Twenty novel tetramic acid derivatives (E)-3-(1-(alkyloxyamino)ethylidene)-1-alkylpyrrolidine-2,4-diones were synthesized by the reaction of 3-(1-hydroxyethylidene)pyrrolidine-2,4-diones with O-alkyl hydroxylamines. The title compounds were confirmed by IR, 1H NMR, MS and elemental analysis. The structure of compound 6r was further verified by X-ray diffraction crystallography. The bioassays showed that most of the title compounds exhibited noticeable herbicidal and fungicidal activities.
MR T1 Image Segmentation of a Prostate Based on Distance Regularized Level Set Evolution

Prostate segmentation from MRI is a necessary first step and plays an important role in clinical decision making process. A MR T1 Image segmentation method of the prostate based on distance regularized level set evolution (DRLSE) is proposed. To smooth the prostate image to reduce the noise, a preprocessed prostate image with Gaussian kernel and get an edge indicator is convolved. We construct an energy function with a distance regularization term and an external energy term containing the edge indicator and minimize it by solving the gradient flow which can be implemented with a finite difference scheme. In segmentation experiments, the impact of parameters λ , μ , α and ε in DRLSE model on the image segmentation is analyzed, and an optimal value of these parameters is given. The experiment results confirm the effectiveness of the MRI segmentation method of prostate for different situation of different patients.
Polymer Nanostructured Components Machined Directly by the Atomic Force Microscopy Scratching Method

This study aims to machine complex three dimensional nano-structures on polymer surfaces directly using the Atomic Force Microscopy (AFM)-based nano-mechanical processing method. Processing parameters including the normal load, the feed rate, the machining speed and the scanning direction are optimized by the Taguchi method. Their effects on the machined depth and the surface roughness of the machined structures are analyzed. Meanwhile, the AFM PZT tube is controlled to achieve the tip’s three-dimensional motions in space. Using the optimized processing parameters, three-dimensional nanostructures on the surface of PMMA are directly fabricated. It is a novel low-cost manufacturing method for machining complex three-dimensional nano-structures or nano parts on polymer surfaces directly.

A melt treatment with a vibrating cooling slope and a semisolid rolling process to produce an AZ61 alloy strip was proposed. The microstructure formation mechanism and the properties of the AZ61 alloy produced by the proposed process were investigated. Due to the high cooling rate and stirring action caused by the vibration cooling slope, the nucleation rate was greatly improved, which caused the formation of fine spherical or rosette primary grains. During the rolling process, the solid fraction increased from the entrance to the exit of the roll gap, and under the shearing action of the roller, the distribution of solute in the melt was homogenous, and the primary grains grew further. When the casting temperature was 680 °C,a strip with a cross section of 4 mm×160 mm was produced and a homogeneous microstructure was obtained. The ultimate tensile strength of the AZ61 alloy strip produced by the proposed method reached 242 MPa, and the corresponding elongation to failure was 4%, which were better than those achieved in previous similar studies.

Esophageal carcinoma (EC) is one of the most aggressive cancers with a poor prognosis. Understanding the molecular mechanisms underlying esophageal cancer progression is a high priority for improved EC diagnosis and prognosis. Recently, MSP58 was shown to behave as an oncogene in colorectal carcinomas and gliomas. However, little is known about its function in esophageal carcinomas. We therefore examined the effects of MSP58 knockdown on the growth of esophageal squamous cell carcinoma (ESCC) cells in vitro and in vivo in order to gain a better understanding of its potential as a tumor therapeutic target. We employed lentiviral-mediated small hairpin RNA (shRNA) to knock down the expression of MSP58 in the ESCC cell lines Eca-109 and EC9706 and demonstrated inhibition of ESCC cell proliferation and colony formation in vitro. Furthermore, flow cytometry and western blot analyses revealed that MSP58 depletion induced cell cycle arrest by regulating the expression of P21, CDK4 and cyclin D1. Notably, the downregulation of MSP58 significantly inhibited the growth of ESCC xenografts in nude mice. Our results suggest that MSP58 may play an important role in ESCC progression.

