      • 고지방식이가 운동시 흰쥐의 글리코겐 이용에 미치는 영향

        Carbohydrate and fat ingestion during exercise are essential nutrient, but a number of opinion is different from their contents of food. In this point of view, to study how high-fat dietary contents affects glycogen utilization, 48 female Sprague-Dawley rats were trained on the treadmill trained for 4wk with consuming two high-fat diet and one high-carbohydrate diet. All animals were fed 79%high-fat diet(n=16, 79F), 40% high-fat diet(n=16, 40F), 70% high-carbohydrate diet(n=16, 70C). After the final day of training, one-half of rats in each the three groups were run to exhaustion (E), whereas the remaining rats served as rested controls (R). All animals were trained on a motorized rodent treadmill. The animals ran for 30min/d at 28m/min(0% grade), analysis items were glycogen, body weight and %Fat were measured. All data were examined by means of one-way ANOVA and significance was set at the p<0.05. The Duncan method was used for a post hoc test. Body weight were similar among the 79F, 40F and 70C groups. %Fat was significantly greater in animals that consumed the 79F group than in those on the 70C group (P<0.05), even though similar body weight each other. Generally liver and muscle glycogen concentration had more in 79FR, on the other hand, had less in 79FE compared with the others. The mean overall glycogen utilization rate was depressed in muscle and liver after animals were on the 79% F diet. In summary, the present study has suggested that consumption of 79% F diet during endurance training can lead to an increase in exercise performance in female rats. Some of the consequences training on F diet are an increase in body fat deposition and slower utilization of glycogen during exercise. These data indicate that rats exposed to a high-fat diet are capable of prolonging exercise in spite of limited glycogen stores. This improved capacity for exercise appears to be partially the result of muscular adaptations to the diet, which apparently increase the ability to oxidize fat and concomitantly spare glycogen.

        인체혈장 중 에탐부톨의 HPLC 분석법의 검증 및 단일용량 투여에 의한 약물동태 연구

        An HPLC method was employed for the determination of ethambutol in human plasma. After addition of internal standard (IS, octylamine, 2 μg/mL) and alkalinization of the plasma with 5 M sodium hydroxide, the drug and IS were extracted into the mixture of chloroform and diethyl ether (40:60, v/v). Following a 15-min vortex-mixing and a 10-min centrifugation, the organic phase was spiked with 100 pL of phenylethylisocyanate (2000 μg/mL) for chemical derivatization, mixed for 5 min and evaporated to dryness under a stream of nitrogen. The residue was reconstituted with 100 μL of mobile phase and 20 pL was injected into Cl8 column with a mobile phase consisting of methanol:water (70:30, v/v). The samples were detected utilizing an ultraviolet detector at 200 nm. The method was specific and validated with a limit of 0.15 μg/mL. Infra- and inter-day precision and accuracy were acceptable for all quality control samples including the lower limit of quantification. The applicability of this method was demonstrated by analysis of human plasma after oral administration of a single 1200-mg dose to 20 healthy subjects. From the plasma ethambutol concentration vs. time curves, the mean AUC was 9.61 ± 1.64 μg hr/mL. and Cmax of 2.68 μg/mL reached 2.73 hr after administration. The mean biological half-life of ethambutol was 3.46 ± 1.21 hr. Based on the results, this simple and validated assay could readily be used in any pharmacokinetic studies using humans.

        휴대형 자동, 수동형 및 탁상형 자동 각막곡률검사기로 측정한 각막곡률의 신뢰성

        Purpose: To compare the keratometry measurements by the handheld Nikon Retinomax K-plus2, the on-table Canon RK-5 autokeratometer, and the manual Topcon OM-4 keratometer and to evaluate the degree of agreement among the three instruments. Methods: Adults (n=30) and children (n=40) presenting toa tertiary eye hospital clinic were examined by an optometrist. Topcon OM-4 keratometer, Nikon Retinomax and the on-table Canon RK-5 autokeratometer analyses were performed in order. The horizontal and vertical keratometry data were measured. The vector components of astigmatism, mean bias and agreement among the three types of measurements were evaluated. Results: There were no significant differences among the three keratometers in horizontal or vertical keratometry or in central corneal astigmatism in both the adult and child groups (p-value>0.05). The keratometers which were estimated with Retinomax K-plus2 were highly correlated with those by on-table RK-5 autokeratometer (min. 0.89, max. 0.97) and Topcon OM-4 keratometer (min. 0.67, max. 0.94) in the adult group, while in the child group, the three instruments showed slightly decreased correlation (min. 0.45, max. 0.97 on RK-5, min. 0.36, max. 0.92 on OM-4), especially in the left eyes. Conclusions: The handheld Retinomax K-plus2 provided comparable data to that of the conventional on-table Topcon and manual keratometers. This instrument will beuseful in the clinic to measure keratometry in children or immobilized patients who have difficulty sitting during the examination.

