This study aimed to identify leaf sheath function through the investigation of effect of removal times and removed number of leaf sheath on growth and flowering in gladiolus. The leaf sheath influenced on the growth and flowering regardless of light condition at early growth stages. When total leaf sheath were removed in early growth stages, flower stalk length and spike length remarkably decreased. Whereas days of flowering were little different by leaf sheath removal. Also when removed numbers of leaf sheath were 1, 2 and 3, the growth and flowering were little different compared to control. But when removed numbers of leaf sheath were 4, flower stalk length decreased above 25㎝ compared to others treatment, spike length and number of flower decreased and stem diameter became thick abnormally.

- Present experiments are conducted to study the seed viability and optimum germination temperature ofPolygonatum odoratumvar. pluriflorumthat is known to have low germination ability and long germination duration. Toenhance germination rate, various growth regulators and inorganic salts were employed. Low germination rate wasobtained with 4 year old seeds, but not with 1∼2 year old seeds. The seeds germinated very well under 22∼25℃ andgerminated speed was rapid. Especially, 1∼2 year old seeds germinated at 25℃ showed 70∼71.2% germination rate.Priming treatments using GA3, IAA, NAA, kinetin, KNO3, KH2PO4, Ca (NO3)2 were effective compared to controlexcept BA. 1 year old seeds treated with GA3 0.5 mM and IAA 1 mM showed 96% and 93% germination rate,respectively.Key words - Seed viability, PGRs, Auxin, Inorganic salts
참취, 좀개미취, 큰금계국 및 기생초 꽃의 추출방법에 따른 항산화 활성 비교

Antioxidant substances were extracted from flowers of Aster scaber, Aster maackii, Coreopsis lanceolata and Coreopsis tinctoria using 2 types of extraction methods- ultrasonic wave and reflux, and antioxidant effects were compared. Higher yield of extract was obtained by ultrasonic wave method in all 4 species. Extraction time was different depending on species, but in all 4 species DPPH and ABTS free radical scavenging and ferrous ion chelating activity were higher or similar by ultrasonic wave than reflux method. Content of total polyphenol and flavonoid was mainly higher by ultrasonic wave than reflux method. Longer the treatment, the more the antioxidant activity and bioactive substances. Anti-lipid peroxidation of ultrasonic and reflux extracts obtained from flowers of Aster scaber and Coreopsis lanceolata were compared. Flower extract of Aster scaber obtained by reflux method showed the highest inhibition effect against peroxidation of linoleic acid, but extract of Coreopsis lanceolata obtained by 15-minute ultrasonic extract showed the highest effect. Ultrasonic wave extraction was more economical and efficient method than reflux extraction. Additional key words - Lipid peroxidation, Phenolic compounds, Radical scavenging, Reflux, Ultrasound 참취, 좀개미취, 큰금계국, 기생초 꽃 추출물의 항산화 효과를 증가시킬 수 있는 적정 추출방법을 개발하기 위하여 환류냉각추출한 추출물과 15, 30, 45분 동안 초음파 추출한 추출물의 항산화 활성을 비교하였다. 추출수율은 모두 초음파 추출물에서 높게 나타났다. 종에 따라 적정 추출시간은 다르게 나타났으나, 4종 모두 초음파추출물이 환류냉각추출물 보다 DPPH와 ABTS radical 소거능 및 Ferrous ion chelating 활성이 우수하거나 유사하였다. 총 폴리페놀 및 플라보노이드의 함량 또한 대체로 초음파추출물에서 높게 나타났으며, 4종 모두 추출시간이 길어질수록 항산화 물질 함량도 증가되는 경향을 보였다. 참취와 큰금계국 꽃 추출물의 지질과산화 억제활성을 비교한 결과, 참취 꽃은 환류냉각추출물, 큰금계국 꽃은 초음파 15분 처리구에서 지질과산화 억제활성이 우수하였다. 연구의 결과, 식물 종 및 목표로 하는 항산화 효과에 따라 추출시간에 차이는 있으나, 초음파추출이 환류냉각추출보다 추출시간이 짧고 항산화 효과를 증가시킬 수 있는 효과적인 추출방법임을 확인할 수 있었다.
