        Differential expression of sex-related fat body proteins during the larval–pupal developmental stages of the silkworm (Bombyx mori)

        The silkworm fat body is the site of many intermediary metabolic processes, and a source of sustenance forgrowth throughout the life cycle. Fat body proteins are responsible for storing nutrients, providing energy, andregulating hormones, and they have been identified using proteomic approaches. However, detailed differentialexpression of sex-related fat body proteins has not previously been evaluated. In the present study, we characterizedthe differential expression of sex-related fat body proteins, by using 2-dimensional gel electrophoresis(2-DE) followed bymass spectrometry identification and bioinformaticsmethods.We extracted the fat body proteinsfrom 5-day-old fifth instar larvae (L5), 10-day-old fifth instar larvae (corresponding to the end of spinning[LE]), and 0-day-old pupae (P0) of the multivoltine silkworm variety “Da Zao”. We confirmed the presence of 11important sex-specific expression proteins and 14 stage-specific expression proteins.We accurately identified 13of these specific expression proteins, including actin, calponin-like protein, 75 kDa subunit NADH, receptor foractivated protein kinase C from Bombyx mori (BmRACK), IMP (inosine monophosphate) cyclohydrolase, tropomyosin1, β-tubulin, hypothetical protein, antichymotrypsin precursor, and 30 K protein precursor.We showedthat BmRACK was differentially expressed betweenmale and female silkworms.Wediscuss the biological roles ofthe specific expression proteins during the larval–pupal developmental stages.

        Soft Tissue Reconstruction of Complete Circumferential Defects of the Upper Extremity

        Background Upper extremity soft tissue defects with complete circumferential involvement are not common. Coupled with the unique anatomy of the upper extremity, the underlying etiology of such circumferential soft tissue defects represent additional reconstructive challenges that require treatment to be tailored to both the patient and the wound. The aim of this study is to review the various options for soft tissue reconstruction of complete circumferential defects in the upper extremity. Methods A literature review of PubMed and MEDLINE up to December 2016 was performed. The current study focuses on forearm and arm defects from the level at or proximal to the wrist and were assessed based on Tajima’s classification (J Trauma 1974). Data reviewed for analysis included patient demographics, causality, defect size, reconstructive technique(s) employed, and postoperative follow-up and functional outcomes (when available). Results In accordance with the Preferred Reporting Items for Systematic Reviews and Meta-Analyses guidelines, 14 unique articles were identified for a total of 50 patients (mean=28.1 years). Underlying etiologies varied from extensive thermal or electrical burns to high impact trauma leading to degloving or avulsion, crush injuries, or even occur iatrogenically after tumor extirpation or extensive debridement. Treatment options ranged from the application of negative pressure wound dressings to the opposite end of the spectrum in hand transplantation. Conclusions With the evolution of reconstructive techniques over time, the extent of functional and aesthetic rehabilitation of these complex upper extremity injuries has also improved. The proposed management algorithm comprehensively addresses the inherent challenges associated with these complex cases.

        Transcriptome sequencing discovers genes related to fatty acid biosynthesis in the seeds of Eucommia ulmoides

        Eucommia ulmoides is a focus of considerable attention because it is used to produce gutta-percha latex and raw materials for traditional Chinese medicine. The species has also been widely planted for vegetable oil production due to the high content of alpha-linolenic acid (ALA) in the seeds. However, little was known about fatty acid biosynthesis in the seeds. Here, we present transcriptome analyses that identified genes related to the fatty acid biosynthesis pathways in the seeds collected 70 and 160 days after flowering (DAF) from two varieties: ‘Huazhong No. 6’ (ALA content of 38.7 %) and ‘Huazhong No. 10’ (ALA content of 67.6 %). De novo assembly generated 96,469 unigenes with an average length of 690 bp. Among them, 11,260 unigenes identified by functional annotation against the Kyoto Encyclopedia of Genes and Genomes database were mapped to 117 metabolic pathways. 65 unigenes involved in fatty acid biosynthesis include 5 coding for 3-oxoacyl-ACP reductase, 2 coding for b-hydroxyacyl-ACP dehydratase, 10 coding for b-ketoacyl-ACP synthase II, and 2 coding for enoyl-ACP reductase I. Expression of the nine differentially expressed unigenes coding for major enzymes in fatty acid biosynthesis was validated by real-time fluorescent quantitative PCR (qPCR) using RNA from six seed stages (70, 88, 106, 124, 142, and 160 DAF) in ‘Huazhong No. 10’. The qPCR analysis showed that expression of these 9 unigenes all changed significantly at 88 DAF. This study provides a large-scale transcriptome annotation of E. ulmoides, which is fundamental information for further characterizing gene functionals involved in the process of seed development.

