<P>Here, we describe and conceptualize the influence of surface engineering for hollow manganese oxide nanoparticles (<B>HMONs</B>)-based contrast agents for magnetic resonance imaging (MRI). A systematic study has been conducted for enhancing the relaxivities of <B>HMONs</B> by functionalizing their surface with various capping ligands having different anchoring groups (carboxylate, alcohol, thiol, and amine). Among all studied ligands, carboxylate-anchored ligands exhibit significant increase in magnetization value upon surface capping of <B>HMON</B>. Contrary to the previous hypotheses based on the surface-Mn<SUP>2+</SUP> ions accessibility to water molecules, we proposed surface capping induced magnetization in <B>HMON</B> is responsible for the enhanced relaxivity (<I>r</I><SUB>1</SUB>) value. Further, <I>in vivo</I> imaging efficacy of oleate-capped HMON (<B>OA-HMON</B>) has been demonstrated in mouse brain. This study provides an insightful understanding of contrast enhancement and modulation by surface ligands on manganese oxide-based T<SUB>1</SUB>-contrast agents.</P> [FIG OMISSION]</BR>

High environmental temperatures observed during the summer month reduce bovine reproduction and fertility. In this study, we examined the effect of maternal thermal stress on the number of immature oocytes retrieved by OPU procedure from Jersey heifers and their subsequent embryo developmental competence. Total 57, 69, 81 immature oocytes were retrieved from same donors (n = 3) in May (spring), June (early summer) and July (summer), respectively. Blastocyst rates were 33.3, 34.8 and 24.8% in spring, early summer and summer, respectively. Interestingly, we observed that the number of retrieved oocytes from donors were higher under extreme hot temperature (summer) than moderated temperature (spring). In contrast, blastocyst rates were lower under the hot weather. It is presumed that the reason for the increased number of retrieved oocytes in summer is associated with increased FSH levels by reduced progesterone levels due to impaired corpus luteum. And reduced embryo developmental competence in hot weather is thought to be associated with disruption of nuclear and cytoplasmic maturation. Taken together, our result suggest that maternal thermal stress affects the number of retrieved oocytes from dairy heifers and embryo developmental competence.

The use of synovial fluid-derived mesenchymal stem cells (SFMSCs) obtained from patients with degenerative arthropathy may serve as an alternative therapeutic strategy in osteoarthritis (OA) and rheumatoid arthritis (RA). For treatment of OA and RA patients, autologous transplantation of differentiated MSCs has several beneficial effects for cartilage regeneration including immunomodulatory activity. In this study, we induced chondrogenic differentiation of SFMSCs by inhibiting protein kinase A (PKA) with a small molecule and microRNA (miRNA). Chondrogenic differentiation was confirmed by PCR and immunocytochemistry using probes specific for aggrecan, the major cartilaginous proteoglycan gene. Absorbance of alcian blue stain to detect chondrogenic differentiation was increased in H-89 and/or miRNA-23b-transfected cells. Furthermore, expression of matrix metalloproteinase (MMP)-9 and MMP-2 was decreased in treated1 cells. Therefore, differentiation of SFMSCs into chondrocytes through inhibition of PKA signaling may be a therapeutic option for OA or RA patients.

Aims: We investigated the clinical impact of Hepatitis B core-related antigen (HBcrAg) level in defining different phases of patients with chronic hepatitis B (CHB). Methods: Stored residual serum samples from longitudinal cohorts of CHB patients in Korea University College of Medicine were studied. Patients were divided into four phases of CHB based on the histology result: hepatitis B e antigen (HBeAg)-positive chronic infection (EPI), HBeAg-positive chronic hepatitis (EPH), HBeAg-negative chronic hepatitis (ENH), HBeAg-negative chronic infection (ENI). Results: In total, 425 patients followed up for 83.1 months were included. The number of patients in each phase are as follows: 26 in EPI, 243 in EPH, 137 in ENH, and 19 in EHI. To evaluate the clinical impact of HBCrAg in differentiating EPI and EPH, patients older than 60 years old, patients with clinically or ultrasonographically evident liver cirrhosis or fibrosis-4 index (> 3.25), and HBV-DNA ≤ 20,000 IU/mL were excluded. In 145 selected patients, 26 patients were in EPI and 119 patients were in EPH. HBCrAg level was significantly higher in EPI than in EPH (8.22 log U/mL vs. 7.57 log U/mL, P=0.003). On multivariate analysis, only higher HBCrAg level (HR 0.447, P=0.013) was significantly associated with an increased probability of EPI. To evaluate the clinical impact of HBCrAg in differentiating ENH and ENI, patients with clinically or ultrasonographically evident liver cirrhosis or fibrosis-4 index (> 3.25), and HBV-DNA > 20,000 IU/mL were excluded. In 33 selected patients, 21 patients were in ENH and 12 patients were ENI. HBCrAg level was significantly higher in ENH patients than in ENI patients (5.24 log U/mL vs. 3.98 log U/mL, P<0.001). Only elevated ALT according to the KASL criteria (HR 4.875, P=0.049) was significantly associated with increased probability of ENH. Conclusions: HBcrAg level is a useful marker in differentiating EPI and EPH in patients with CHB.