        단일 밴드 중적외선 영상으로부터 표면온도 추정을 위한 기초연구

        3~5μm 파장대의 중적외선 영상은 화산 활동이나 산불과 같이 고온 현상을 관측하는데 효과적이다. 그러나 중적외선 영역은 지표의 복사율과 대기의 영향으로 인한 변화가 매우 심하고, 특히 낮 영상의 경우 태양 복사량에 의한 영향도 고려해야 하는 어려움이 있다. 따라서 단일밴드인 중적외선 영상을 이용하여 표면온도를 얻기 위해서는 영상이 취득된 시간과 장소에서 관측된 태양 복사량 및 여러 가지 대기 변수가 필요하다. 이 연구는 기존의 다중밴드 기반의 중적외선 영상 활용방법과 달리 단일 밴드 중적외선 영상을 이용하여 표면온도 측정을 위한 기초연구에 그 목적이 있다. 이를 위하여 MODIS 영상을 대상으로 MODTRAN을 사용하여 중적외선 영역의 대기보정 기법을 적용 한 뒤 복사전달 모델을 이용하여 지표의 온도를 측정하였다. 획득된 온도 영상의 정밀도를 측정하기 위해 기존의 온도 알고리즘인 MODIS Sea Surface Temperature 알고리즘에 의해 얻어진 해수온도와 비교를 통하여 오차 원인에 대해 분석을 실시하였다. 두 결과의 온도 차는 낮 영상의 경우 0.89±0.54℃ 밤 영상의 경우 1.25±0.41℃로 비교적 긍정적인 결과를 보였다. 그러나 낮 영상의 육지의 경우 대기에 의한 영향보다 태양빛의 반사가 주된 오차의 원인이 되며 이는 지표 복사율에 의한 영향이 매우 크게 작용하고 있음을 추정할 수 있다. 이 연구는 현재까지 해수에 대한 적용에 국한된 것으로 육상의 경우 복사율 변화가 매우 크기 때문에 중적외선 단일밴드에 의한 온도추정이 매우 어려울 것으로 예상된다. Middle-infrared (MIR) spectral region between 3.0 and 5.0 μm in wavelength is useful for observing high temperature events such as volcanic activities and forest fire. However, atmospheric effects and sun irradiance in day time has not been well studied for this MIR spectral band. The objectives of this basic study is to evaluate atmospheric effects and eventually to estimate surface temperature from a single channel MIR image, although a typical approach utilize split-window method using more than two channels. Several parameters are involved for the correction including various atmospheric data and sun-irradiance at the area of interest. To evaluate the effect of sun irradiance, MODIS MIR images acquired in day and night times were used for comparison. Atmospheric parameters were modeled by MODTRAN, and applied to a radiative transfer model for estimating the sea surface temperature. MODIS Sea Surface Temperature algorithm based upon multi-channel observation was performed in comparison with results from the radiative transfer model from a single channel. Temperature difference of the two methods was 0.89±0.54℃ and 1.25±0.41℃ from the day-time and night-time images, respectively. It is also shown that the emissivity effect has by more largely influenced on the estimated temperature than atmospheric effects. Although the test results encourage using a single channel MIR observation, it must be noted that the results were obtained from water body not from land surface. Because emissivity greatly varies on land, it is very difficult to retrieval land surface temperature from a single channel MIR data.

