        Correlations of Four Genetic Single Nucleotide Polymorphisms in Brain-Derived Neurotrophic Factor with Posttraumatic Stress Disorder

        Objective-To investigate the correlations of four genetic single nucleotide polymorphisms (SNPs) of brain-derived neurotrophic factor (BDNF) with posttraumatic stress disorder (PTSD). Methods-A total of 300 patients with sporadic PTSD and 150 healthy subjects (the control group) were selected according to the diagnostic criteria of PTSD (DSM-IV), and the genotypes of the BDNF SNPs G-712A, C270T, rs6265, and rs7103411 were detected by polymerase chain reaction and direct DNA sequencing to determine intergroup differences in the genotypes and allele frequencies; the pvalues were corrected with the permutation test. Results-The genotypes and allele frequencies of the SNPs G-712A, rs6265, and rs7103411 of BDNF showed no significant intergroup differences (p>0.05). However, the genotype and allele frequencies of C270T showed significant differences between the PTSD group and the control group (p<0.05). Conclusion-The SNP C270T of BDNF may be associated with PTSD. Individuals carrying the polymorphic T allele of C270T may be more likely to suffer from PTSD.

        Structural configurations and dynamic performances of flexible riser with distributed buoyancy modules based on FEM simulations

        Flexible risers are usually used as conveying systems to bring ocean resources from sea bed up to onshore. Under ocean environments, risers need to bear complex loads and it is crucial to comprehensively examine riser's configurations and to analyze structural dynamic performances under excitation of bottom vehicle motions, to guarantee structural safe operation and required service lives. In this study, considering a saddle-shaped riser, the influences of some important design parameters, including installation position of buoyancy modules, buoyancy ratio and motion of mining vehicle, on riser's configuration and response are carefully examined. Through our FEM simulations, the spatial distributions of structural tensions and curvatures along of riser length, under different configurations, are compared. Then, the impacts of mining vehicle motion on riser dynamic response are discussed, and structural tolerance performance is assessed. The results show that modules installation position and buoyancy ratio have significant impacts on riser configurations. And, an appropriate riser configuration is obtained through comprehensive analysis on the modules positions and buoyancy ratios. Under this proposed configuration, the structural tension and curvature could moderately change with buoyancy modules and bottom-end conditions, in other words, the proposed saddle-shaped riser has a good tolerance performance to various load excitations.

        Carbon disulfide induces mitochondria-mediated apoptosis in Sertoli-germ cells coculture

        Carbon disulfide (CS2), a common organic solvent, induces a variety of adverse effects in the male reproductive system. In this study, we investigated the cytotoxicity, ultrastructural changes, and potential apoptotic induction mechanisms of CS2 in mixed cultures of Sertoli and germ cells. Sertoli and germ cells were cocultured and treated with CS2 for 24 h. Growth rates were noted, and apoptotic cells were identified by Hoechst 33258 staining. Ultrastructure changes were observed via transmission electron microscopy (TEM). Mitochondrial membrane potential and expressions of apoptosis-related factors (cytochrome c, Bax, Bcl- 2, caspase-3 and caspase-9) were examined by JC-1 staining, western blot, and real-time PCR. The results showed that CS2 treatment was associated with reduced growth rates of Sertoli-germ cells. Ultrastructure changes in Sertoli-germ cells treated with CS2 were typical of apoptosis. In addition, CS2 treatment depolarized mitochondrial membrane potential, upregulated Bax levels and downregulated Bcl-2 levels, released cytochrome c from the mitochondrial intermembrane space to the cytosol, and triggered mitochondria-mediated apoptosis. Subsequently, caspase-9 and caspase-3 were activated, resulting in Sertoli-germ cells apoptosis. The above data suggest that CS2 has adverse effect on the viability of Sertoli-germ cells and induces apoptosis through mitochondrial pathway.

