        Ex vivo bone morphogenetic protein-2 gene delivery using gingival fibroblasts promotes bone regeneration in rats

J Clin Periodontol 2009; 37: 305–311. doi: 10.1111/j.1600-051X.2009.01522.x</I>.</P><P>Abstract</P><P>Aim</P><P>The aim of the present study was to investigate bone regeneration following ex vivo bone morphogenetic protein-2 (BMP-2) gene delivery using human gingival fibroblasts (HGFs) in rat calvarial defects.</P><P>Materials and Methods</P><P>An 8 mm craniotomy defect was created in Sprague–Dawley rats. The animals were divided into four groups: (1) non-grafted group, the defect was left empty; (2) collagen matrix group, the defect was filled with collagen matrix only; (3) HGF group, the defect was filled with non-transduced HGFs on collagen matrix; (4) BMP-2/HGF group, the defect was filled with BMP-2 gene-transduced HGFs on collagen matrix. Animals were sacrificed at 2 and 4 weeks after surgery, and micro-computed tomographic and histologic observations were performed.</P><P>Results</P><P>The BMP-2/HGF group showed promoted osseous healing of calvarial defects, as compared with the other groups. At both 2 and 4 weeks, regenerated bone area was significantly greater in the BMP-2/HGF group than the other three groups. Quite a few number of transplanted HGFs were observed within the regenerated bone tissues.</P><P>Conclusions</P><P>The results of this study suggest that ex vivo BMP-2 gene delivery induces prominent bone regeneration in vivo and HGFs may be useful as target cells for ex vivo gene therapy.</P>

      • The Expression of IL-4 and Interferon-gamma Genes in Proportion to Serum IgE Levels in Peripheral Blood Mononuclear Cell of Patien

        IL-4 and IFN-γ gene expressions in peripheral mononuclear cells(PBMC) were compared by semi-quantitative reverse transcription-polymerase chain reaction (semi-quantitative RT-PCR) between atopic dermatitis(AD) patients showing high serum IgE level and those showing low serum IgE level. The subjects of this study were 15 cages; 5 AD patients with high serum IgE(>2,000kU/l), 5 AD patients with low serum IgE(<100kU/l), and 5 healthy controls. Cultures of PBMC were harvested at 0 hr(unstimulated), 3 hr, 6 hr, 12 hr, and 24 hr(stimulated). Phorbol 12-myristate 13-acetate in combination with calcium ionophore A231879(PMA / CA) was used as a stimulant. Semi-quantitative RT-PCR was performed. The results were as follws; 1) IL-4 gene expression in unstimulated PBMC was higher at AD patient groups than at control group but statistically higher only at AD patient group with high serum IgE level than at control group (P<0.05). 2) IFN-γ gene expression in unstimulnted PBMC was higher at AD patient groups than at control group without statistical significance. 3) IL-4 mRNA was first detected at 3 hr after stimulation in vitro and persisted until 6 hr, and slowly decreased in all three groups. IL-4 gene expression was higher at 3 hr than at 12 hr or 24 hr in AD patient group with high IgE level(P<0.05). 4) IFN-γ gene expression was first detected at 3 hr and Persisted until 24 hr. IFN-γ gene expression was lower at 24hr than at 3 hr, 6hr, and 12hr in AD patient group with low IgE level(P<0.05). 5) IL-4 and IFN-γ gene expressions in stimulated PBMC were not different among all three groups. In light of our results, it is suggested that T cells of AD patients are more activated in vivo because IL-4 and IFN-γ gene expressions are increased at AD patient groups than at control group. Since there are no differences of IL-4 and IFN-γ gene expressions in PBMC stimulated with PMA / CA, it may be suggested that AD is not characterized by the shift in the reciprocal relationship between IL-4 and IFN-γ production when T cells are stimulated under antigen presenting cell - independent conditions.

      • Electrical loop antenna using stacked patches for 433.92 MHz RFID reader

        A novel and compact electrical loop antenna for an RFID reader at 433.92 MHz is proposed. Using modified and stacked patches, in-phase and uniform currents are excited so that an omnidirectional radiation pattern with a φ-component of the electric field is obtained. The antenna is analysed based on simulated and measured results.

      • 동맥성 발기부전증 환자의 진단에서 선택적 혈관조영술의 역할의 재평가

        For the evaluation of role of internal pudendal arteriogram in arteriogenic impotence, 70 selective arteriograms of 35 suspected postraumatic impotence patients were analyzed and compared to results of penile bracial index(PBI), duples ultrasonograms of cavernosal artery and cavernosal artery systolic occlusion pressure(CASOP). The results of selective internal pudendal arteriography(SIPA) were classified as 44 cases with normal vascular findings and 26 with abnormal findings. Between two groups, ther was no significant difference in peak velocity and diameter of cavernosal artery in duples ultrasonograms, in pressure difference between systolic pressure of brachial artery and CASOP, and in PBI. These results are also true even in cases with nonvisualization of cavernosal artery casued by complete occlusion of internal pudendal artery. In conclusion, selective pudendal arteriogram is the only one reliable study for the evaluation of arteriogenic impotence, although further study is needed to define the role of other noninvasive approaches.

