Enhanced Macrophage Antitumor Effects Protein A in Combination with IFN-γ

In this study we examined the potential for the synergistic augmentation of the antitumor activity of inflammatory mouse peritoneal macrophages by stimulation with protein A combined with IFN-γ. The moderate augmentative effect induced by preincubation with protein A was demonstrated to be concentration-dependent, whereas IFN-γ had a very low activating effect. Following preincubation with both protein A and IFN-γ, a marked enhancement of macrophage activity was noted. In addition, based on the utilization of neutralizing antibody to TNF-α or the inhibition of NO production, TNF-α and NO were proven to be involved as mediators during the activation of tumoricidal macrophages by protein A in combination with IFN-γ. We also demonstrated that supernatants from macrophages treated with protein A plus IFN-γ contained both TNF-α and NO at markedly increased levels. Thus, tumor cell lysis in the combined system was mediated via TNF-α or NO. These results demontstrate the synergistic effects on mouse peritoneal macrophage function of protein A in combination with IFN-γand suggest that combinations of such agents may serve as the basis for future in vivo immunotherapy.
The Mechanisms of Interferon-α and Interferon-γ Induced Antiviral Activity in Peritoneal Macrophages

대식세포는 세포 내에서 바이러스복제를 억제함으로서 바이러스 감염에 대한 숙주반응에서 중요한 역할을 한다. 인터페론은 바이러스 감염시 세포가 분비하는 항바이러스 효과를 갖고 있는 단백질로서 인터페론 생성을 유도하는 poly I : C와 같이 대식세포에서 다른 인터페론을 유도하여 항바이러스작용을 나타낼 가능성이 있기 때문에 인터페론에 의해 활성화된 대식세포가 HSV-1의 감염에 대한 저항작용기전을 연구하였다. 대식세포를 인터페론-α 또는 인터페론-γ 및 인터페론 항체를 함께 처리하여 HSV-1감염에 대하여 저항하는 대식세포의 능력을 평가하였다. 인터페론 항체는 HSV-1에 감염된 대식세포에서 인터페론의 보호효과를 중화시키지 못하였다. 또한 인터페론을 처리한 대식세포의 상층액에서 인터페론을 탐지할 수 없었다. 이는 poly I : C가 인터페론-β 생성을 유도하여 항바이러스효과를 나타내는 작용기전 (Pyo et al. J. Leukocyte Biol. 50:479)과는 달리 인터페론-α와 인터페론-γ는 인터페론-β 생성과정을 경유하지 않고 직접적으로 대식세포에 작용하여 항바이러스 효과를 나타내는 듯하다.

Recently, Rhei Rhizoma extracts (RRE) have begun to receive more attention as potential biological response modifiers. In the present study, we studied the antiproliferative effect of RRE on tumor cells and the effect of RRE on macrophage function. A variety of tumor cells and macrophages were treated with RRE at various concentrations. The effect of RRE on cell proliferation was measured by MTT assay and the effect of RRE on the production of nitric oxide (NO) was determined in the macrophage-like cell lines Raw264.7, C6 and peritoneal macrophages (pMQ). RRE inhibited the growth of tumor cells (e.g., B16, HOS). However, the effects of RRE on the production of NO varied with macrophage types. RRE had no effect on C6 cell growth and slightly increased the growth of Raw264.7 cells. In addition, treatment of normal pMQ with RRE enhanced NO production in a concentration-dependent manner, whereas RRE suppressed NO production at 50 ㎍/mL in both Raw264.7 and C6 cells. However, RRE suppressed NO production in LPS/IFN-γ-stimulated C6 cells. Overall, these results suggest that RRE elicits an antiproliferative property and differentially modulates NO production in various macrophages, and have a potential for therapeutic application.
Immunomodulatory Effects of Cimicifugae Rhizoma Extracts in Macrophages

Cimicifugae Rhizoma (CR) belongs to the Ranunculaceae family, which has been traditionally used to treat climacteric complaints, antipyretics and diaphoresis as an alternative medicine for estrogen hormone replacement therapy with estrogens. Recently, it has been reported that different extract fractions of CR have various effects such as anti-allergic, anti-inflammatory and anti-proliferative activities. The current study investigated the immunomodulatory effects of Cimicifugae Rhizoma water extracts (CRE) in the macrophage-like cell line, Raw 264.7. Our results showed that CRE (1~50 ㎍/mL) stimulated tumoricidal activity and NO production, whereas phagocytic activity was inhibited at the same concentrations. Additionally, iNOS mRNA expression was significantly increased in Raw 264.7 exposed to CRE as demonstrated by RT-PCR. These results indicate that the tumoricidal activity induced by CRE may be mediated by the production of NO and these activities may be useful for the treatment of diseases such as cancer.

Abstract - Poly I:C에 의해 유도된 대식세포의 항바이러스 작용에 RNA 또는 단백질 합성이 필요한지를 결정하였다. Actinomycin D 또는 cycloheximide로 대식세포를 전처리 하였을 때 poly I:C의 항바이러스 효과가 감소되었으나 actinomycin D 또는 cycloheximide를 후처리하였을 때는 억제되지않아 poly I:C의 항바이러스 작용에 RNA와 단백질 합성이 요구되는 듯 하며 poly I:C 처리의 초기단계에서 interferon이 생성되는 듯하다.
