Effects of Modafinil on Behavioral Learning and Hippocampal Synaptic Transmission in Rats

Purpose: Modafinil is a wake-promoting agent that has been proposed to improve cognitive performance at the preclinical and clinical levels. Since there is insufficient evidence for modafinil to be regarded as a cognitive enhancer, the aim of this study was to investigate the effects of chronic modafinil administration on behavioral learning in healthy adult rats. Methods: Y-maze training was used to assess learning performance, and the whole-cell patch clamp technique was used to assess synaptic transmission in pyramidal neurons of the hippocampal CA1 region of rats. Results: Intraperitoneal administration of modafinil at 200 mg/kg or 300 mg/kg significantly improved learning performance. Furthermore, perfusion with 1mM modafinil enhanced the frequency and amplitude of spontaneous postsynaptic currents and spontaneous excitatory postsynaptic currents in CA1 pyramidal neurons in hippocampal slices. However, the frequency and amplitude of spontaneous inhibitory postsynaptic currents in CA1 pyramidal neurons were inhibited by treatment with 1mM modafinil. Conclusions: These results indicate that modafinil improves learning and memory in rats possibly by enhancing glutamatergic excitatory synaptic transmission and inhibiting GABAergic (gamma-aminobutyric acid-ergic) inhibitory synaptic transmission.
Verticillium Wilt of Okra Caused by Verticillium dahliae Kleb. in China

Okra (Abelmoschus esculentus (L.) Moench) has gained more popularity as an economically significant plant for its nutritional and medicinal value, especially in China. During 2014-2016, the root disease of okra was discovered in four okra commercial fields surveyed in China. A fungul was isolated from the infected tissues, and was identified by Verticillium dahliae based on morphological characteristics. Pathogenicity test demonstrated that the fungus was pathogenic on okra, and fulfilled Koch’s postulates. The analysis of three sequences revealed 99-100% identity with the reported V. dahliae strain in GenBank. Neighbor-joining analysis of the gene sequences revealed that the representative isolates were clustered with V. dahliae. To the best of our knowledge, this is the first report of Verticillium wilt of okra in China.

Although BMP9 is highly capable of promoting osteogenic differentiation of mesenchymal stem cell (MSCs), the molecular mechanism involved remains to be fully elucidated. Here, we explore the possible involvement and detail role of JNKs (c-Jun N-terminal kinases) in BMP9-induced osteogenic differentiation of MSCs. It was found that BMP9 stimulated the activation of JNKs in MSCs. BMP9-induced osteogenic differentiation of MSCs was dramatically inhibited by JNKs inhibitor SP600125. Moreover, BMP9-activated Smads signaling was decreased by SP600125 treatment in MSCs. The effects of inhibitor are reproduced with adenoviruses expressing siRNA targeted JNKs. Taken together, our results revealed that JNKs was activated in BMP9-induced osteogenic differentiation of MSCs. What is most noteworthy, however, is that inhibition of JNKs activity resulted in reduction of BMP9-induced osteogenic differentiation of MSCs, implying that activation of JNKs is essential for BMP9 osteoinductive activity. The effects of sintering temperature on the pore characteristics and strength of porous corundum-mullite ceramics made from Al(OH)3, kaolinite gangue and MgCO3 were investigated through an X-ray diffractometer (XRD), scanning electron microscopy (SEM), mercury porosimetry measurement, etc. It was found that the sintering temperature strongly affects the formation of secondary mullite, and then changes the pore characteristics and strength. The formation of secondary mullite takes place extremely quickly at 1500 oC. The pore size distributions are bimodal in samples sintered at 1300 oC and 1400 oC,and mono-modal in samples sintered at 1500 oC and 1600 oC. The strength are the same and low when the sintering temperatures are 1300 oC and 1400 oC, and increase sharply when the sintering temperature is increased to 1500 oC. The most apposite mode is a sample sintered at 1500 oC which has a high mullite content (86.6 wt%), high apparent porosity (42%), high crushing strength (52 MPa) and a homogeneous pore size distribution.
Antimicrobial glycoalkaloids from the tubers of Stephania succifera

Three new glycoalkaloids, N-formyl-asimilobine-2-O-b-D-glucoside (1), (-)-1-O-b-D-glucoside-8-oxotetrahydropalmatine(2), and 1-N-monomethylcarbamateargentinine-3-O-b-D-glucoside (3) were isolated fromtubers of Stephania succifera. The structures were establishedbased on spectroscopic analysis, and the antimicrobialactivities of the three glycoalkaloids are reported.

