        Interleukin-2가 Mouse 골수에 미치는 영향

        골수에서 혈구형성과정은 interleukin-3, interleukin-4 및 임파구 분비물질 등의 액소성 인자와 대식세포 및 임파구에 의하여 조절되며, 흉선 및 비장에서 형성되는 임파구의 조절은 interleukin-2가 관여한다. Interleukin-2 는 T 임파구에서 분비되는 T 임파구 성장인자로서 세포독성임파구의 증식을 유도하고 임파구의 활성을 증가시켜 여러 육종 및 암종에 탁월한 효과를 나타내는 종양치료제이나 interleukin-2를 암환자에 투여하면 가역적 호산구증가증, 말초혈액임파구의 변동 등 혈액학적 독성이 야기된다. 이에 저자는 mouse에 interleukin-2를 투여하면 흉골 골수에서 임파구 발생이 변동될 것으로 사료되어 mouse에 interleukin-2를 투여한 후 나타나는 흉골임파구의 변동을 관찰하였다. 실험동물로는 체중 15gm 내외의 웅성 mouse를 사용하였으며 체중 ㎏당 200만 단위의 interleukin-2, 0.2ml를 복강내에 주사하고 3시간, 6시간, 12시간, 24시간 및 48시간 경과 후에 각각 희생시켜 흉골을 적출하고 Zenker 용액에 고정한 후 Hematoxylin-eosin-azure II 염색을 시행하여 다음과 같은 결과를 얻었다. 1. Interleukin-2 투여 3시간 경과군의 mouse 흉골골수에서는 소형임파구가 약간 증가하였으며 6시간 경과군 및 12시간 경과군에서는 소형임파구가 현저히 증가하였다. 2. Interleukin-2 투여 24시간 경과군 및 48시간 경과군의 mouse 흉골골수에서는 미분화 혈구세포들이 다수 관찰되었다. 이상의 결과를 종합하면 interleukin-2는 흉골의 임파구 형성에 관여하는 것으로 사료된다. It is known that hemopoiesis is regulated by lymphocytes, macrophages and T lymphocyte-derived lymphokines, and lymphopoiesis in thymus is controlled by the interleukin-2. Interleukin-2 is a T-cell growth factor produced by antigen or mitogen activated T lymphocyte, and mediates antitumor effect by stimulating the differentiation of precurssor T cells into lymphokine activated killer cells. Although, interleukin-2 shows therapeutic effects on various sarcoma and carcinoma, it induces many hematologic toxicities, reversible eosinophilia and marked changes in peripheral lymphoid cells. Therefore the author undertook the present study to pursue the effect of interleukin-2 on the lymphocytes in the sternal marrow. Albino mice, DDY strain, weighing 15 gm were used as experimental animals. Experimental animals were killed at 3, 6, 12, 24 and 48 hours after administration of 2,000,000 unit/㎏ inter leukin-2. The specimens obtained from the sternum were stained with Hematoxylin-eosin-Azure II for histological observation. The results were as follows. 1. The number of small lymphocytes were increased slightly in the sternal marrow at 3 hours after interleukin-2 injection and considerably increased in the 6 hours and 12 hours group. 2. Undifferentiated hemopoietic cells were observed numerously at the 24 and 48 hours after interleukin-2 injection. Consequently, it is suggested that interleukin-2 would influence the lymphopoiesis in the mouse sternal marrow.

        Abstracts-Prostaglandins exhibit diverse pharmacological properties and are now recognized as local hormones that control a multitude of important physiological process. (R)-4-hydroxy-2-cyclope-ntenone is a very useful intermediate to synthesize the prostaglandin derivatives. The optically active cyclopentenone building blocks were synthesized in various ways ; by resoultion of the racemic hydroxy ketone ; by preparation from chiral natural products via resolved synthetic intermediates, by asymmetric synthesis, by microbial or enzymatic techniques, etc. The hydroxy ketone can be produced by preparation from chiral building block via microbial organisms. The key chiral building block is dimethyl-5-hydroxy-4R-hydroxy-2-oxopentane phosphate which can be synthesized the optically active cyclpentenone via Horner Emmon reaction. This paper describes the preparation of the chiral diol compound 2 in detail.

        조구등(釣鉤藤)의 4-VO로 유발한 흰쥐뇌허혈에 대한 신경방어효과

        Uncariae Ramulus et Uncus(UR, ???) has sweet in flavour and slightly cold in property, acting on the liver and pericardium channels. This drug was described in a medical classic as having the ability to remove "heat", check hyperfunction of the liver and relieve dizziness, tremors, and convulsion, and subdue "endogenous wind". So this study was planned to check the neuroprotective effect of UR on the global ischemia induced by 4-vessel occlusion in Wistar rats. and UR extract was lyophilized after extraction with 70% methanol. We induced 4-vessel occlusion for 10 minutes and reperfused again. The number of CA1 pyramidal neurons were counted after 7 days of reperfusion under the cresyl violet staining. In 4-VO ischemia model, UR showed significantly neuroprotective effects(1,000 and 500 ㎎/㎏ of UR extracts, p<0.05) compared with control group. Each neuroprotective ratio was about 23.0%, 19.0% respectively. Consequently, Uncariae Ramulus et Uncus has neuroprotective effects on the global ischermia induced by 4-vessel occlusion in Wistar rats. So we expect that Uncariae Ramulus et Uncus can be used as a drug for neurodegenerative disease.

