      • STAT3-RANTES autocrine signaling is essential for tamoxifen resistance in human breast cancer cells.

        <P>The acquisition of tamoxifen resistance is a major therapeutic problem in breast cancer. We developed a tamoxifen-resistant MCF-7 (TRM-7) cell line to elucidate the molecular mechanisms and factors associated with acquisition of such resistance. We showed that phosphorylation of STAT3 at tyrosine 705 (Y705) and RANTES expression are increased in response to tamoxifen in human breast cancer cells. On the basis of these results, we hypothesize that upregulated STAT3 phosphorylation and RANTES may be correlated with the development of drug resistance. Here, we showed that STAT3 and RANTES contribute to the maintenance of drug resistance. STAT3 phosphorylation is constitutively retained via a RANTES autocrine loop, which in turn upregulates anti-apoptotic signals in TRM-7 cells. STAT3-RANTES autocrine signaling affected expression of anti-apoptotic BCL-2 family genes and prevented TRM-7 cells from undergoing programmed cell death by inhibiting PARP and caspase-9 cleavage. Subsequently, blockade of STAT3 and RANTES in TRM-7 cells resulted in reduction of anti-apoptotic signals, which was rescued by exogenous RANTES treatment; drug resistance was also restored. Taken together, our results suggested that STAT3-RANTES autocrine signaling is essential for maintenance of drug resistance and inhibition of programmed cell death. These mechanisms of STAT3-RANTES autocrine signaling suggest a novel strategy for management of patients with tamoxifen-resistant tumors.</P>

        한 언어를 구문 분석하기 위해서는 해당 언어를 효과적인 방법으로 문법적 형태로 표현할 필요가 있다. 본 논문은 단일화 기반의 다양한 문법 형식을 표현할 수 있는 문법 기술도 구인 PATRII로 쓰여진 문법을 번역하여 함수의 형태를 가지는 문법 번역기를 구현한다. 본 문법 번역기는 단일화식을 미리 번역함으로써 파싱 실행시간의 향상을 기대할 수 있으며 파싱 메카니즘에 관계없이 이용될 수 있는 장점을 가지고 있다. 또한 한국어 구문 분석에 필요한 선택적 단일화를 수용할 수 있도록 문법 기술 도구를 확장한다.

        Solution Structure of a GSK 3$\beta$ Binding Motif, A $AXIN^{pep}$

        Axin is a scaffold protein of the APC/axin/GSK complex, binding to all of the other signalling components. Axin interacts with Glycogen synthase kinase 3$\beta$ (GSK 3$\beta$) and functions as a negative regulator of Wnt signalling pathways. To determine the solution structure of the GSK3$\beta$ binding regions of the axin, we initiated NMR study of axin fragment comprising residues 3$Val^{388} - Arg^{401}$using circular dichroism (CD) and two-dimensional NMR spectroscopy. The CD spectra of 3$axin^{pep}$ in the presence of 30% TFE displayed a standard 3$\alpha$-helical conformation, exhibiting the bound structure of 3$axin^{pep}$ to GSK3$\bata$. On the basis of experimental restraints including $NOE_s$, and $^3J_{HN\alpha} $ coupling constants, the solution conformation of $axin^{pep}$ was determined with program CNS. The 20 lowest energy structures were selected out of 50 final simulated-annealing structures in both water and TFE environment, respectively. The $RMSD_s$ for the 20 structures in TFE solution were 0.086 nm for backbone atoms and 0.195 nm for all heavy atoms, respectively. The Ramachandran plot indicates that the $\varphi$, $\psi$ angles of the 20 final structures is properly distributed in energetically acceptable regions. $Axin^pep$ in aqueous solutions consists of a stable $\alpha$-helix spanning residues form $Glu^{391}$ to $Val^{391} $, which is an interacting motif with GSK3$\beta$.

        The purpose of this paper is to analyze and evaluate the tendency of collective habitat model in traditional urban area proposed by the architectural competition. The analysis and evaluation of these works is made by reference-frame such as community, locality, and economy. As results, the summaries of findings are as follows: 1. In the tentative plan of the collective habitat models, these have a tendency to encourage community life as the characteristic of traditionality and locality. That is, these concepts are represented by the consideration of environs, the positive use of site conditions, and so on. 2. In evaluation of these works, the critics are composed of these groups-specialist, developer, residents. The specialist groups make an emphasis on the community life and environs, the developer groups on the plan and structure system, and the residents on the present life-cycle and conditions. 3. The collective habitat models find their traditional and local expression in the alley, court, arrangement, exterior space, and traditional space.