        Scanning Electron Microscopic Observation of Human Skin Replica

        The skin is the largest organ of the integument system whose surface is closely related with many physiological and pathological conditions. Various methods are used to understand the structural and functional status of human skin. We would like to present usefulness of scanning electron microscopic (SEM) observation of skin replica and its significance of training module for a novice. The silicon replicas from several regions of the body (hand, finger, forearm, lip, and face) were casted by applying Exafine$^{(R)}$ mixture. The positive replicas were prepared by applying EPON 812 mixture on negative silicon replicas. Some of the negative silicon replicas were cut with a razor blade and surface profiles were observed. The negative and positive replicas were coated with platinum and were observed under the scanning electron microscope. We could investigate the detailed structures of the human skin surface without any physical damage to the subject. The positive replicas depicted real surface structure of the human skin vividly. The cross sectional view of the negative silicon replicas provided surface profile clearly. The scanning electron microscopic observation of the human skin replicas would be useful to study skin surface structures and to evaluate medical and esthetical applications.

        Communication Redundancy for Reliability Improvement in an Industrial Monitoring and Control System

        In development of monitoring and control systems, one of the most important points is to consider a redundancy so that the system can be operated normally although hardware faults are partly occurred. The purpose of this paper is to introduce a monitoring and control system with a redundancy function for I/O servers and communication networks. I/O servers composed with an active server and a standby server. Each server also has 3 communication ports, 2 ports of them were connected to field units and the other 1 port was connected to the other server. Field units have to be constructed to 2 communication ports connected I/O servers through communication lines. Also, server communication module was implemented for analyzing and handling fault elements. and was submodularized for linking easily with a monitoring and control module. An experiment with 2 servers and 2 field units was constructed to demonstrate its effectiveness.

        The Expression of IL-4 and Interferon-gamma Genes in Peripheral Blood Mononuclear Cells of Patients with Severe Atopic Dermatitis : Evaluation in Proportion to Serum IgE Levels

        Background : It is not yet clear whether the abnormal cytokine production in relation to serum IgE levels in atopic dermatitis (AD) is associated with the amount of mRNA of cytokine gene. Objective : Our purpose was to delineate the effect of reciprocal correlation in the level of mRNA between interleukin-4 (IL-4) and interferon-gamma (IFN-γ) in severe AD. Methods : We examined 1S cases including 5 AD patients with high serum IgE ($gt;2,000 kU /liter), 5 AD patients with low serum IgE ($lt;100 kU/liter), and 5 healthy controls. Using semi quantitative reverse transcription-polymerase chain reaction, IL-4 and IFN-γ gene expressions in peripheral mononuclear cells (PBMC) were examined. Results : 1) IL-4 gene expression in spontaneous PBMC was higher in AD patient groups than in control group, significantly higher only in AD patient group with high serum IgE level (p $lt; 0.05 ). 2) IFN-γ gene expression in spontaneous PBMC showed increased tendency in AD patient groups than in control group without statistical significance. 3) IL-4 and IFN-γ gene expressions in stimulated PBMC were not different among all three groups. Conclusion : In light of our results, high and low IgE subgroups in AD can exist and AD may not be characterized by the shift in the reciprocal relationship between IL-4 and IFN-γ when T cells are stimulated under antigen presenting cell-independent conditions.

      • Flurbiprofen 함유 키토산 제제가 치은 섬유아세포에 미치는 영향

        The main goal of periodontal regeneration is to be achieved by epithelial exclusion, periodontal ligament cell activation or alveolar bone regeneration. The purpose of this study was to investigate on the physico-chemical and biological characteristics of biodegradable chitosan beads. Chitosan beads were fabricated by ionic gelation with sodium tripolyphosphate and they had the size in 300㎛ diameter. As therapeutic agent, flurbiprofen was incorporated into the beads by 10, 20% loading contents. The release of drugs from the chitosan beads was measured in vitro. Also, biological activity tests of flurbiprofen loaded chitosan beads including cytotoxicity test, ihhibition of IL-1β production, suppression to PGE_2 production, collagenase inhibition test, the ability of total protein synthesis, and tissue response were evaluated. The amount of flurbiprofen released from chitosan was 33-50% during 7 days. Minimal cytotoxicity was observed in chitosan beads. Flurbiprofen released from chitosan beads significantly suppressed the IL-1β production of monocyte, PGE_2 production and markedly inhibited collagenase activity. Meanwhile, flurbiprofen released from this system showed increased ability for protein synthesis. Throughout 4-week implantation period, no significant inflammatory cell infiltrated around chitosan bead and also fibroblast like cell types at the beads-tissue interface were revealed with gradual degradation of implanted chitosan beads. From these results, it was suggested that flurbiprofen loaded chitosan beads can be effectively useful for biocompatible local delivery system in periodontal regeneration.
