injection timing. The fuel injection timing was adjusted to 23,21,19bTDC(before Top Dead Center) with insert shims between fuel pump block and cylinder block. Energy release schedules were determined using an one-zone thermodynamics analysis with measured cylinder pressure as a function of real time. The major conclusion of this work include:ⅰ)advanced fuel injection timing led to higher cylinder pressure occurred at around top dead center;ⅱ)advanced fuel injection timing led to longer ignition delay, and,ⅲ)retarded fuel injection timing produced fewer smoke from exhaust pipe by 50%;finally ⅳ)fuel injection timing may play the key role of affect the combustion processes and smoke density in agricultural diesel engine.