        Acinetobacter brisouii sp. nov., Isolated from a Wetland in Korea

        A bacterial strain $5YN5-8^T$ was isolated from peat layer on Yongneup in Korea. Cells of strain $5YN5-8^T$ were strictly aerobic, Gram-negative, coccobacilli, non-spore forming, and non-motile. The isolate exhibited optimal growth at $28^{\circ}C$, pH 7.0, and 0-1% NaCl. Results of 16S rRNA gene sequence analyses indicated a close relationship of this isolate to Acinetobacter calcoaceticus (97.8% similarity for strain DSM $30006^T$). It also exhibited 94.4-97.8% 16S rRNA gene sequence similarities to the validly published Acinetobacter species. The value for DNA-DNA hybridization between strain $5YN5-8^T$ and other members of the genus Acinetobacter ranged from 16 to 28%. Predominant cellular fatty acids were $C_{18:1}$ ${\omega}9c$, summed feature 4 containing $C_{15:0}$ iso 2-OH and/or $C_{16:1}$ ${\omega}7c$, and $C_{16:0}$. The DNA G+C content was 43.9 mol%. Phylogenetic, phenotypic, and chemotaxonomic data accumulated in this study revealed that the isolate could be classified in a novel species of the genus Acinetobacter. The name Acinetobacter brisouii sp. nov. is proposed for the novel species, with $5YN5-8^T$ (=KACC $11602^T$ =DSM $18516^T$) as the type strain.

        CCoAOMT 유전자 발현 억제에 의한 현사시나무의 화학조성 변화

        본 연구에서는 리그닌 전구물질 생합성에 관여하는 CCoAOMT (Caffeoyl-CoA-O-methyltransferase)유전자의 발현을 분자생물학적으로 억제시킨 상태와 정상적인 상태에서 생장한 현사시나무를 대상으로 홀로셀룰로오스, 리그닌과 단당류 조성에 관한 정량 분석을 각각 실시하였다. 그리고 각각의 목질바이오매스로부터 MWL (milled wood lignin)을 추출하여 메톡실기정량, DFRC, Py-GC/MS, GPC, 13C NMR 분석 등 리그닌의 화학 구조적 특성을 비교하였다. 정상재와 형질전환체의 홀로셀룰로오스 함량은 각각 81.6%와 82.3%로 큰 차이는 없었지만, 리그닌 함량은 각각 21.7%와 18.3%로 형질전환체가 약 3.4% 정도 낮았다. 정상재와 형질전환체의 구성 단당류 분석결과, 글루코오스는 각각 511.0 mg/g와 584.8 mg/g, 그리고 자일로오스는 각각 217.8 mg/g와 187.5 mg/g로 나타나 CCoAOMT 발현억제는 단당류 조성에 영향을 미치는 것으로 분석되었다. 각 시료로부터 단리한 리그닌(MWL)의 메톡실기 정량에 따르면 CCoAOMT 발현을 억제한 시료에서 약 7% 정도의 메톡실기가 감소하는 것으로 나타났고, 이는 13C-NMR 분석과 Py-GC/MS 분석을 통하여 확인할 수 있었다. 또한 Py-GC/MS 분석에 의하면 정상재와 형질전환체 리그닌의 G/S 비율이 각각 0.59와 0.44로 나타난 점으로 미루어 CCoAOMT 발현 억제는 리그닌 생합성 과정에서 G unit 생합성보다 S unit 생합성에 더 많은 영향을 미치는 것으로 예측할 수 있다. In this study, chemical compositions-holocellulose, lignin and monomeric sugars-were characterized with two poplar wood cell walls, one of which was grown at normal condition (CPW) and the other was genetically modified by antisence suppression of CCoAOMT gene expression (ACPW). Milled wood lignins were isolated from CPW and ACPW and subjected to methoxyl group, DFRC, Py-GC/MS, GPC, 13C-NMR analysis, respectively. There were few differences in holocellulose contents in both cell walls, which were determined to 81.6% in CPW and to 82.3% in ACPW. However, lignin contents in ACPW was clearly decreased by the suppression of CCoAOMT gene expression. In CPW 21.7% of lignin contents was determined, while lignin contents in ACPW was lowered to 18.3%. The relative poor solubility of ACPW in alkali solution could be attributed to the reduction of lignin content. The glucose contents of CPW and ACPW were measured to 511.0 mg/g and 584.8 mg/g and xylose contents 217.8 mg/g and 187.5 mg/g, respectively, indicating that suppression of CCoAOMT gene expression could be also influenced to the formation of monomeric sugar compositions. In depth investigation for milled wood lignin (MWL) isolated from both samples revealed that the methoxyl contents at ACPW was decreased by 7% in comparison to that of CPW, which were indirectly evidenced by 13C-NMR spectra and Py-GC/MS. According to the data from Py-GC/MS S/G ratios of lignin in CPW and ACPW were determined to 0.59 and 0.44, respectively. As conclusive remark, the biosynthesis of syringyl unit could be further influenced by antisense suppression of CCoAOMT during phenylpropanoid pathway in the plant cell wall rather than that of guaiacyl unit.