        Highly Selective Production of Compound K from Ginsenoside Rd by Hydrolyzing Glucose at C-3 Glycoside Using β-Glucosidase of Bifidobacterium breve ATCC 15700

        To investigate a novel β-glucosidase from Bifidobacterium breve ATCC 15700 (BbBgl) to produce compound K (CK) via ginsenoside F2 by highly selective and efficient hydrolysis of the C-3 glycoside from ginsenoside Rd, the BbBgl gene was cloned and expressed in E. coli BL21. The recombinant BbBgl was purified by Ni-NTA magnetic beads to obtain an enzyme with specific activity of 37 U/mg protein using pNP-Glc as substrate. The enzyme activity was optimized at pH 5.0, 35°C, 2 or 6 U/ml, and its activity was enhanced by Mn<sup>2+</sup> significantly. Under the optimal conditions, the half-life of the BbBgl is 180 h, much longer than the characterized β-glycosidases, and the Km and V<sub>max</sub> values are 2.7 mM and 39.8 μmol/mg/min for ginsenoside Rd. Moreover, the enzyme exhibits strong tolerance against high substrate concentration (up to 40 g/l ginsenoside Rd) with a molar biotransformation rate of 96% within 12 h. The good enzymatic properties and gram-scale conversion capacity of BbBgl provide an attractive method for large-scale production of rare ginsenoside CK using a single enzyme or a combination of enzymes.

        Health-related quality of life of patients with inflammatory bowel disease in Singapore

        Background/Aims: Inflammatory bowel disease (IBD) is associated with considerable impairment of patients’ health-related quality of life (HRQoL). Knowledge of factors that significantly affect IBD patients’ HRQoL can contribute to better patient care. However, the HRQoL of IBD patients in non-Western countries are limited. Hence, we assessed the HRQoL of Singaporean IBD patients and identified its determinants. Methods: A prospective, cross-sectional study was conducted at Singapore General Hospital outpatient IBD Centre. The HRQoL of IBD patients was assessed using the short IBD questionnaire (SIBDQ), Short Form-36 physical and mental component summary (SF-36 PCS/MCS) and EuroQol 5-dimensions 3-levels (EQ-5D-3L) and visual analogue scale (VAS). Independent samples t-test was used to compare HRQoL between Crohn’s disease (CD) and ulcerative colitis (UC). Determinants of HRQoL were identified through multiple linear regression. Results: A total of 195 IBD patients (103 UC, 92 CD) with a mean disease duration of 11.2 years were included. There was no significant difference in HRQoL between patients with UC and CD. Factors that significantly worsened HRQoL were presence of active disease (b=-6.293 [SIBDQ], -9.409 [PCS], -9.743 [MCS], -7.254 [VAS]), corticosteroids use (b=-7.392 [SIBDQ], -10.390 [PCS], -8.827 [MCS]), poor medication adherence (b=-4.049 [SIBDQ], -1.320 [MCS], -8.961 [VAS]), presence of extraintestinal manifestations (b=-13.381 [PCS]), comorbidities (b=-4.531 [PCS]), non-employment (b=-9.738 [MCS], -0.104 [EQ-5D-3L]) and public housing (b=-8.070 [PCS], -9.207 [VAS]). Conclusions: The HRQoL is impaired in this Asian cohort of IBD. The magnitude of HRQoL impairment was similar in UC and CD. Clinical characteristics were better determinants of patients’ HRQoL than socio-demographic factors. Recognizing the factors that impact patients’ HRQoL would improve the holistic management of IBD patients. (Intest Res 2019;17:107-118)

        In Vivo Protein Transduction: Delivery of PEP-1-SOD1 Fusion Protein into Myocardium Efficiently Protects against Ischemic Insult

        Myocardial ischemia-reperfusion injury is a medical problem occurring as damage to the myocardium following blood flow restoration after a critical period of coronary occlusion. Oxygen free radicals (OFR) are implicated in reperfusion injury after myocardial ischemia. The antioxidant enzyme, Cu, Zn-superoxide dismutase (Cu, Zn-SOD, also called SOD1) is one of the major means by which cells counteract the deleterious effects of OFR after ischemia. Recently, we reported that a PEP-1-SOD1 fusion protein was efficiently delivered into cultured cells and isolated rat hearts with ischemia-reperfusion injury. In the present study, we investigated the protective effects of the PEP-1-SOD1 fusion protein after ischemic insult. Immunofluorescecnce analysis revealed that the expressed and purified PEP-1-SOD1 fusion protein injected into rat tail veins was efficiently transduced into the myocardium with its native protein structure intact. When injected into Sprague-Dawley rat tail veins, the PEP-1-SOD1 fusion protein significantly attenuated myocardial ischemia-reperfusion damage; characterized by improving cardiac function of the left ventricle, decreasing infarct size, reducing the level of malondialdehyde (MDA), decreasing the release of creatine kinase (CK) and lactate dehydrogenase (LDH), and relieving cardiomyocyte apoptosis. These results suggest that the biologically active intact forms of PEP-1-SOD1 fusion protein will provide an efficient strategy for therapeutic delivery in various diseases related to SOD1 or to OFR.