        거자법(巨刺法)에 의한 전침자극(電鍼刺戟)이 흰쥐의 formalin 유도(誘導) 통증(痛症)에 미치는 영향(影響)

        Acupuncture has been used for treatment of numerous diseases, especially for pain control in the oriental culture. However, the mechanism of pain control by acupuncture was not clear so far. The present study was examined that the effects of electro-acupuncture (EA) applied to the acu-point of extra-segmental area on modulation of formalin induced pain in Sprague - Dawley rats. In order to apply EA to acu-points in the plantar area of right fore paws, a pair of teflon - coated stainless steel wires were implanted in HT 7 (shin-mun) and PC 7 (dae-neong) 5 days before behavioral test. A behavioral test was performed by means of video camera after injection of 5% formalin ($50{\mu}l$) into the lateral plantar region of left hind paw. EA was delivered by a constant current stimulator at 4~5 mA, 2 ms, and 10 Hz for 30 min. The electromyographic activities were recorded in the biceps femoris muscle under chloral hydrate anesthesia. Test stimuli with 1~9mA were applied to the sural nerve territory including the medial portion of the 4th toe and the lateral portion of the 5th toe. Behavioral responses including favoring, flinching and bitting were occured in the biphasic pattern, such as the lst phase (0~5 min) and the 2nd phase (20~45 min) after formalin injection. However, EA (4~5 mA, 2 ms, 10 Hz) significantly inhibited Che behavioral responses. EMG activities of flexor reflex had a latency of 100~300 ms and thresholds of test stimuli for EMG were 4~5 mA in normal rats. Injection of formalin decreased threshold of test stimuli and increased EMG activities for 2hrs after injection. However, EA significantly inhibited EMG activities of flexor reflex increased by formalin and recovered EMG evoked thresholds. These results suggest that contralateral extra-segmental EA inhibits the first and second phases of formalin induced pain but their mechanism be needed to examine additionally.

        中學 科學敎育의 探究學習에 대한 評價問項 開發 (Ⅱ)

        This study was accomplished to develope the evaluation items for inquiry learning in the 2nd grade Middle School Science for the consecutive study of the evaluation items for inquiry learning in the 1st grade Middle School Science(U-Hang Ki et al, 1984). In this study, paper and pencil test items and performance test items are made by analyzing the abilities of inquiry according to the contents and four basic experiments from each unit in the 2nd grade Middle school science. These evaluation items were applied to tke five classes of the 2nd grade of middle school to test their validity. It is desirable that performance test schuld be used for the evaluation for the abilities of inquiry which can not be evaluated by paper and pencil test. In the evaluation methods of performance test, tester evaluation, peer evaluation and self-evaluation can be applied to the science class in a multi-student class. In higher grade, however tester evaluation is more desirable than peer and self evaluation. It is found that peer evaluation and self-evaluation make possible the perfect study by feedback.

        This study was conducted to investigate the characteristics of nonstarchy poly-saccharides in Korean ginseng, (Panax ginseng C.A. Meyer). The results obtained are as follows. 1. The total sugar content of ginseng roots were decreased with increasing the cultural period. On the other hand, the crude fiber content was increased with that of the ginseng leaves or stems. But the crude fiber in root was much less than that of leaves and stems. 2. The dietary fiber content of ginseng root on 5 years old was 14.20% as neutral detergent fiber, 9.08% as acid detergent fiber, hemicellulose 5.12%, cellulose 7.98% and lignin of 1.10%, respectively. 3. Much more pectin was found in ginseng roots which was cultivated for shorter period. And it was contained much more in the root than in the leaves and stems. 4. Ginseng hemicellulose content in root was 5% to 10%. It was decreased with increasing cultivated period. Hemicellulose was constituted of xylose, arabinose, glucose, rhamnose and xylose of these sugars was the predominant. 5. X-ray diffraction pattern of ginseng cellulose showed maximum intensity at the interplanar angle of 4.1˚.

      • 가정과 교사의 환경교육 수행실태

        The purpose of this study was to investigate the performance of environmental education by Home Economics teachers. The researchers used a questionnaire for 153 Home Economics teachers of secondary schools in Gwang-Ju. The results are as follows : Home Economics teachers had a fairly high level of environmental consciousness, but they showed low levels of environmental knowledge and environmental education performance. Married teachers with more educational experience kept a high level of environmental consciousness. Unmarried teachers had more environmental knowledge than married ones. Older teachers with more educational experience performed more environmental education than the others. The teachers with more chances for environmental information had a relatively high level of environmental knowledge. The teachers from an environment-friendly community who had more educational experience and chances for environmental information showed a relatively high level of environmental education performance.