      • Parapedobacter luteus sp. nov. and Parapedobacter composti sp. nov., isolated from cotton waste compost

        <P>Two aerobic, Gram-reaction-negative, non-spore-forming bacterial strains, 4M29<SUP>T</SUP> and 4M40<SUP>T</SUP>, were isolated from cotton composts. The two strains grew in the presence of 0-5 % (w/v) NaCl (optimum growth in the absence of NaCl), at pH 6.0-8.0 (optimum, pH 7.0) and at 15-45 °C (optimum, 30 °C). The strains shared 97.1 % 16S rRNA gene sequence similarity. Strains 4M29<SUP>T</SUP> and 4M40<SUP>T</SUP> showed the next highest levels of 16S rRNA gene sequence similarity to <I>Parapedobacter koreensis</I> Jip14<SUP>T</SUP> (95.6 and 94.4 %, respectively) and <I>Parapedobacter soli</I> DCY14<SUP>T</SUP> (95.2 and 93.8 %). The level of DNA-DNA relatedness between strains 4M29<SUP>T</SUP> and 4M40<SUP>T</SUP> was 38 %. The two strains contained iso-C15 : 0, summed feature 3 (comprising iso-C15 : 0 2-OH and/or C16 : 1<I>ω</I>7<I>c</I>) and iso-C17 : 0 3-OH as major fatty acids, MK-7 as the major respiratory quinone, homospermidine as the only polyamine and phosphatidylethanolamine as the major polar lipid. The DNA G+C contents of strains 4M29<SUP>T</SUP> and 4M40<SUP>T</SUP> were 47.6 and 48.6 mol%, respectively. On the basis of phylogenetic and phenotypic data, strains 4M29<SUP>T</SUP> and 4M40<SUP>T</SUP> are considered to represent two novel species of the genus <I>Parapedobacter</I>, for which the names <I>Parapedobacter luteus</I> sp. nov. (type strain 4M29<SUP>T</SUP> =KACC 10955<SUP>T</SUP> =JCM 15977<SUP>T</SUP>) and <I>Parapedobacter composti</I> sp. nov. (type strain 4M40<SUP>T</SUP> =KACC 10972<SUP>T</SUP> =JCM 15978<SUP>T</SUP>) are proposed.</P>

        A Magnetic Nanoprobe Technology for Detecting Molecular Interactions in Live Cells

        <P>Technologies to assess the molecular targets of biomolecules in living cells are lacking. We have developed a technology called magnetism-based interaction capture (MAGIC) that identifies molecular targets on the basis of induced movement of superparamagnetic nanoparticles inside living cells. Efficient intracellular uptake of superparamagnetic nanoparticles (coated with a small molecule of interest) was mediated by a transducible fusogenic peptide. These nanoprobes captured the small molecule’s labeled target protein and were translocated in a direction specified by the magnetic field. Use of MAGIC in genome-wide expression screening identified multiple protein targets of a drug. MAGIC was also used to monitor signal-dependent modification and multiple interactions of proteins.</P>

        Acute Appendicitis: A Rare But Probable Manifestation of Kawasaki Disease

        Kawasaki disease (KD) is an acute, systemic inflammatory disorder that often targets coronary arteries. Being the common cause of acquired heart disease in children, timely diagnosis and intravenous immunoglobulin treatment are crucial. However, it is challenging for physicians to diagnose KD if it presents with atypical manifestations. We report the case of a 5-year-old boy who initially presented with appendicitis; after an appendectomy, he had a prolonged fever. He was finally diagnosed with atypical KD and successfully recovered after intravenous immunoglobulin treatment. Through a literature review, we found 21 cases of appendicitis associated with KD. In most cases, the patients were male with a mean age of 5.3 years. Most had higher proportions of incomplete KD and coronary artery complications than expected for typical KD. In conclusion, appendicitis could be a rare complication of KD; therefore, multidisciplinary cooperation and early recognition of atypical KD are essential for timely diagnosis.
