        둥시 장아찌 제조 과정 중 이화학적 특성 변화

        떫은감 품종의 하나인 등시를 이용하여 감장아찌 제조 중 과육의 이화학적 변화를 조사한 결과, 염도 변화는 저농도 간장 및 된장 침지의 경우 다소 완만한 증가를 보였고, 고농도의 간장 및 된장 침지의 경우 침지 기간내내 증가하는 것으로 나타났다. 간장 침지액 감장아찌의 경도는 간장 및 된장 모두 20-80% 농도에서는 침지 초기에는 증가하다가 침지 기간이 경과할수록 감소하는 경향을 나타내었다. 간장 침지 감장아찌의 색도 변화는 저장기간이 경과할수록 명도가 낮아지는 경향이었고, 적색도와 황색도는 침지액 농도를 달리하였을 경우 농도가 낮을수록, 침지 기간이 길어질수록 높았으며, 침지액 농도에 따른 차이는 크지 않았다. 침지액의 간장 및 된장의 농도를 달리하였을 때 감장아찌 중의 탄닌함량의 변화는 모든 침지액 농도에서 침지 기간이 지속될수록 탄닌의 함량은 저장 초기에 서서히 감소되다가 침지 기간이 길어질수록 급격히 감소하였으며, 첨가량에 따른 탄닌함량의 변화폭은 크지 않았다. 침지액의 간장 및 된장의 농도를 달리하였을 때 감과육에 존재하는 연화효소인 polygalacturonase와 pectinesterase 활성 변화는 간장침지 및 된장침지 모두 침지 초기부터 효소활성이 증가하기 시작하였으며 침지 기간이 경화할수록 효소활성은 더욱 높아지는 것을 알 수 있었고, 침지액의 농도가 높아질수록 효소활성이 억제되었다. Changes of physicochemical properties during the preparation of persimmon pickles were investigated. The salinity in persimmon pickles increased during storage time (0~50 days) with soy sauce and soy paste. When the fermented soy sauce and soy paste was added to the soaking solution with 80~100%, the salinity increased more rapidly. The hardness of persimmon pickles with soy sauce and soy paste was slightly increased up to the 20 or 30th day of storage and then decreased. L value of persimmon pickles was gradually decreased, but a and b value were slightly increased. The concentration of soluble tannin in persimmon pickles was slightly decreased down to the 20th day of storage and then decreased rapidly. The activities of polygalacturonase and pectinesterase as softening enzyme in persimmon pickles with soy sauce and soy paste increased during storage time (0-50 days) and enzyme activity was inhibited by high concentration of soaking solution.

        Sclerotic fibroma is a rare skin neoplasm that can occur seen sporadically or in association with Cowden`s disease. Clinically, it presents as asymptomatic flesh-colored or pinkish slow-growing papules or nodules with a wide anatomical distribution. We report a rare case of pedunculated type of sclerotic fibroma resembling soft fibroma, which has not been reported in Korea. (Korean J Dermatol 2011;49(11):1032∼1034)

      • Transcutaneous Delivery of Skin-penetrating Phosphatase Modulates Inflammatory Cytokine Signaling for Atopic Dermatitis and Psoriasis

        Background: Atopic dermatitis and psoriasis are the most common chronic inflammatory skin diseases. However, there are high unmet medical needs with limitations for transcutaneous drug development due to the skin barrier. Objective: We aimed to identify a novel transdermal delivery peptide and develop a transcutaneously applicable immune modulatory protein by targeting cytokine signaling in atopic dermatitis and psoriasis. Methods: We generated reporter proteins conjugated with a human-originated transdermal delivery peptide, AP and analyzed intracellular delivery efficiency in mouse or human skin cells and tissues. Especially, we utilized multi-photon confocal microscopy for visualizing the localized proteins. We also generated a recombinant therapeutic protein, AP-rPTP, which is conjugated with AP and phosphatase domain of TC-PTP targeting multiple STATs. Immune modulatory function of AP-rPTP was confirmed in splenocytes upon cytokine stimulation and TcR stimulation. Finally, we confirmed in vivo efficacy of AP-rPTP in OXA-induced acute dermatitis, OVA-induced chronic dermatitis, and imiquimod-induced psoriasis models. Results: AP-conjugated reporter proteins exhibited significant intracellular transduction efficacy in keratinocytes, fibroblasts, and immune cells. In addition, transcutaneous administration of AP-dTomato showed significant localization into the dermis and epidermis in mouse skin. AP-rPTP could inhibit pSTAT1, pSTAT3, and pSTAT6 in splenocytes and also regulate T cell activation and proliferation. Transcutaneous paper-patch of AP-rPTP protein significantly ameliorated ear thickness, tissue inflammation, and cytokine expression in allergy- as well as psoriasis-like dermatitis models. Conclusion: We identified a 9 amino acid-long novel transdermal delivery peptide and suggest the feasibility of utilizing AP for transcutaneous biologic drug development such as AP-rPTP as a novel immune modulatory drug candidate for dermatitis.