        황금(黃芩)의 4-VO로 유발한 흰쥐뇌허혈에 대한 신경방어효과

        Scutellariae Radix(SR), the root of Scutellaria baicalensis G_EORGI, whose property, flavour and channel tropism is bitter in flavour, cold in property(寒), acting on the lung, gallbladder, stomach and large intestine channels(歸?,?,?,大??), has the effects of clearing away heat and dampness(淸熱??), purging fire, detoxicating(?火解?), stopping bleeding(止血) and preventing miscarriage(安?). This drug is one of the popular drugs in traditional Korean medicine, which has been used to antipyretic, antibacterial, antitoxic and antihypersensitive effects. So this study was planned to check the neuroprotective effect of SR on the global ischemia induced by 4-vessel occlusion in Wister rats. and SR extract was lyophilized after extraction with 70% methanol.         Regio- and Stereoselective Reactions of (S)-(1-Methylpyrrolidin-2-yl)methyl Allyl Sulfide

        The synthesis and regioselective reactions of a chiral allyl sulfide. (S)-(+)-(1-methylpyrrolidin-2-yl)methyl allyl sulfide (MAS, 1) are described. Remarkable ${\alpha}-regioselectivity$ was observed in the alkylation of the carbanion of MAS while 1:3 mixtures of ${\alpha}-and\;{\gamma}-adducts$ were produced in the addition of the MAS anion to aldehydes. However, a dramatic change of the regioselectivity was witnessed when Lewis acids such as $Et_3Al$, $Et_3B$, and $Ti(O^iPr)_4$ were used as additives in the addition reaction. In these cases, ${\alpha}-adducts$ were formed exclusively. A rationale for the change of regioselectivity is provided. And the stereochemical aspect of the addition reaction is also described.

        복수중에서의 혈청 및 복수내 Adenosine Deaminase 활성도 측정의 진단적 가치

        Objectives: In this study, we would like to evaluate the diagnostic value of adenosine deaminase activity in serum and ascitic fluid from patients with ascites of various causes, including tuberculous peritonitis, liver cirrhosis with or without hepatoma, and cancer peritonitis. Methods: The ADA was assessed in serum and ascitic fluid, and simultaneous calculation of ascitic fluid/ serum ADA ratios were obtained in 89 patients with ascites due to various causes; 42 for liver cirrhosis, 18 for hepatoma, 14 for tuberculous peritonitis, 15 for malignant ascites. Results: 1) Serum ADA activity was significantly higher in patients with liver cirrhosis than those with other causes of ascites, with the level of 61.3±49.0 U/L, and the corresponding ADA levels of other diseases were 42.8±20.4 U/L in hepatoma, 46.1±21.9 U/L in tuberculous peritonitis, 24.0±19.8 U/L in malignant ascites (p<0.05). 2) Ascitic fluid ADA activity was highest in patients with tuberculous peritonitis than those with non-tuber-culous causes of ascites, with the level of 82.0±72.3 U/L, and the corresponding ADA levels of other diseases were 21.1±31.7U/L in liver cirrhosis, 8.8±7.7U/L in hepatoma, 29.0±19.7 U/L in malignant ascites (p<0.05). Ascitic fluid ADA level greater than 80 U/L had a specificity of 97%, sensitivity of 50%, and diagnostic efficiency of 76% for the diagnosis of tuberculous peritonitis. 3) The ascitic fluid/serum ADA ratios were significantly higher in patients with malignant ascites than those with other causes of ascites, with the level of 3.5±7.0, and the corresponding levels of other diseases were 0.8±1.8 in liver cirrhosis, 0.3±0.4 in hepatoma, 1.7±1.3 in tuberculous peritonitis (p<0.05). Conclusion: These results suggest that the assessment of serum and ascitic fluid ADA activity, and simultaneous calculation of ascitic fluid/serum ADA ratios would be useful in the differential diagnosis between patients with tuberculous peritnnitis and non-tuberculous causes of ascites.

        Ovarian tumors in children present difficult problems in diagnosis and management. Clinicians has difficult problems in distinguishing the functional varients from the pathologic aberrations which make up ovarian tumors in childhood and in distinguishing benign or malignant neoplasms. The authors undertook this study because of clinical survey of juvenile ovarian tumors as observed 40 cases. The results obtained are as follows: 1) The incidence of ovarian tumors under age of 20(1960-1968) was 5.79% of all ovarian tumors. 2) The incidence of benign ovarian cystadenoma under age of 20 was dermoid cyst 43.5%, mucinous cyst 26%, serous cyst 19.5%. 3) Age distribution of juvenile ovarian tumors was between 9-20 years. The tumors occured frequently between 18-20 years. 4) The incidence of malignant ovarian tumors was about 10%. 5) Among the subjective symtpoms complained of by the patient, about 80% noticed abdominal mass, 15% complained of G-I troubles. 6) As to the objective signs among the patients, about 97% noticed abdominal mass. Abdominal tenderness was observed in 27%. 7) Benign ovarian tumors were observed 44.4% in right ovary, 38.8% in left and 16.6% in bilateral ovaries. We can see the greate roccurence of tumors in right ovary in benign ovarian tumors. 8) The size of benign and malignant ovarian tumors were nearly over adult fist size if the lutein cyst was excluded. The mucinous cysts and serous cysts generally were the largest. 9) The incidence of pedicle torsion of all ovarian tumors was 22.4%. 10) The treatment for benign ovarian tumors of 36 cases was unilateral salpingo-oophorectomy in 30 cases and bilateral salpingo-oophorectomy in 6 cases, and for malignant was surgical primarily and the principle of it was total hysterectomy with bilateral salpingo-oophorectomy with or without omentectomy, Abjuvant treatment, X-ray irradiation was applied to all malignant ovarian tumors.