The existence of glycine residues in long-chain scorpion toxins has been well documented. However, their role as analgesics has not been evaluated. To address this issue, we investigated the functional role of glycines in the C-terminal end of Chinese-scorpion toxin from Buthus martensii Karsch (BmK AGP-SYPU2) using site-directed mutagenesis and analgesic activity assays. Recombinant BmK AGP-SYPU2 and its mutants were efficiently expressed in E. coli and purified to homogeneity using immobilized metal ion affinity chromatography (IMAC) and cation exchange chromatography. The mouse-twisting test was used to detect the analgesic activity of BmK AGP-SYPU2 and its mutants. As a result, we identified glycines at the C-terminal end that, when altered, significantly affected analgesic activity. Also, Mut6566 was significantly decreased compared to BmK AGP-SYPU2. These data indicate that the glycines at the C-terminal end are important for the analgesic activity of BmK AGP-SYPU2. [BMB reports 2010; 43(12): 801-806]

Phytochemical investigation of Verbascum thapsus led to the isolation and identification of one new iridoid compound named verbathasin A, along with ten known compounds. The structure and relative stereochemistry of verbathasin A were elucidated by analysis of spectroscopic data. All the isolates except 10-deoxyeucommiol and ajugol were tested for antiangiogenic and antiproliferative activities, and compounds luteolin and 3-O-fucopyranosylsaikogenin F showed promising antiproliferative activities, with an obvious effect of inducing apoptosis of A549 lung cancer cells.

운동유발전위(motor evoked potential;MEP)는 척수 및 운동신경계의 기능에 대한 임상 및 진단, 그리고 척수손상 연구 모형에서와 같은 동물을 이용한 실험적 연구에 많이 이용되고 있다. 그러나 이들 연구들간에는 자극대상, 즉 뇌용적이 작고 추체외로가 상대적으로 잘 발달한 쥐와 같은 설치류에서는 자극방법이나 자극위치에 따라서 MEP의 발생과 형태가 상이하며, 추체로운동유발전위(pyramidal motor evoked potential;PMEP)와 추체외로운동유발전위(extrapyramidal motor evoked potential;EPMEP)가 혼입되어 포착되는 것으로 생각된다. 이러한 문제점을 보완하기 위해서, 본 연구는 새로운 전극을 사용해서 쥐의 대뇌피질을 자극하여 발생시킨 PMEP와 망상핵을 자극하여 발생시킨 EPMEP의 특성을 관찰기록하여, PMEP와 EPMEP의 특성을 비교고찰하였다. 이를 위해 본 연구에서 사용한 새로운 전극은 대뇌피질에 직경이 약 1.4㎜인 원판을 접촉시켜서 이를 음전극으로 하고, 원판의 중심부에서 돌출된 0.2㎜의 두께와 1.5㎜ 길이의 절연된 심을 양전극으로 이용할 수 있게 제작되었다. 그리고 PMEP와 EPMEP는 제6경수위와 제9흉수위에서 기록하였다. 본 연구에서 새로운 자극전극을 이용하여 대뇌피질을 자극함으로써 전류의 전파를 국소화시킨 결과 비교적 일관적인 PMEP를 포착할 수 있었으며, PMEP와 EPMEP의 파형이 상이함을 보여주었다. Motor evoked potentials(MEPs) have been used widely both experimentally and clinically to study the function of the motor nervous system. However, the origins and the waveforms of MEPs in small animals like rats may be different from those of potentials evoked by intracortical microstimulation in carnivore like cats because of current spread. The patterns and characteristics of MEPs also could be affected by recording techinques and methods of stimulation. In the present study, we used a specially designed electrode to stimulate localized cortical area. The pointed tip of a special stimulating electrode was inserted into the cerebral cortex perpendicular to the cortical surface so that the round area gently contacted with the cortical surface. We investigated the characteristics of pyramidal motor evoked potentials (PMEPs) and extrapyramidal motor evoked potentials (EPMEPs) evoked by the electrical stimulation of the mortex cortex and the gigantocellular reticular nucleus. Using this technique, we could constantly obtain the PMEP and EPMEP. The results of the present study showed that the PMEP and the EPMEP were different in their latencies and waveforms.