Objective: Using a pulsating coronary artery phantom at high heart rate settings, we investigated the efficacy of a motion correction algorithm (MCA) to improve the image quality in dual-energy spectral coronary CT angiography (CCTA). Materials and Methods: Coronary flow phantoms were scanned at heart rates of 60−100 beats/min at 10-beats/min increments, using dual-energy spectral CT mode. Virtual monochromatic images were reconstructed from 50 to 90 keV at 10-keV increments. Two blinded observers assessed image quality using a 4-point Likert Scale (1 = non-diagnostic, 4 = excellent) and the fraction of interpretable segments using MCA versus conventional algorithm (CA). Comparison of variables was performed with the Wilcoxon rank sum test and McNemar test. Results: At heart rates of 70, 80, 90, and 100 beats/min, images with MCA were rated as higher image scores compared to those with CA on monochromatic levels of 50, 60, and 70 keV (each p < 0.05). Meanwhile, at a heart rate of 90 beats/min, image interpretability was improved by MCA at a monochromatic level of 60 keV (p < 0.05) and 70 keV (p < 0.05). At a heart rate of 100 beats/min, image interpretability was improved by MCA at monochromatic levels of 50 keV (from 69.4% to 86.1%, p < 0.05), 60 keV (from 55.6% to 83.3%, p < 0.05) and 70 keV (from 33.3% to 69.3%, p < 0.05). Conclusion: Low-keV monochromatic images combined with MCA improves image quality and image interpretability in CCTAs at high heart rates.

Silibinin, a natural flavonoid antioxidant isolated from extracts of the milk thistle herb, has recently been identified as having antihepatotoxic and anticancer properties. In this paper, we investigated the effects of silibinin on behavior and neuroplasticity in mice subjected to chronic unpredictable mild stress (CUMS). After 5 consecutive weeks of CUMS, the mice were treated with silibinin (100 mg/kg, 200 mg/kg and 400 mg/kg by oral gavage) for 3 consecutive weeks. The results showed that silibinin administration significantly alleviated the CUMS-induced depressive-like behavior, including the total number of squares crossed and the frequency of rearing in the open field test, the immobility time in the tail suspension test and the forced swimming test. Furthermore, silibinin treatment increased the levels of brain-derived neurotrophic factor (BDNF), serotonin (5-HT) and norepinephrine (NE) in the prefrontal cortex and hippocampus. Our study provides new insight into the protective effects of silibinin on the depressive status of CUMS mice, specifically by improving neuroplasticity and neurotransmission.
Fault Diagnosis Model Based on Multi-level Information Fusion for CNC Machine Tools

The difficulty of CNC machine tools fault diagnosis is bigger than other general equipments because of the complex structure and the coupling among subsystems. The fault diagnosis model based on multi-level information fusion and hybrid intelligence is studied to improve reliability of fault diagnosis. Information from built-in sensors is used to monitor the status of CNC machine tools. The diagnosis principles of internal parameters-motor current, torque, temperature and following error are analyzed. Internal information and external sensors are two main sources which provide data to diagnosis. In order to detect effective fault signal, features of time domain, frequency domain and time-frequency domain are extracted. All these features constitute the feature set. The features are selected by the method of Kernel Principal Component Analysis (KCPA). Then the sensitive feature set is obtained. The method of multiple classifier fusion based on fuzzy comprehensive evaluation is researched. The determination method of weight based on information entropy is proposed. This diagnosis model has been tested feed system mechanical fault diagnosis of CNC machine tools and the results show which is effective and versatile.

Substructure pseudo-dynamic hybrid simulation (SPDHS) is an advanced structural seismic testing method which combines physical experiment and numerical simulation. Generally, the key components which display nonlinearity first are taken as experimental substructures for actual test, and the remaining parts are modeled in simulation. Model updating techniques can be effectively applied to enhance the model precision of nonlinear numerical elements. Specifically, the constitutive model of the experimental substructure is identified online by the instantaneously-measured data, and the corresponding numerical elements with similar hysteretic behaviors are updated synchronously. Artificial neural network (ANN) can recognize the system which cannot be represented by definite numerical model, and thus avoids the structural response distortion caused by the inherent numerical model defects. In this study, a framework for online model updating in SPDHS with ANN method is expanded to implement actual test validation. Moreover, the effectiveness of ANN method is demonstrated by practical tests of a two-story frame model with bending dampers. Additionally, the unscented Kalman filter technique and offline ANN identification approach are both examined in the test validation. The experimental results show that, under the identical loading history, the online ANN method can significantly reduce the model errors and improve the accuracy of SPDHS.