Purpose : Smoking adversely affects the health of smokers and passive smoking threatens the health of people around smokers. The purpose of this review was to summarize the effect of smoking on sleep. Methods : We reviewed 430 articles on smoking and the effect of medication for smoking on sleep. These 430 articles consisted of 78 articles on the physiological effects of nicotine, 20 articles on the effects of nicotine on sleep, 56 articles on smoking related diseases provoking a sleep disorder, 183 articles on other substances (except nicotine) provoking a sleep disorder, and 93 articles on the effects of stopping smoking and medication for stopping smoking on sleep. Results : We summarized the physiological effects of nicotine on sleep, the effects of other substances such as acetaldehyde on sleep, smoking related illnesses including obstructive sleep apnea, chronic obstructive pulmonary disease, chronic rhinosinusitis, cardiovascular and cerebrovascular disease, depression, sleep bruxism, and restless legs syndrome, and the effects of smoking cessation on sleep. Conclusion : Research on the effect of smoking on sleep is necessary to understand the mechanism of smoking. More research on smoking, sleep, and brain mechanisms will give us a closer insight into the relationship between smoking and sleep.
국내 항공사 예매 웹 사이트에 대한 사용성 평가도구 제작 및 유효성 검증

최근 국내 각 항공사 웹사이트를 통해 항공편을 예약하는 사용자가 급격히 증가하고 있으며, 이 중 적지 않은 사람들이 웹사이트의 사용과 관련된 불만을 제기하고 있다. 하지만, 아직 웹사이트 인터페이스에 대한 실효성 높은 사용성 평가 도구나 사용성 개선을 위한 가이드라인이 없기 때문에 각 항공사 웹사이트 인터페이스에 대한 사용자의 불만족은 더욱 심화되고 있다. 이에, 본 연구에서는 웹사이트의 사용성을 평가할 수 있는 도구를 개발하여 국내 항공사 웹사이트를 대상으로 사용성 평가를 실시함으로써, 그 유효성을 검증하고자 하며, 웹사이트 인터페이스에 대한 사용성 개선을 위해 착안해야 할 주요 사항을 확인해 보고자 한다.

In this study, we investigated conception rates after timed embryo transfer (ET) with frozen-thawed in vivo produced embryos from Holstein and Jersey cattle between 2008 to 2021 by four seasons at our dairy centre located in Cheonan-si, South Korea. Frozen-thawed embryos (Holstein; 416, Jersey; 169) were transferred to Holstein heifers (age = 12.5 to 31 mo). The conception rate of ET results by breed, Holstein-derived embryos (37.7%) tended to be higher than Jersey embryos (35.5%). It is presumed that the reason for the lower conception rate in Jersey-derived frozen embryos than Holstein is associated with high intracellular lipid content, causing increased damage to embryos during cryopreservation. The conception rate of ET results by four seasons, summer (34.9%) and autumn (34.4%) significantly lower than spring (38.7%) and winter (39.6%) (P < 0.05). Interestingly, according to the previous study, conception rates in summer decreased by 20-30% compared to other seasons in artificial insemination (AI), whereas ET decreased by approximately 7% compared to other seasons in this study. It may be advantageous to ET instead of AI in commercial dairy farms under summer heat stress conditions.

<P><B>Abstract</B></P> <P>Microfluidic polymerase chain reaction (PCR) has been of great interest owing to its ability to perform rapid and specific nucleic acid amplification and analysis on small volumes of samples. One of the major drawbacks of microfluidic PCR is bubble generation and reagent evaporation, which can cause malfunctions. Here, through theoretical modeling and characterization of bubble behavior, we propose a bubble-free microfluidic PCR device via controlled fluid transfer. Our approach exploits a thin impermeable polyethylene (PE) top layer that minimizes the generation of bubbles by inhibiting mass transport along a vertical direction. Simulation results demonstrate that a calculated mass flow difference of approximately 370% can be obtained by utilizing an impermeable membrane as the vertical barrier layer. To demonstrate proof-of-concept, two nanoporous polymeric materials, poly(dimethylsiloxane) (PDMS) and PE, were used for stand-alone self-powered sample loading (approximately 70 s) and for use as a vertical barrier layer, respectively. Consequently, we demonstrate successful amplification of the cMET gene, a nucleic acid (NA) biomarker for lung cancer, and complete an ultrafast PCR test in less than 3 min using a high powered Peltier-based thermal cycler under bubble-free conditions.