        가상 인플루언서 특성에 따른 사용자 호감도 연구

        현대인들은 인플루언서와의 적극적인 소통을 통해 손쉽게 정보를 얻고 소비한다. 이러한 인플루언서 산업의 진화 과정에서 인간을 대신하는 가상 인플루언서가 등장하며 큰 관심을 끌고 있다. 가상 인플루언서 시장은 앞으로 여러 분야와 융합하여 확장될 것으로 전망되며 이를 활용한 콘텐츠의 활성화를 위해 가상 인플루언서와 사용자 경험에 관한 실증연구와 논의가 요구된다. 이에 본 연구는 가상 인플루언서가 가지는 특성이 사용자들의 호감도에 미치는 영향을 살펴보았다. 현존하는 두 명의 대표 가상 인플루언서의 SNS를 실험물로 제작하여 가상 인플루언서의 특성인 매력성, 신뢰성, 친밀성, 전문성, 상호작용성, 자아 일치성에 대한 설문과 심층 인터뷰를 진행하였다. 연구 결과 매력성과 신뢰성은 호감도에 유의미한 영향을 미치는 것으로 조사되었으나 전문성과 상호작용성, 자아 일치성은 사용자 호감도를 낮추는 특성으로 확인되었고, 친밀성의 경우 직접적인 영향을 미치지 못하는 것으로 나타났다. 본 연구는 가상 인플루언서의 특성이 사용자들의 호감도에 미치는 영향과 관계성을 파악하여 다양한 콘텐츠 산업 분야의 가상 인플루언서 활용 가능성에 도움이 될 자료를 제시했다는 것에 그 의의가 있다. These days, we easily get and consume information through active communication with influencer. In the process of evolution of the influencer industry, a virtual influencer has emerged and has been attracting great attention. In order to activate content using a virtual influencer, an empirical research and discussion on virtual influencer and an user experience are required. Therefore, this study examined the effect of the characteristics of virtual influencer on users' favorability. By prototyping the SNS of the two virtual influencers, surveys and in-depth interviews were conducted on the characteristics of the virtual influencer, such as attractiveness, reliability, intimacy, expertise, interaction, and self-consistency. As a result of the study, attractiveness and reliability were found to have a positive effect on favorability, but expertise, interaction, and self-consistency were found to be characteristics that lower user favorability, and intimacy did not have a direct effect. This study is meaningful in that it identified the effect and relationship of the characteristics of virtual influencer on users' favorability and presented the possibility of use.

        US Preventive Services Task Force prostate-specific antigen screening guidelines result in higher Gleason score diagnoses

        Purpose: To evaluate the impact that the 2012 US Preventive Services Task Force (USPSTF) prostate-specific antigen (PSA) screening guidelines have had on the diagnosis of prostate cancer, we compared the incidence and distribution of new cases diagnosed in 2011-before the USPSTF PSA screening recommendations versus 2014 at which time the guidelines were widely adopted. Materials and Methods: We identified all prostate biopsies performed by a large urology group practice utilizing a centralized pathology lab. We examined total biopsies performed, percentage of positive biopsies, and for those with positive biopsies examined for differences in patient age, PSA, and Gleason score. Results: A total of 4,178 biopsies were identified – 2,513 in 2011 and 1,665 in 2014. The percentage of positive biopsies was 27% in 2011 versus 34% in 2014 (p<0.0001). Among patients with positive biopsies, we found statistically significant differences between the 2 cohorts in the median ages and Gleason scores. Patients were about 1 year younger in 2014 compared to 2011 (t-test; p=0.043). High Gleason scores (8–10) were diagnosed in 19% of the 2014 positive biopsies versus 9% in the 2011 positive biopsies (chi square; p<0.0001). Conclusions: After the widespread implementation of the 2011 USPTF PSA screening guidelines, 34% fewer biopsies were performed with a 29% increase in positive biopsy rates. We found a significantly higher incidence of high grade disease in 2014 compared with 2011. The percentage of patients with positive biopsies having Gleason scores 8–10 more than doubled in 2014. The higher incidence of these more aggressive cancers must be part of the discussion regarding PSA screening.