      • 문화재 주변 역사문화경관 보전을 위한 분석기법

        본 연구에서는 한양도성 내부의 온전하게 보존되어 있는 조망축을 보존하면서 역사문화자원과 도시개발이 이룰 수 있는 건축물 고도제한을 제시하고자 하였다. 구역별 고도제한은 구역별로 조망축 내부의 지형적 특성으로 고려하여 고도제한의 기준을 제시하였으며, 고도제한에 대한 적용기준은 다음과 같이 요약 할 수 있다.9)시곡면 분석 결과를 바탕으로 사선기준에 따라 설정된 경관고도 설정값에 따라 경관시뮬레이션을 실시한 결과창덕궁에서의 조망축이 유지되어 역사문화경관의 보존이 가능한 것으로 분석되었다. 첫째, A구간과 B구간은 선잠단지 주변으로 2층 이하로 설정하였으며, 지면높이를 포함하여 70m를 넘지 하였으며, 혜화동 로터리 구간은 3층에서 17층까지 가능한 것으로 분석되었으나, 서울 문묘와 성균관 사선제한 적용하여 2층에서 10층으로 하도록 하였다. 둘째, C구간은 창경궁과 서울 대한의원 등이 분포하고 있어 3층을 최고층수 높이로 설정하기 위해 창경궁기점으로 사선제한을 7°로 설정하였다. 셋째, D, E구간에서는 종묘 정전에서 건물이 보이지 않는 5°의 사선제한으로 3층에서 13층까지 가능하나 내부에서는 3층~10층으로 설정하였으며, 역사문화환경보존지역 에서는 3~5층, 그 외 지역에서는 10층 이하로 설정하였다. 넷째, F, G구간은 성락원, 창덕궁, 종묘의 조망축이 중첩되는 구간으로 개발압력이 높다. 이곳은 중 종묘의 범위에서는 건물의 높이가 표고75m를 넘지 않도록 하였으며, 창덕궁과 종묘의 조망이 중첩되는 구간은 건물의80m 이내로 하였다. 그동안의 역사문화경관의 보존을 위해 단일문화재에 대한 조망점을 보존하고 있었으나, 도심지의 역사문화자원의 광역적인 조망경관축의 보존을 위한 고도제한 기준을 제시함으로써 도시의 경관관리의 정량적인 관리방안을 제시하였다는데 의의가 있다. In this study tried to propose the plan of limiting the height of the building so that the view axis of the Hanyangdoseong Ancient Fortress can be maintained and the cultural and historical resources can be harmonized with the urban development. The limitation of the height of the buildings has been suggested for each zone taking into consideration of the topographical features of the area inside the view axis here. First, Zone A is located around Seonjamdanji, and the building in Zone A shall be 2-story building or below. The height of building shall not exceed 70m including the height of the ground. Secondly, Zone B is located from Hangyangdoseong Fortress, Seoul to the rotary of Hyehwadong. The height of the building in this Zone is found to be possible from 3-story building to 17-story builing. But if the setback criteria is applied from the Munmyo and Seongkyunggwan, the height should be from 2-story building to 10-story building Thirdly, Zone C is located around Changgyeonggung Palace and Seoul Daehanuiwon Korea Traditional Medical Clinic. To set the 3 story building at the highest building height, the setback was set using the slope line of 7° from the point of Changgyeonggung Palace. Fourth, Zone F and G has the buildings of Seongrakwon, Changgyeonggung Palace and Jongmyo and the property owners here want to develop their properties higher. From the view axis of Jongmyo, the building height should be less than 75m from the ground while it should be less than 80m when the views from Changdeokgung Palace and Jongmyo are overlaid. Up to now the view point for the single cultural remains has been maintained for the preservation of the single cultural remains. But in this study, I suggested the hight limitation for preservation of historical and cultural remains of the urban area taking into consideration the broader view axis. So, this study has meaning in that it has proposed the quantitive management method for the control of landscape in urban area.

        Protein tyrosine phosphatase conjugated with a novel transdermal delivery peptide, astrotactin 1–derived peptide recombinant protein tyrosine phosphatase (AP-rPTP), alleviates both atopic dermatitis–like and psoriasis-like dermatitis

        <P><B>Background</B></P> <P>Atopic dermatitis (AD) and psoriasis are the 2 most common chronic inflammatory skin diseases. There is an unmet medical need to overcome limitations for transcutaneous drug development posed by the skin barrier.</P> <P><B>Objective</B></P> <P>We aimed to identify a novel transdermal delivery peptide and to develop a transcutaneously applicable immunomodulatory protein for treating AD and psoriasis.</P> <P><B>Methods</B></P> <P>We identified and generated reporter proteins conjugated to astrotactin 1–derived peptide (AP), a novel transdermal delivery peptide of human origin, and analyzed the intracellular delivery efficiency of these proteins in mouse and human skin cells and tissues using multiphoton confocal microscopy. We also generated a recombinant therapeutic protein, AP–recombinant protein tyrosine phosphatase (rPTP), consisting of the phosphatase domain of the T-cell protein tyrosine phosphatase conjugated to AP. The immunomodulatory function of AP-rPTP was confirmed in splenocytes on cytokine stimulation and T-cell receptor stimulation. Finally, we confirmed the <I>in vivo</I> efficacy of AP-rPTP transdermal delivery in patients with oxazolone-induced contact hypersensitivity, ovalbumin-induced AD-like, and imiquimod-induced psoriasis-like skin inflammation models.</P> <P><B>Results</B></P> <P>AP-conjugated reporter proteins exhibited significant intracellular transduction efficacy in keratinocytes, fibroblasts, and immune cells. In addition, transcutaneous administration of AP-dTomato resulted in significant localization into the dermis and epidermis in both mouse and human skin. AP-rPTP inhibited phosphorylated signal transducer and activator of transcription (STAT) 1, STAT3, and STAT6 in splenocytes and also regulated T-cell activation and proliferation. Transcutaneous administration of AP-rPTP through the paper-patch technique significantly ameliorated skin tissue thickening, inflammation, and cytokine expression in both AD-like and psoriasis-like dermatitis models.</P> <P><B>Conclusion</B></P> <P>We identified a 9-amino-acid novel transdermal delivery peptide, AP, and demonstrated its feasibility for transcutaneous biologic drug development.
